    NOD/Scid IL2Rγnull (NSG) mice reconstituted with peripheral blood mononuclear cells (PBMC) donated by patients with ulcerative colitis or Crohn’s disease highly reflect the respective pathological phenotype. To determine if these findings could be applicable to atopic dermatitis (AD) and psoriasis vulgaris (PV), PBMCs isolated from AD and PV patients were first subjected to immunological profiling. Subsequently, NSG mice were reconstituted with these PBMCs. Hierarchical clustering and network analysis revealed a distinct profile of AD and PV patients with activated CD4+ T cells (CD69, CD25) occupying a central position in the AD network and CD4+ CD134+ cells acting as the main hub in the PV network. Following dermal application of DMSO, both NSG-AD and NSG-PV mice exhibited increased clinical, skin and histological scores. Immuno-histochemical analysis, frequencies of splenic human leukocytes, and cytokine expression levels indicated that CD4+ CD69+ cells, M1 and TSLPR-expressing monocytes, switched B cells and MCP-3 were the driving factors of inflammation in NSG-AD mice. In contrast, inflammation in NSG-PV mice was characterized by an increase in fibroblasts in the epidermis, frequencies of CD1a-expressing monocytes and IL-17 levels. Therefore, the pathological phenotypes of NSG-AD and NSG-PV mice differ and partially reflect the respective human disease

    Recurrence-Free Survival after Synovectomy and Subsequent Radiosynoviorthesis in Patients with Synovitis of the Knee—A Retrospective Data Analysis

    Background: Persistent knee synovitis leads to joint discomfort, incapacitating inflammation, and functional limitations. The conventional approach has involved surgical procedures to eliminate the actively inflamed synovial membrane. This study aims to investigate the recurrence-free survival and functional outcome after synovectomy and subsequent radiosynoviorthesis (RSO) in patients with knee synovitis. Methods: Thirty-seven knees diagnosed with pigmented villonodular synovitis (PVNS), rheumatoid arthritis (RA), and peripheral spondyloarthritis underwent synovectomy and subsequent RSO between May 2005 and October 2016. The mean age was 34.9 ± 15.1 years, and the mean follow-up period was 84 ± 36.4 months. Clinical outcomes were assessed using the Oxford Knee Score and the presence of swelling and pain at the last follow-up. Recurrence-free survival denotes the duration from synovectomy to surgical re-synovectomy. Results: In general, twelve knees underwent re-synovectomy after a mean follow-up of 34.8 ± 24.9 months. The recurrence-free survival was 83.8% at two years, 71.3% at five years, and 61.7% at ten years. The subgroup analysis revealed recurrence-free survival at two years in 63.6% of patients with PVNS, 86.7% of those with RA, and 100% of individuals with peripheral spondyloarthritis. Conclusions: This study demonstrates that combined therapy for synovitis is an effective approach, significantly improving clinical outcomes

    Improved Detection of Foreign Bodies on Radiographs Using X-ray Dark-Field and Phase-Contrast Imaging

    Purpose: The aim of this study was to investigate whether the detection of foreign bodies can be improved using dark-field and phase-contrast radiography compared with conventional (transmission) radiographs. Materials and Methods: Experiments were performed using ex vivo pig paws, which were prepared with differently sized foreign bodies of metal, wood, and glass (n = 10 each). Paws without foreign bodies served as controls (n = 30). All images were acquired using an experimental grating-based large object radiography system. Five blinded readers (second- to fourth-year radiology residents) were asked to assess the presence or absence of any foreign body. Sensitivity and specificity for the detection of metal, wood, glass, and any foreign body were calculated and compared using McNemar test and generalized linear mixed models. Results: Sensitivity for the detection of metal foreign bodies was 100% for all readers and image combinations. The sensitivity for the detection of wooden foreign bodies increased from 2% for transmission images to 78% when dark-field images were added (P < 0.0001). For glass foreign bodies, sensitivity increased from 84% for transmission images to 96% when adding phase-contrast images (P = 0.041). Sensitivity for the detection of any foreign body was 91% when transmission, dark-field, and phase-contrast images were viewed simultaneously, compared with 62% for transmission images alone (P < 0.0001). Specificity was 99% to 100% across all readers and radiography modalities. Conclusions: Adding dark-field images substantially improves the detection of wooden foreign bodies compared with the analysis of conventional (transmission) radiographs alone.     Deutschlands EU-Ratspräsidentschaft: Chancen zur Gestaltung der Zukunftsfähigkeit Europas nutzen

    Deutschland übernimmt zum 1. Januar 2007 für sechs Monate die EU-Ratspräsidentschaft und zugleich für ein Jahr den Vorsitz in der Gruppe der G-8 Staaten. Gerade durch das zeitliche Zusammenfallen beider Aufgaben kann Deutschland einen Beitrag zur richtungsweisenden Gestaltung der europäischen Politik leisten. Im offiziellen Ratspräsidentschafts-Programm der Bundesregierung wird die Bedeutung der Handlungsfähigkeit der EU nicht nur für den binnenwirtschaftlichen, sondern auch für den außenpolitischen und außenwirtschaftlichen Erfolg Europas betont. Die Agenda reicht von der Vollendung des Binnenmarktes, über Energiepolitik und Beschäftigungssicherung bis zur Frage nach der Gestaltung der Entwicklungspolitik. In einer Prioritätenliste wurden unter anderem die stärkere Koordination innerhalb der EU, die Überwindung außenwirtschaftlicher Ungleichgewichte, die Energiesicherheit und die Ausgestaltung der vertraglichen Beziehungen zur Russischen Föderation als zentrale Themen festgelegt. In jüngerer Zeit zeichnet sich eine Fokussierung auf die Themen "Energiesicherheit" und "Klimaschutz" (Kasten 1) ab. Das DIW Berlin greift einige zentrale Problemfelder aus dem Präsidentschaftsprogramm auf und zeigt, dass tragfähige Lösungen vor allem dann entwickelt werden können, wenn die EU als Gemeinschaft auf dem internationalen Parkett agiert.

    Just pain, no gain?: Data management systems and biodiversity data

    Scientists often see research data management as a burden. At first glance, it appears as yet another task with lots of effort and no apparent benefits. In fact, proper research data management provides many benefits and is important for biodiversity research projects. It is a key factor for their success and for the long-term impact of the research. Data management systems provide established informatics standards such as online access, versioning, and backup. Other features support scientists to organize, document, and verify their data according to accuracy and validity. Such system offers functionalities for data search, access control and traceability. This allows a collaborative, all-time available and trustable teamwork. Beside it, more and more funding agencies and publishers ask for accessible and reproducible data with a guarantee of long-term availability. In addition, stakeholders and the community request data access. Data management system curated data is ready to be published and cited. We believe that, in order to truly support researchers and ensure that they can reap the benefits of their efforts, data management platforms are needed that deal with the entire data lifecycle. We have developed the data management system “Biodiversity Exploratories Information System”. It acts as a platform to support researchers of the Priority Program “Biodiversity Exploratories” of the German Science Foundation (DFG). It guaranties data curation and enables data interchange and reuse since nearly 10 years. The system is also the foundation of the BEXIS2 data management platform. With our poster, we show the role of a data management system as a service to researchers and projects. We illustrate the benefits of such a system inside the whole life cycle of data seen from researcher and project perspective

    Superior Functional Outcome and Comparable Health-Related Quality of Life after Enhanced Recovery vs. Conventional THA: A Retrospective Matched Pair Analysis

    Background: The concept of enhanced recovery after total hip arthroplasty is gaining worldwide interest, as it shortens the length of hospital stay without an increase of complications. The aim of the study was to investigate the functional outcome and health-related quality of life 12 months after cementless total hip arthroplasty with the use of an enhanced recovery concept in comparison to a conventional rehabilitation. Material and Methods: 320 patients were retrospectively analyzed who underwent primary cementless total hip arthroplasty (THA). A total of 123 of the patients received an enhanced recovery program (ERAS) and 197 patients a conventional rehabilitation (Non-ERAS). Twelve months postoperatively, a clinical examination was performed regarding satisfaction, function and pain. Results were evaluated using WOMAC, EQ-5D-5L and EQ-VAS. A 1:1 matching was performed to correct for confounding variables, regarding age, sex and ASA score. Finally, 122 patients (n = 61, in each group) were analyzed and compared. Results: Patients showed a significant improvement of WOMAC total score, subscale pain, subscale stiffness and subscale function from preoperative to the follow up after 12 months in both groups, with significantly superior results for the WOMAC total score for the ERAS group (p = 0.042). EQ-5D and EQ-5D VAS showed a significant improvement from preoperative to 12 months postoperative (p < 0.001) for both groups, while no difference regarding the group-comparison was shown. Conclusion: Health-related quality of life and functional outcome increased to excellent values after total hip arthroplasty with the use of an enhanced recovery concept and a conventional rehabilitation, with a superior WOMAC total score for ERAS and a tendency to better results for health-related quality of life for patients with ERAS within the follow up after 12 months

    Exposure to night-time traffic noise, melatonin-regulating gene variants and change in glycemia in adults

    Traffic noise has been linked to diabetes, with limited understanding of its mechanisms. We hypothesize that night-time road traffic noise (RTN) may impair glucose homeostasis through circadian rhythm disturbances. We prospectively investigated the relationship between residential night-time RTN and subsequent eight-year change in glycosylated hemoglobin (ΔHbA1c) in 3350 participants of the Swiss Cohort Study on Air Pollution and Lung and Heart Diseases in Adults (SAPALDIA), adjusting for diabetes risk factors and air pollution levels. Annual average RTN (Lnight) was assigned to participants in 2001 using validated Swiss noise models. HbA1c was measured in 2002 and 2011 using liquid chromatography. We applied mixed linear models to explore RTN-ΔHbA1c association and its modification by a genetic risk score of six common circadian-related MTNR1B variants (MGRS). A 10 dB difference in RTN was associated with a 0.02% (0.003-0.04%) increase in mean ΔHbA1c in 2142 non-movers. RTN-ΔHbA1c association was modified by MGRS among diabetic participants (Pinteraction = 0.001). A similar trend in non-diabetic participants was non-significant. Among the single variants, we observed strongest interactions with rs10830963, an acknowledged diabetes risk variant also implicated in melatonin profile dysregulation. Night-time RTN may impair glycemic control, especially in diabetic individuals, through circadian rhythm disturbances. Experimental sleep studies are needed to test whether noise control may help individuals to attain optimal glycemic levels
