    Neutron Radiographic Study of the Effect of Heat-Driven Water Transport on the Tensile Strength of Bentonite-Bonded Moulding Sand

    Wet tensile testing is a common method to assess the stability of bentonite bonded moulding sands. For wet tensile testing, a specimen is first heated from above in order to simulate heat-driven moisture transport induced by the casting process. Then, tensile stress is applied until rupture. In this study, neutron radiography imaging was applied to moulding sands in-situ during heating and wet tensile testing in order to investigate the effects of water kinematics on the tensile strength. Neutron radiography allowed the localization of the rupture plane and the quantitative determination of the local water content with sub-mm resolution. Quantification of the temperature at the rupture plane and of the heat kinematics within the specimen was accomplished by temperature measurements both in-situ and ex-situ. In this way, experimental data correlating the wet tensile strength with the specific conditions of moulding sands at the rupture plane were obtained for the first time. Series of experiments with different initial sand moisture contents were conducted. The results show that the weakest location within a sand profile can be pinpointed at the interface between evaporation and condensation zone (i.e., at the 100 °C isotherm), where water vaporisation starts and the water bridges connecting the sand grains collapse. This weakest location has maximum strength, if the local water content at the rupture plane is between 5 and 9 wt.%. Less water leads to a strong decrease of wet tensile strength. More water requires an initial water content above 5 wt.%, which leads to a decrease of the tensile strength of the unheated sand

    Using the Multiple Analysis Approach to Reconstruct Phylogenetic Relationships among Planktonic Foraminifera from Highly Divergent and Length-polymorphic SSU rDNA Sequences

    The high sequence divergence within the small subunit ribosomal RNA gene (SSU rDNA) of foraminifera makes it difficult to establish the homology of individual nucleotides across taxa. Alignment-based approaches so far relied on time-consuming manual alignments and discarded up to 50% of the sequenced nucleotides prior to phylogenetic inference. Here, we investigate the potential of the multiple analysis approach to infer a molecular phylogeny of all modern planktonic foraminiferal taxa by using a matrix of 146 new and 153 previously published SSU rDNA sequences. Our multiple analysis approach is based on eleven different automated alignments, analysed separately under the maximum likelihood criterion. The high degree of congruence between the phylogenies derived from our novel approach, traditional manually homologized culled alignments and the fossil record indicates that poorly resolved nucleotide homology does not represent the most significant obstacle when exploring the phylogenetic structure of the SSU rDNA in planktonic foraminifera. We show that approaches designed to extract phylogenetically valuable signals from complete sequences show more promise to resolve the backbone of the planktonic foraminifer tree than attempts to establish strictly homologous base calls in a manual alignment

    During heme biosynthesis the oxygen-independent coproporphyrinogen III oxidase HemN catalyzes the oxidative decarboxylation of the two propionate side chains on rings A and B of coproporphyrinogen III to the corresponding vinyl groups to yield protoporphyrinogen IX. Here, the sequence of the two decarboxylation steps during HemN catalysis was investigated. A reaction intermediate of HemN activity was isolated by HPLC analysis and identified as monovinyltripropionic acid porphyrin by mass spectrometry. This monovinylic reaction intermediate exhibited identical chromatographic behavior during HPLC analysis as harderoporphyrin (3-vinyl-8,13,17-tripropionic acid-2,7,12,18- tetramethylporphyrin). Furthermore, HemN was able to utilize chemically synthesized harderoporphyrinogen as substrate and converted it to protoporphyrinogen IX. These results suggest that during HemN catalysis the propionate side chain of ring A of coproporphyrinogen III is decarboxylated prior to that of ring B. © by Walter de Gruyter

    A family case of fertile human 45,X,psu dic(15;Y) males

    We report on a familial case including four male probands from three generations with a 45,X,psu dic(15;Y)(p11.2;q12) karyotype. 45,X is usually associated with a female phenotype and only rarely with maleness, due to translocation of small Y chromosomal fragments to autosomes. These male patients are commonly infertile because of missing azoospermia factor regions from the Y long arm. In our familial case we found a pseudodicentric translocation chromosome, that contains almost the entire chromosomes 15 and Y. The translocation took place in an unknown male ancestor of our probands and has no apparent effect on fertility and phenotype of the carrier. FISH analysis demonstrated the deletion of the pseudoautosomal region 2 (PAR2) from the Y chromosome and the loss of the nucleolus organizing region (NOR) from chromosome 15. The formation of the psu dic(15;Y) chromosome is a reciprocal event to the formation of the satellited Y chromosome (Yqs). Statistically, the formation of 45,X,psu dic(15;Y) (p11.2;q12) is as likely as the formation of Yqs. Nevertheless, it has not been described yet. This can be explained by the dicentricity of this translocation chromosome that usually leads to mitotic instability and meiotic imbalances. A second event, a stable inactivation of one of the two centromeres is obligatory to enable the transmission of the translocation chromosome and thus a stably reduced chromosome number from father to every son in this family

    STED nanoscopy of the centrosome linker reveals a CEP68-organized, periodic rootletin network anchored to a C-Nap1 ring at centrioles

    The centrosome linker proteins C-Nap1, rootletin, and CEP68 connect the two centrosomes of a cell during interphase into one microtubule-organizing center. This coupling is important for cell migration, cilia formation, and timing of mitotic spindle formation. Very little is known about the structure of the centrosome linker. Here, we used stimulated emission depletion (STED) microscopy to show that each C-Nap1 ring at the proximal end of the two centrioles organizes a rootletin ring and, in addition, multiple rootletin/CEP68 fibers. Rootletin/CEP68 fibers originating from the two centrosomes form a web-like, interdigitating network, explaining the flexible nature of the centrosome linker. The rootletin/CEP68 filaments are repetitive and highly ordered. Staggered rootletin molecules (N-to-N and C-to-C) within the filaments are 75 nm apart. Rootletin binds CEP68 via its C-terminal spectrin repeat-containing region in 75-nm intervals. The N-to-C distance of two rootletin molecules is ∌35 to 40 nm, leading to an estimated minimal rootletin length of ∌110 nm. CEP68 is important in forming rootletin filaments that branch off centrioles and to modulate the thickness of rootletin fibers. Thus, the centrosome linker consists of a vast network of repeating rootletin units with C-Nap1 as ring organizer and CEP68 as filament modulator

The combined effects of these processes on marine planktic calcifiers at decadal to centennial timescales are poorly understood. Here, we analyze size normalized planktic foraminiferal shell weight, shell geochemistry, and supporting proxies from 3 sediment cores in the Mediterranean Sea spanning several centuries. Our results allow us to investigate the response of surface-dwelling planktic foraminifera to increases in atmospheric carbon dioxide. We find that increased anthropogenic carbon dioxide levels led to basin wide reductions in size normalized weights by modulating foraminiferal calcification. Carbon (ή13C) and boron (ή11B) isotopic compositions also indicate the increasing influence of fossil fuel derived carbon dioxide and decreasing pH, respectively. Alkenone concentrations and test accumulation rates indicate that warming and changes in biological productivity are insufficient to offset acidification effects. We suggest that further increases in atmospheric carbon dioxide will drive ongoing reductions in marine biogenic calcification in the Mediterranean Sea.Publisher PDFPeer reviewe

    Bentonitgebundene Formsande gehören aufgrund ihrer Umweltfreundlichkeit und Wiederverwendbarkeit zu den wichtigsten Formstoffen in Gießereien. Es liegt daher nahe, das Verhalten und die Eigenschaften dieser Formsande genau zu untersuchen. Im durchgefĂŒhrten Forschungsprojekt wurden die Nasszugfestigkeiten von Formstoffproben in Echtzeit wĂ€hrend einer Durchstrahlung mit Neutronen gemessen. Dabei konnten auch der Riss abgebildet und die lokalen Wassergehalte ermittelt werden
