Evaluation and scale-up of single-use bioreactors for the production and harvesting of a hepatitis C vaccine candidate
The present work examines the suitability of single-use bioreactors for production of a Hepatitis C Virus-Like Particle (VLP) vaccine candidate using the baculovirus expression system with Sf9 cells. It can be shown that a Mobius® 3L bioreactor results in viable cell concentration, viability, growth kinetics, stability and VLP production that are comparable to standard glass bioreactors. A simple translation of hydrodynamic working parameters between the two systems is adequate to match performance. Furthermore, we report on the successful scale-up of this disposable alternative from a 3L to a 50L scale using minimal optimization. These results demonstrate the potential and ease of use of this technology for the production of complex biopharmaceutical products. Using the 50 liters harvested from the run, we evaluated depth filtration and compared the results to centrifugation. Multiple filter trains with different properties were tested and the results on recovery, turbidity and impurity reduction will be presented and discussed
Un habitat de la fin de la période gauloise et un fanum du Haut-Empire à Nécy « La Martinière » (Orne)
Dans le cadre de l’amĂ©nagement autoroutier A88 entre Falaise et Argentan, une fouille archĂ©ologique prĂ©ventive a Ă©tĂ© rĂ©alisĂ©e en 2005 Ă NĂ©cy au lieu-dit « La Martinière », en limite dĂ©partementale de l’Orne et du Calvados. Bien que la zone d’étude soit de faible superficie, il a Ă©tĂ© possible d’explorer deux installations successives : l’une de la fin de la pĂ©riode gauloise et l’autre du Haut-Empire. L’intĂ©rĂŞt de ce site rĂ©side, d’une part, en la perduration de l’occupation après la conquĂŞte romaine et, d’autre part, en la mutation d’un habitat commun en lieu cultuel. Peu de comparaisons sont connues Ă ce jour en Basse-Normandie.Within the context of motorway building between Falaise and Argentan, a preventive archaeological excavation was carried out in 2005 at NĂ©cy “La Martinière”, on the departmental limit of the Orne and the Calvados. Although the study area was small, it was possible to explore two successive settlements, one dating from the end of the Iron Age and other from the Early Roman Empire. The interest of this site resides, on the one hand, in the continuity of the occupation after the Roman conquest and, on the other, the mutation of its function from settlement to place of worship. Few comparisons are known to date in Lower Normandy.En el marco del desarrollo de la autopista A88 entre Falaise y Argentan, una excavaciĂłn arqueolĂłgica preventiva se llevĂł a cabo en 2005 a NĂ©cy en un lugar llamado “La Martinière” en el lĂmite departamental de Orne y Calvados. Aunque el área de estudio es de pequeño tamaño, fue posible explorar dos instalaciones sucesivas: una al final del perĂodo galo y uno del Alto Imperio romano. El interĂ©s de este sitio se encuentra, por un lado, en la perduraciĂłn de la ocupaciĂłn despuĂ©s de la conquista romana y, en segundo lugar, en la mutaciĂłn de un hábitat comĂşn en un lugar de culto. L’âge du Fer en Basse-Normandie. Gestes funéraires en Gaule au Second-Âge du Fer. Volumes I et II
Cet ouvrage est la publication des actes du 33e colloque international de l’AFEAF, qui s’est tenu à Caen, du 20 au 24 mai 2009. De même que le colloque, il comprend deux parties. La première, qui contient dix-sept contributions, traite de l’actualité des recherches sur l’Âge du fer en Basse-Normandie, une région qui a connu un renouvellement important de la documentation permettant d’appréhender les dynamiques d’occupation et les formes d’organisation territoriale durant la Protohistoire. Plusieurs bilans synthétiques concernant l’habitat rural, les sites fortifiés et la culture matérielle, sont, entre autres, proposés ici. La seconde partie de l’ouvrage traite du thème de la mort et des pratiques funéraires durant la période Ve-Ier siècles avant J.-C. ; elle contient vingt-deux contributions. Ce sujet a été abordé en privilégiant quelques axes de réflexion, de façon à faciliter les comparaisons entre régions. À travers des bilans synthétiques, le thème spécialisé du colloque est traité à l’échelle de la Gaule, avec les éclairages de plusieurs régions de l’Europe celtique.This is the publication of the proceedings of the 33rd AFEAF symposium held in Caen from the 20th to the 24th of May 2009. Like the symposium, this book is divided in two parts. The first part, with its 17 contributions, deals with the current knowledge about the Iron Age in Lower Normandy. An important renewal of the documentation concerning this region allows a better understanding of the occupational processes and territorial organization existing throughout Protohistory. This first part also includes several summary reports on rural settlements, fortified sites as well as finds. The second part of the book, with 22 contributions, deals with the subject of death and funerary practices during the 5th to 1st centuries BC. The subject was discussed focussing on a chosen number of thoughts as to make it easier to compare between different regions. Various summary reports allow an overview of this specialized topic throughout Gaul with perspectives from other regions of Celtic Europe
