Darkness-induced effects on gene expression in Cosmarium crenatum (Zygnematophyceae) from a polar habitat
Light is a key environmental regulator in all photosynthetic organisms. Many studies focused on the physiologic response to changes in light availability of species from the Zygnematophyceae, but the impact of the absence of light and the molecular acclimation process on the other side have been poorly understood. Here we present transcriptomic analyses of Cosmarium crenatum from a polar habitat exposed to darkness. The algae were cultured in dark for one week; cell number and quantum yield of photosystem II (Fv/Fm) were monitored. Cell number was stable, but the Fv/Fm decreased in both groups, darkness-treated and control. Gene expression analysis revealed a strong repression of transcripts associated with photosynthesis, photorespiration and cell wall development. General carbohydrate and lipid metabolism were differentially regulated, but starch is shown to be the primary energy source in these conditions. Additionally, C. crenatum induced mRNA responsible for epigenetic modifications which may be a specific response to an adaption and acclimation to polar conditions. Our study sheds light on the molecular acclimation process to darkness and provides ecological implications for new perspectives in this specialized group of green algae
PROCESSO SAÚDE-DOENÇA NO CONTEXTO DE TRABALHO EM SAÚDE: PERCEPÇÕES DOS TÉCNICOS DE ENFERMAGEM DE UM AMBULATÓRIO HOSPITALAR
O estudo teve como objetivo identificar os significados atribuÃdos pelos técnicos de enfermagem de um ambulatório hospitalar ao processo saúde-doença no contexto de trabalho em saúde. Os resultados indicam que a concepção sobre saúde-doença predominante está ancorada no modelo biomédico, refletido diretamente nas práticas de cuidado desenvolvidas e na esfera do autocuidado. A manifestação de certa insatisfação no trabalho, expressa na forma de descontentamento e falta de motivação dos trabalhadores, não está relacionada ao tempo de serviço na organização. Esta pode estar indicando a influência de fatores psicossociais à saúde do trabalhador que sofre em meio ao contexto e que se encontra desamparado pela atual polÃtica interna da saúde do trabalhador, apesar de observar-se o desenvolvimento de vários projetos visando à saúde dos trabalhadores da organização. Na percepção dos entrevistados a instituição acaba evidenciando a valorização da estrutura fÃsica ao investimento de melhorias na qualificação dos trabalhadores, o que não corresponde à s expectativas da maioria dos que participaram do estudo. Portanto, ressalta-se a necessidade da instituição reavaliar conceitos, projetos e ações que estão sendo efetivados na área, buscando encontrar alternativas que favoreçam a formação de ambientes para processos de trabalho mais saudáveis
Finishing the euchromatic sequence of the human genome
The sequence of the human genome encodes the genetic instructions for human physiology, as well as rich information about human evolution. In 2001, the International Human Genome Sequencing Consortium reported a draft sequence of the euchromatic portion of the human genome. Since then, the international collaboration has worked to convert this draft into a genome sequence with high accuracy and nearly complete coverage. Here, we report the result of this finishing process. The current genome sequence (Build 35) contains 2.85 billion nucleotides interrupted by only 341 gaps. It covers ∼99% of the euchromatic genome and is accurate to an error rate of ∼1 event per 100,000 bases. Many of the remaining euchromatic gaps are associated with segmental duplications and will require focused work with new methods. The near-complete sequence, the first for a vertebrate, greatly improves the precision of biological analyses of the human genome including studies of gene number, birth and death. Notably, the human enome seems to encode only 20,000-25,000 protein-coding genes. The genome sequence reported here should serve as a firm foundation for biomedical research in the decades ahead
Stabilizing mandibular complete dentures by a single midline implant-influence on quality of life: 2-year results from a randomized clinical trial comparing different loading protocols.
OBJECTIVES
The knowledge about the influence of dental treatment on health-related quality of life (HRQoL) is still limited. The aim of this multicenter randomized controlled clinical trial was to assess the effect of stabilizing an existing complete denture, by means of a single mandibular implant, on HRQoL. Furthermore, the impact of the loading protocol, i.e., immediate or delayed loading, in edentulous patients was evaluated.
METHODS
One hundred fifty-eight participants aged 60-89 years were randomly assigned to study group A (immediate loading; n = 81) and to group B (delayed loading; n = 78). All participants received a single midline implant in the mandible. The implants were either immediately loaded (group A) or after a closed healing period of 3 months (group B) by connecting the existing mandibular complete dentures to ball attachments. HRQoL was assessed with the Short Form-36 questionnaire of health (SF-36) at baseline, 4 months, and 24 months after implant loading.
RESULTS
Improvement of HRQoL by means of a single implant-retained mandibular overdenture could not be demonstrated after 4 and 24Â months of implant loading. Furthermore, the application of two different loading protocols did not influence HRQoL ratings of study participants.
CONCLUSION
The loading protocol is not a factor, influencing HRQoL in patients treated by a single midline implant in the edentulous mandible.
CLINICAL RELEVANCE
A single midline implant in the edentulous mandible, stabilizing a mandibular complete denture, cannot be recommended for improving HRQoL
