    Dynamic regulation of serum aryl hydrocarbon receptor agonists in MS

    Objective: Several factors influence the clinical course of autoimmune inflammatory diseases such as MS and inflammatory bowel disease. Only recently, the complex interaction between the gut microbiome, dietary factors, and metabolism has started to be appreciated with regard to its potential to modulate acute and chronic inflammation. One of the molecular sensors that mediates the effects of these environmental signals on the immune response is the aryl hydrocarbon receptor (AHR), a ligand-activated transcription factor with key functions in immune cells. Methods: In this study, we analyzed the levels of AHR agonists in serum samples from patients with MS and healthy controls in a case-control study. Results: We detected a global decrease of circulating AHR agonists in relapsing-remitting MS patients as compared to controls. However, during acute CNS inflammation in clinically isolated syndrome or active MS, we measured increased AHR agonistic activity. Moreover, AHR ligand levels in patients with benign MS with relatively mild clinical impairment despite longstanding disease were unaltered as compared to healthy controls. Conclusions: Collectively, these data suggest that AHR agonists in serum are dynamically modulated during the course of MS.     Predicting the physicochemical properties and geographical ORIGIN of lentils using near infrared spectroscopy

    [EN]Calibration statistical descriptors for both whole and ground lentils using Near Infrared Spectroscopy (NIRS), combined with fiber-optic probe, are presented and discussed. The models were developed for estimating the weight, size, total raw protein, moisture, total fat, total fiber, and ash. Standard methods were used to determine compositional parameters of 42 samples of different varieties of lentils. The calibration curves show a wide range of validity for all parameters. The results showed excellent predictability for the determination of weight, fiber, and ash in whole lentils. However, size, moisture, and total fat were predicted satisfactorily in ground lentils. The total protein content could be predicted for both whole and ground lentils. Moreover, NIRS and Direct Partial Least Squares (DPLS) were used to determine whether a sample of lentils belonged to the Protected Geographical Indication (PGI) “Lenteja de La Armuña” or not. The results showed that 95% of the samples were correctly classified as belonging to a PGI. This result demonstrates that this technique allows the differentiation of samples from nearby regions

    Influence of the electric field on the latent heat of the ferroelectric phase transition in KDP

    The specific heat, heat flux (DTA trace) and dielectric constant of KDP ferroelectric crystal have been measured simultaneously for various electric fields with a conduction calorimeter. The specific heat presents a strong anomaly but these simultaneous measurements allow us to evaluate the latent heat accurately. Latent heat decreases with field and the value of critical electric field --that where latent heat disappears-- is estimated to be (0.44\pm0.03) kV/cm. Incidentally, we have measured simultaneously the dielectric permittivity which suggests that latent heat is developed as domains are growing.Comment: 7 pages, 6 figures, ReVTeX, twocolumn format, to appear in J.     Estudio epidemiológico de los casos de triquinelosis registrados en la provincia de Santa Fe, Argentina, 1998?2009

    Objetivo. Caracterizar desde los puntos de vista epidemiológico, clínico y de laboratorio los casos y brotes de triquinelosis que se registraron en la provincia de Santa Fe, Argentina, entre 1998 y 2009. Métodos. Se analizaron 1 519 fichas epidemiológicas de personas que presentaron síntomas y signos compatibles con triquinelosis en la provincia de Santa Fe en el período 1998?2009. Se realizó el análisis estadístico descriptivo de la información contenida en las fichas epidemiológicas y los resultados clínicos y de laboratorio; se aplicó el análisis bifactorial para determinar las posibles asociaciones con factores epidemiológicos y ambientales. Resultados. Se encontraron síntomas de triquinelosis en 1 276 casos; 372 cumplían la definición de caso clínico y 224 muestras resultaron positivas por inmunofluorescencia indirecta, independientemente de su clasificación como caso clínico. Se identificaron 27 brotes que involucraron a 1 157 casos en ocho departamentos, con mayor frecuencia en el centro y el sur de la provincia. La distribución anual de los casos asociados epidemiológicamente entre sí fue heterogénea, con una mayor frecuencia en el período 2000?2003 y en los meses de agosto a octubre. Hubo mayor probabilidad de enfermar cuando se consumieron productos de fuentes informales (OR = 3,69; P = 0,014) y en las áreas rurales (OR = 1,799; P = 0,011). El período de incubación de la enfermedad (mediana) fue de 12 días. Las personas que consumieron carne o derivados de cerdo tuvieron 2,06 veces más riesgo de enfermar que las que no la consumieron. Conclusiones. El mayor número de brotes ocurrió en los departamentos más poblados y con mayor producción porcina. Se deben diseñar acciones específicas de prevención y control de la enfermedad en esos departamentos.     Synthesis and characterization of adsorbents for the elimination of nitrates and bromates from water aiming to develop a continuous oxyanion water elimination system

    It is known that the excess of oxyanions such as NO 3 and BrO 3 in drinking water affects its quality. In this work, three adsorbents (montmorillonite (Mt), silica (Si), and diatomaceous earth) loaded with hexadecyl- (H) and octadecyl-trimethylammonium (O) were used to remove these oxyanions from aqueous solutions by adsorption. In batch systems, the highest NO 3 removal was obtained with Mt modified with H and O (Mt-H and Mt-O), attaining 33% and 50%, respectively, while for BrO 3 removal Si modified with H and O, Si-H and Si-O samples, reached 38% and 42%, respectively. A direct relationship between the adsorption capacity of NO 3 and BrO 3 and the mass of the adsorbent was found in column filtration tests with Mt-O and Mt-H samples in standard solution and real groundwater samples. The adsorption capacity of the column, in the groundwater sample, remained constant after two reuses.The results obtained are promising for the development of a continuous oxyanion removal system containing the low-cost clay Mt modified with either H or O.Fil: Azaro, M..     Adsorption of picloram herbicide on iron oxide pillared montmorillonite

    The adsorption of picloram on Fe Oxide Pillared Clays (PILC) as a possible basis for the development of adsorbent materials for the remediation of polluted environments was studied. To this end, after characterizing PILC obtained from raw clay from Wyoming, USA by XRD, elemental analysis, Mössbauer spectroscopy, DTA, TGA and N2 adsorption (BET), the picloram adsorption was studied at constant pH and ionic strength after 48 h equilibration with an aqueous solution of PCM. PCM adsorbed samples were characterized by SEM, zeta-potential vs pH functions, FTIR and TGA-DTA and the adsorption was modellated using Langmuir and Freundlich models. The results reveal that the surface area and interlayer space of PILC are bigger than those of raw montmorillonite. An analysis of the adsorption isotherms of picloram on the PILC showed that picloram adsorption increases with decreasing pH. The adsorption of PCM on PILC was 150 times higher than that found for raw montmorillonite under similar conditions. The zeta-potential vs pH functions shape and FTIR indicated that PCM coordinates the surface iron centers throughout pyridinic nitrogen atom forming inner-sphere complexes. The much greater PCM adsorption on PILC turns the process into an excellent candidate to be used for dosing or remediation of pesticides.Fil: Marco Brown, José Luis.     Interacting resident epicardium-derived fibroblasts and recruited bone marrow cells form myocardial infarction scar

    Background: Although efforts continue to find new therapies to regenerate infarcted heart tissue, knowledge of the cellular and molecular mechanisms involved remains poor. Objectives: This study sought to identify the origin of cardiac fibroblasts (CFs) in the infarcted heart to better understand the pathophysiology of ventricular remodeling following myocardial infarction (MI). Methods: Permanent genetic tracing of epicardium-derived cell (EPDC) and bone marrow-derived blood cell (BMC) lineages was established using Cre/LoxP technology. In vivo gene and protein expression studies, as well as in vitro cell culture assays, were developed to characterize EPDC and BMC interaction and properties. Results: EPDCs, which colonize the cardiac interstitium during embryogenesis, massively differentiate into CFs after MI. This response is disease-specific, because angiotensin II-induced pressure overload does not trigger significant EPDC fibroblastic differentiation. The expansion of epicardial-derived CFs follows BMC infiltration into the infarct site; the number of EPDCs equals that of BMCs 1 week post-infarction. BMC-EPDC interaction leads to cell polarization, packing, massive collagen deposition, and scar formation. Moreover, epicardium-derived CFs display stromal properties with respect to BMCs, contributing to the sustained recruitment of circulating cells to the damaged zone and the cardiac persistence of hematopoietic progenitors/stem cells after MI. Conclusions: EPDCs, but not BMCs, are the main origin of CFs in the ischemic heart. Adult resident EPDC contribution to the CF compartment is time- and disease-dependent. Our findings are relevant to the understanding of post-MI ventricular remodeling and may contribute to the development of new therapies to treat this disease.1.     Radiomarcaje y estudios de biodistribución de nanopartículas poliméricas como adyuvantes para la vacunación oftálmica frente a la brucelosis

    Objetivos: Optimizar el radiomarcaje con 99mTc de nanopartículas de Gantrez® manosiladas y cargadas con el antígeno de Brucella Ovis (Man-NP-HS) y llevar a cabo estudios de biodistribución en ratón tras la administración de las nanopartículas por vía ocular. Metodología: Las Man-NP-HS se obtuvieron por el método de desplazamiento de disolvente. Se purificaron, liofilizaron y caracterizaron. A continuación, se marcaron con 74 MBq de 99mTcO4 - previamente reducido con una disolución ácida de cloruro de estaño, trabajando en ausencia de oxígeno y con un pH final de 4. El rendimiento del marcaje se evaluó mediante TLC. Los estudios de biodistribución se llevaron a cabo en ratones tras la administración oftálmica de la formulación y de un control de 99mTcO4 - libre. Para ello, se sacrificaron los animales a las 2 y a las 24 horas tras la administración ocular y se contaron los órganos en un contador gamma. Resultados: Se obtuvo un rendimiento de marcaje superior al 90%. Los estudios de biodistribución de 99mTc-Man-NP-HS permitieron detectar la actividad concentrada en mucosa nasal y ocular y tracto gastrointestinal tanto a las 2 como a las 24 horas, frente a la biodistribución de 99mTcO4 - libre que permaneció concentrado en la piel alrededor del ojo y en tracto gastrointestinal. Conclusión: Los estudios de biodistribución de 99mTc-Man-NP-HS tras administración oftálmica han permitido demostrar su biodistribución en mucosas y tracto gastrointestinal, característica indispensable como sistema de liberación de antígenos a través de mucosa ocular. Esto, junto con su elevada respuesta inmune, efectiva protección y no virulencia, convierte a estas nanopartículas en una vacuna ideal anti Brucelosis

    Conjunctival vaccination against Brucella ovis in mice with mannosylated nanoparticles

    The use of sub-unit vaccines can solve some drawbacks associated with traditional attenuated or inactivated ones. However, in order to improve their immunogenicity, these vaccines needs to be associated to an appropriate adjuvant which, adequately selected, may also offer an alternative pathway for administration. The aim of this work was to evaluate the protection offered by the hot saline complex extracted from Brucella ovis (HS) encapsulated in mannosylated nanoparticles (MAN-NP-HS) when instilled conjunctivally in mice. Nanoparticles displayed a size of 300 nm and the antigen loading was close to 30 μg per mg nanoparticle. Importantly, encapsulated HS maintained its protein profile, structural integrity and antigenicity during and after the preparative process of nanoparticles. The ocular immunization was performed on BALB/c mice. Eight weeks after vaccination animals were challenged with B. ovis, and 3 weeks later, were slaughtered for bacteriological examinations. Animals immunized with MAN-NP-HS displayed a 3-log reduction in spleen CFU compared with unvaccinated animals. This degree of protection was significantly higher than that observed for the commercial vaccine (Rev1) subcutaneously administered. Interestingly, the mucosal IgA response induced by MAN-NP-HS was found to be much more intense than that offered by Rev1 and prolonged in time. Furthermore, the elicited IL-2, IL-4 and γ-IFN levels showed good correlation with the degree of protection. On the other hand, biodistribution studies in animals were performed with nanoparticles labelled with either 99mtechnetium or rhodamine B isothiocyanate. The biodistribution revealed that, after instillation, MAN-NP-HS moved from the palpebral area to the nasal region and, the gastrointestinal tract. This profile of distribution was different to that observed for free 99mTcO4− colloids, which remained for at least 24 h in the site of administration. In summary, mannosylated nanoparticles appear to be a safe and suitable adjuvant for conjunctival vaccination
