The initial objective was elucidation of the molecular mechanisms underlying insect metamorphosis and embryonic diapause in respect to the expression of neuropeptide hormones and their physiological effects and we consider to have reached the goal at an appreciable level.Bombyxin, an insulin-related neuropeptide, consists of 30 gene copies and their expression pattern in Bombyx genome and the nucleotide sequences of upstream region of all the 30 genes have been accomplished. Bombyxin receptor has also been elucidated by cDNA cloning to possess the tyrosine kinase domein similar to insulin receptor. Ultra-micro assay method of bombyxin was developed using time-resolved fluoroimmunoassay (TR-FIA) which enabled to quantify a very small amount of the hormone as low as few atto-moles and to determine the detailed changes in hemolymph bombyxin concentrations through the larval-pupal-adult period. The same method became applicable to measurement of hemolymph PTTH titer, which showed a daily secretion of PTTH even before head critical period (HCP). PTTH secretion is stimulated by a neurotransmitter, acetylcholine, indicating that acetylcholine neuron may be involved in the regulation of PTTH cells. As a part of elucidation of molecular mechanisms of PTTH stimulation of prothoracic glands, purification of PTTH receptor and elucidation of its physical properties are in progress. Expression dynamics of diapause hormone (DH) mRNA showed to be different in embryonic and larval stages of animals that are destined to produce diapausing eggs from those to produce non-diapausing eggs. One of the major DH effects was involved in the expression of trehalase gene in ovary. DH gene was expressed in 3 clusters of neurosecretory cells in suboesophageal ganglion. DH is processed and secreted from the posterior cluster while pheromone biosynthesis activating neurohormone which is processed from the same precursor molecule as DH is secreted from the anterior and middle clusters. Also, we showed the gene response to 20E in vitro using anterior silk glands of gut-purged larvae. The in vivo and in vitro results indicated that a heterodimeric EcR-B1 and USP-2 may be responsible for the cell death Results also indicated involvement of E74, E75, BHR3 and BR-C isoforms, but not Ftz-F1. We are not succeeded in gene cloning of the putative membrane ecdysone receptor yet. We examined pharmacologically the signaling pathway from mEcR to cellular responses, i.e. cell condensation, nuclear condensation, DNA fragmentation and nuclear fragmentation. Ca acts as the second messenger The mEcR is suggested to be a G-protein coupled receptor (GPCR) associated with Gαq, followed by a serial activation of phospholipase c-β, generation of inositol 3-phosphate (IP_3), and release of Ca from endoplasmic reticulum probably through IP3 receptor Then, Ca activates protein kinase C (PKC) and caspase 3-like protease. This signaling pathway culminates in nuclear fragmentation and nuclear fragmentation. Nuclear condensation is regulated by a different pathway involving calmodulin and calmodulin-dependent protein kinase II (CaMK-II). However, this pathway was not activated by Ca, and therefore it is unknown whether Gαq is involved in this pathway. Rectal sac distention is induced by 20-hydroxyecdysone in the pupa of Bombyx mori
Holometabolous insects do not excrete but store metabolic wastes during the pupal period. The waste is called meconium and is purged after adult emergence. Although the contents of meconium are well-studied, the developmental and physiological regulation of meconium accumulation is poorly understood. In Bombyx mori, meconium is accumulated in the rectal sac; thereby, the rectal sac distends at the late pupal stage. Here, we show that rectal sac distention occurs between 4 and 5 days after pupation. The distention is halted by brain-removal just after larval-pupal ecdysis but not by brain-removal 1 day after pupation. In the pupae, brain-removal just after ecdysis kept the hemolymph ecdysteroid titer low during early and mid-pupal stages. An injection of 20-hydroxyecdysone (20E) evoked the distention that was halted by brain-removal in a dose-dependent manner. Therefore, brain-removal caused the lack of ecdysteroid, and rectal sac distention did not appear in the brain-removed pupae because of the lack of ecdysteroid. We conclude that rectal sac distention is one of the developmental events regulated by 20E during the pupal period in B. mori. © 2008 Elsevier Ltd. All rights reserved
