    Outer Rotation Curve of the Galaxy with VERA I: Trigonometric parallax of IRAS 05168+3634

    We report measurement of trigonometric parallax of IRAS 05168+3634 with VERA. The parallax is 0.532 +/- 0.053 mas, corresponding to a distance of 1.88+0.21/-0.17 kpc. This result is significantly smaller than the previous distance estimate of 6 kpc based on kinematic distance. This drastic change in the source distance revises not only physical parameters of IRAS 05168+3634, but also its location of the source, placing it in the Perseus arm rather than the Outer arm. We also measure proper motions of the source. A combination of the distance and the proper motions with systemic velocity yields rotation velocity ({\Theta}) of 227+9/-11 km s-1 at the source, assuming {\Theta}0 = 240 km s-1. Our result combined with previous VLBI results for six sources in the Perseus arm indicates that the sources rotate systematically slower than the Galactic rotation velocity at the LSR. In fact, we show observed disk peculiar motions averaged over the seven sources in the Perseus arm as (Umean, Vmean) = (11 +/- 3, -17 +/- 3) km s-1, indicating that these seven sources are systematically moving toward the Galactic center, and lag behind the Galactic rotation.Comment: 11 pages, 4 figures and 6 tables, accepted for the publication in PAS

    Retrieving Photorecombination Cross Sections of Atoms from High-Order Harmonic Spectra

    We observe high-order harmonic spectra generated from a thin atomic medium, Ar, Kr, and Xe, by intense 800-nm and 1300-nm femtosecond pulses. A clear signature of a single-atom response is observed in the harmonic spectra. Especially in the case of Ar, a Cooper minimum, reflecting the electronic structure of the atom, is observed in the harmonic spectra. We successfully extract the photorecombination cross sections of the atoms in the field-free condition with the help of an accurate recolliding electron wave packet. The present protocol paves the way for exploring ultrafast imaging of molecular dynamics with attosecond resolution

    Detection of Lung Cancer Cells in Solutions Using a Terahertz Chemical Microscope

    Cancer genome analysis has recently attracted attention for personalized cancer treatment. In this treatment, evaluation of the ratio of cancer cells in a specimen tissue is essential for the precise analysis of the genome. Conventionally, the evaluation takes at least two days and depends on the skill of the pathologist. In our group, a terahertz chemical microscope (TCM) was developed to easily and quickly measure the number of cancer cells in a solution. In this study, an antibody was immobilized on a sensing plate using an avidin-biotin reaction to immobilize it for high density and to improve antibody alignment. In addition, as the detected terahertz signals vary depending on the sensitivity of the sensing plate, the sensitivity was evaluated using pH measurement. The result of the cancer cell detection was corrected using the result of pH measurement. These results indicate that a TCM is expected to be an excellent candidate for liquid biopsies in cancer diagnosis

    Serotonin enhances the production of type IV collagen by human mesangial cells.BackgroundThe plasma concentration of 5-hydroxytryptamine (5-HT) in diabetic patients is higher than that in normal subjects. Since recent reports have demonstrated the presence of 5-HT2A receptor in glomerular mesangial cells, it is possible that 5-HT may be involved in the development of diabetic nephropathy through the 5-HT2A receptor in mesangial cells. Because expansion of the glomerular mesangial lesion is a characteristic feature of diabetic nephropathy, we examined the effect of 5-HT on the production of type IV collagen by human mesangial cells.MethodsHuman mesangial cells were incubated with 5-HT with or without 5-HT receptor antagonists, protein kinase C (PKC) inhibitor or transforming growth factor-β (TGF-β) antibody. Type IV collagen mRNA and protein concentration in medium were measured by Northern blot analysis and enzyme-linked immunosorbent assay (ELISA), respectively. TGF-β mRNA and bioactivity in the medium were measured by Northern blot analysis and bioassay using mink lung epithelial cells, respectively.Results5-HT stimulated the production of type IV collagen by human mesangial cells, which was inhibited by ketanserin and sarpogrelate hydrochloride, 5-HT2A receptor antagonists, but not by ondansetron, a 5-HT3 receptor antagonist. 5-HT increased the bioactivities of both active and total TGF-β. However, the 5-HT-enhanced production of type IV collagen was completely inhibited by an anti-TGF-β antibody. Furthermore, a PKC inhibitor, calphostin C, inhibited the 5-HT-induced increase in type IV collagen secretion, and the activity of membrane PKC was increased by 5-HT. Phorbol ester activated type IV collagen production as well as active and total TGF-β. Calphostin C completely inhibited the 5-HT-enhanced activity of active TGF-β, but did not inhibit exogenous TGF-β-induced increase in type IV collagen secretion.ConclusionsOur results suggest that 5-HT-enhanced production of type IV collagen by human mesangial cells is mediated by activation of PKC and subsequent increase in active TGF-β activity
