    Genetic characterization of alpine sheep breeds

    The aim of this study was to characterize from the genetic point of view eight alpine sheep breeds reared in Italy (Bergamasca, Biellese, Schwarzbraunes Bergschaf and Tiroler Bergschaf), Germany (Brillenschaf and Weisses Bergschaf) and Slovenia (Bov\u161ka and Jezerzko-Sol\u10davska) through the use of microsatellite molecular markers. Allelic richness was rather high in each breed highlighting a considerable genetic diversity. However, the study evidenced a significant departure from Hardy-Weinberg equilibrium in all analyzed breeds caused by a heterozygote deficiency. Such lack seems to be caused by a rather high level of inbreeding. The genetic differentiation among breeds was rather low (F ST = 0.064) but significant. The neighbour-joining tree obtained from Reynolds\u2019 genetic distance estimates, showed the presence of three groups formed by the three Bergschaf breeds, the Italian Bergamasca and Biellese and the two Slovenian breeds together with the German Brillenschaf. Such grouping is in accordance with the breeds\u2019 region of origin and with their known history. Concluding, microsatellite resulted to be a useful tool to investigate breed variability and to characterize alpine sheep breeds. Obtained findings suggest the need to set up a conservation plan aiming to safeguard and increase the genetic variability of the studied breeds compromised by the high level of inbreeding. Microsatellites genotyping could help to monitor breed variability and to organize matings

    Effect of dietary nitrogen level and source on mRNA expression of urea transporters in the rumen epithelium of fattening bulls

    This paper aims to study the effect of the dietary treatments on mRNA expression of urea transporter B (UT-B) and some aquaporins (AQP) in rumen epithelium of Italian Simmental young bulls. Eighty animals allocated to 16 pens were fed from about 500 to 650 kg body weight with four experimental diets, which resulted from the combination of two crude protein levels (125 and 110 g/kg dry matter, diets M and L, respectively) and two nitrogen sources (soybean meal (SBM) or SBM partly replaced by an isonitrogenous mixture of corn and urea; diets −U and +U, respectively). At slaughtering samples of blood and rumen epithelium were collected from six bulls for each diet. Blood samples were analysed for haematological parameters and quantitative PCR was carried out on the mRNA extracted from the rumen epithelium samples. The bulls fed diets M had lower plasma concentrations of aspartate aminotransferase than those receiving diets L (78.9 vs. 88.3 U/l, p = 0.04). Plasma urea was higher (p = 0.03) for diets M and lower for diets +U (2.0 vs. 2.5 and 1.73 vs. 2.00 mmol/l, respectively, in M and L diets, p = 0.04). The effect of dietary treatments on rumen UT expression were limited to AQP3, which was down regulated (p = 0.01) in diets +U. Finally, a high positive correlation (R2 = 0.871) between the expressions of AQP7 and AQP10 was found. In conclusion, the AQP3 appears very responsive to dietary treatments and therefore it is a candidate to be further studied in rumen metabolism experiments. The close relationship between mRNA expression of AQP7 and AQP10 indicates a similar function of these two proteins

    Variability in the characteristics of fresh meat and thighs in relationship to genetic type of the heavy pig

    The variability in the characteristics of fresh meat and thighs in relationship with genetic type was studied on 22 lots of heavy pigs, comprising 615 female and castrated males, randomly chosen during slaughtering in groups of 20 to 30 animals per lot. Four different genetic types reared for the production of the San Daniele dry-cured ham PDO were considered: two traditionals (Large White or Duroc x (Landrace x Large White) and two industrial hybrids (GOLAND and DANBRED). Fresh meat samples from longissimus dorsi muscle were collected to perform chemical and physical analysis. The right trimmed thighs were used to evaluate some morphological parameters along with weight losses during seasoning. The genetic type confirmed its important influence on pig carcass, meat and ham quality. The two industrial types, DANBRED in particular, tended to present a lower back fat values and a leaner carcass than the two Italian genetic types. Within the most important characteristics for the production of dry cured hams (subcutaneous fat thickness and seasoning losses) the highest differences were found among the industrials genetic types, which cannot be simply considered as an undifferentiated homogenous group

    Understanding the Pathogenesis of Red Mark Syndrome in Rainbow Trout (Oncorhynchus mykiss) through an Integrated Morphological and Molecular Approach

    Red mark syndrome (RMS) is a widespread skin disorder of rainbow trout in freshwater aquaculture, believed to be caused by a Midichloria-like organism (MLO). Here, we aimed to study the pathologic mechanisms at the origin of RMS by analyzing field samples from a recent outbreak through gene expression, MLO PCR, quantitative PCR, and a histopathological scoring system proposed for RMS lesions. Statistical analyses included a One-Way Analysis of Variance (ANOVA) with a Dunnett’s multiple comparisons test to assess differences among gene expression groups and a nonparametric Spearman correlation between various categories of skin lesions and PCR results. In short, the results confirmed the presence of a high quantity of 16S gene copy numbers of Midichloria-like organisms in diseased skin tissues. However, the number of Midichloria-like organisms detected was not correlated to the degree of severity of skin disease. Midichloria-like organism DNA was found in the spleen and head kidney. The spleen showed pathologic changes mainly of hyperplastic type, reflecting its direct involvement during infection. The most severe skin lesions were characterized by a high level of inflammatory cytokines sustaining and modulating the severe inflammatory process. IL-1 β, IL-6, IL-10, MHC-II, and TCR were upregulated in severe skin lesions, while IL-10 was highly expressed in moderate to severe ones. In the moderate form, the response was driven to produce immunoglobulins, which appeared crucial in controlling the skin disease’s severity. Altogether our results illustrated a complex immune interaction between the host and Midichloria-like organism

    The aim of this study was to assess the effect of heat stress on dairy cow performance and on the expression of selected genes involved in milk protein metabolism. Eight Italian Holstein Friesian cows were kept under thermoneutral conditions (temperature\u2013humidity index (THI) 0.05), CSN3 (p > 0.05), HSPA8 (p > 0.05), and STAT5B (p > 0.05) mRNA. Mild heat stress reduced the performance of dairy cows without affecting the expression of genes coding for caseins

    Ataxia with oculomotor apraxia type 2: a clinical, pathologic, and genetic study

    BACKGROUND: Ataxia with oculomotor apraxia type 2 (AOA2) is characterized by onset between age 10 and 22 years, cerebellar atrophy, peripheral neuropathy, oculomotor apraxia (OMA), and elevated serum alpha-fetoprotein (AFP) levels. Recessive mutations in SETX have been described in AOA2 patients. OBJECTIVE: To describe the clinical features of AOA2 and to identify the SETX mutations in 10 patients from four Italian families. METHODS: The patients underwent clinical examination, routine laboratory tests, nerve conduction studies, sural nerve biopsy, and brain MRI. All were screened for SETX mutations. RESULTS: All the patients had cerebellar features, including limb and truncal ataxia, and slurred speech. OMA was observed in two patients, extrapyramidal symptoms in two, and mental impairment in three. High serum AFP levels, motor and sensory axonal neuropathy, and marked cerebellar atrophy on MRI were detected in all the patients who underwent these examinations. Sural nerve biopsy revealed a severe depletion of large myelinated fibers in one patient, and both large and small myelinated fibers in another. Postmortem findings are also reported in one of the patients. Four different homozygous SETX mutations were found (a large-scale deletion, a missense change, a single-base deletion, and a splice-site mutation). CONCLUSIONS: The clinical phenotype of oculomotor apraxia type 2 is fairly homogeneous, showing only subtle intrafamilial variability. OMA is an inconstant finding. The identification of new mutations expands the array of SETX variants, and the finding of a missense change outside the helicase domain suggests the existence of at least one more functional region in the N-terminus of senataxin

    Characteristics of carcass, meat and subcutaneous fat of Mangalica pigs reared outdoors

    Importance of the work: Mangalica pig meat has a reputation for superior quality. Little information is available on this breed reared for heavy pig production. Objectives: To evaluate the effects of sex on the carcass, loin and lard characteristics (inner and outer layers) of Blonde Mangalica pigs. Materials & Methods: In total, 15 Blonde Mangalica pigs were considered (10 castrated males and 5 females) that were reared together outdoors. The pigs had ad libitum access to the same diet and were slaughtered at age 22.9 mth, with an average carcass weight of 141.4 kg. Results: Sex did not affect the carcass, meat or lard characteristics. In general, the Mangalica pigs had an average fat thickness of 70 mm and a loin cholesterol content of 0.674 mg/g. The loin had 39.0%, 52.5% and 8.5% saturated (SFA), monounsaturated fatty acids and polyunsaturated fatty acids (PUFA), respectively. Significant differences were found in the characteristics of the two lard layers. In particular, the inner layer was brighter and had a higher dry matter content than the outer layer. Regarding the fatty acids content, the inner layer had a lower PUFA but higher SFA content. Main finding: Sex did not influence the carcass, meat or lard characteristics of the Mangalica pigs. The results improved the characterization and filled knowledge gaps regarding meat and lard quality parameters of heavy Mangalica pigs reared outdoors
