    The walkthrough method : an approach to the study of apps

    Software applications (apps) are now prevalent in the digital media environment. They are the site of significant sociocultural and economic transformations across many domains, from health and relationships to entertainment and everyday finance. As relatively closed technical systems, apps pose new methodological challenges for sociocultural digital media research. This paper describes a method, grounded in a combination of science and technology studies with cultural studies, through which researchers can perform a critical analysis of a given app. The method involves establishing an app’s environment of expected use by identifying and describing its vision, operating model, and modes of governance. It then deploys a walkthrough technique to systematically and forensically step through the various stages of app registration and entry, everyday use, and discontinuation of use. The walkthrough method establishes a foundational corpus of data upon which can be built a more detailed analysis of an app’s intended purpose, embedded cultural meanings, and implied ideal users and uses. The walkthrough also serves as a foundation for further user-centred research that can identify how users resist these arrangements and appropriate app technology for their own purposes

    Components of SurA Required for Outer Membrane Biogenesis in Uropathogenic Escherichia coli

    Background: SurA is a periplasmic peptidyl-prolyl isomerase (PPIase) and chaperone of Escherichia coli and other Gramnegative bacteria. In contrast to other PPIases, SurA appears to have a distinct role in chaperoning newly synthesized porins destined for insertion into the outer membrane. Previous studies have indicated that the chaperone activity of SurA rests in its ‘‘core module’ ’ (the N- plus C-terminal domains), based on in vivo envelope phenotypes and in vitro binding and protection of non-native substrates. Methodology/Principal Findings: In this study, we determined the components of SurA required for chaperone activity using in vivo phenotypes relevant to disease causation by uropathogenic E. coli (UPEC), namely membrane resistance to permeation by antimicrobials and maturation of the type 1 pilus usher FimD. FimD is a SurA-dependent, integral outer membrane protein through which heteropolymeric type 1 pili, which confer bladder epithelial binding and invasion capacity upon uropathogenic E. coli, are assembled and extruded. Consistent with prior results, the in vivo chaperone activity of SurA in UPEC rested primarily in the core module. However, the PPIase domains I and II were not expendable for wild-type resistance to novobiocin in broth culture. Steady-state levels of FimD were substantially restored in the UPEC surA mutant complemented with the SurA N- plus C-terminal domains.     Acute physiological stress down-regulates mRNA expressions of growth-related genes in coho salmon

    Growth and development in fish are regulated to a major extent by growth-related factors, such as liver-derived insulin-like growth factor (IGF) -1 in response to pituitary-secreted growth hormone (GH) binding to the GH receptor (GHR). Here, we report on the changes in the expressions of gh, ghr, and igf1 genes and the circulating levels of GH and IGF-1 proteins in juvenile coho salmon (Oncorhynchus kisutch) in response to handling as an acute physiological stressor. Plasma GH levels were not significantly different between stressed fish and prestressed control. Plasma IGF-1 concentrations in stressed fish 1.5 h post-stress were the same as in control fish, but levels in stressed fish decreased significantly 16 h post-stress. Real-time quantitative PCR (qPCR) analysis showed that ghr mRNA levels in pituitary, liver, and muscle decreased gradually in response to the stressor. After exposure to stress, hepatic igf1 expression transiently increased, whereas levels decreased 16 h post-stress. On the other hand, the pituitary gh mRNA level did not change in response to the stressor. These observations indicate that expression of gh, ghr, and igf1 responded differently to stress. Our results show that acute physiological stress can mainly down-regulate the expressions of growth-related genes in coho salmon in vivo. This study also suggests that a relationship between the neuroendocrine stress response and growth-related factors exists in fish.Peer reviewed: YesNRC publication: Ye

    Habitat attributes of landscape mosaics along a gradient of matrix development intensity: Matrix management matters

    The matrix is an important element of landscape mosaics that influences wildlife indirectly through its influence on habitat, and directly, if they live in ormove through it.Therefore, to quantify andmanage habitat quality for wildlife in modified landscapes, it is necessary to consider the characteristics of both patch and matrix elements of the whole landscape mosaic. To isolate matrix effects from the often simultaneous and confounding influence of patch and landscape characteristics, we identified nineteen 500 mradius landscapes in southeastQueensland, Australiawith similar remnant forest patch attributes, habitat loss, and fragmentation, but exhibiting a marked gradient from rural through high-density suburban development of the matrix, quantified by a weighted road-length metric. We measured habitat disturbance, structure, and floristics in patch core, patch edge andmatrix landscape elements to characterise how landscape habitat quality changes for small mammals. Correlation analyses identified that with increased matrix development intensity, human disturbance of core sites increased, predators and exotic plant species richness in matrix sites increased, and structural complexity (e.g. logs and stumps) in the matrix decreased. Ordination analyses showed landscape elements were most similar in habitat structure and floristics at low to moderate levels of matrix development, suggesting enhanced landscape habitat quality. Matrix development intensity was not, however, the greatest source of overall variation of habitat throughout landscapes. Many variables, such as landholder behaviour, complicate the relationship. For enhanced conservation outcomes the matrix needs to be managed to control disturbances and strategically plan for matrix habitat retention and restoration
