Adhesive and degradative properties of human placental cytotrophoblast cells in vitro.
Human fetal development depends on the embryo rapidly gaining access to the maternal circulation. The trophoblast cells that form the fetal portion of the human placenta have solved this problem by transiently exhibiting certain tumor-like properties. Thus, during early pregnancy fetal cytotrophoblast cells invade the uterus and its arterial network. This process peaks during the twelfth week of pregnancy and declines rapidly thereafter, suggesting that the highly specialized, invasive behavior of the cytotrophoblast cells is closely regulated. Since little is known about the actual mechanisms involved, we developed an isolation procedure for cytotrophoblasts from placentas of different gestational ages to study their adhesive and invasive properties in vitro. Cytotrophoblasts isolated from first, second, and third trimester human placentas were plated on the basement membrane-like extracellular matrix produced by the PF HR9 teratocarcinoma cell line. Cells from all trimesters expressed the calcium-dependent cell adhesion molecule cell-CAM 120/80 (E-cadherin) which, in the placenta, is specific for cytotrophoblasts. However, only the first trimester cytotrophoblast cells degraded the matrices on which they were cultured, leaving large gaps in the basement membrane substrates and releasing low molecular mass 3H-labeled matrix components into the medium. No similar degradative activity was observed when second or third trimester cytotrophoblast cells, first trimester human placental fibroblasts, or the human choriocarcinoma cell lines BeWo and JAR were cultured on radiolabeled matrices. To begin to understand the biochemical basis of this degradative behavior, the substrate gel technique was used to analyze the cell-associated and secreted proteinase activities expressed by early, mid, and late gestation cytotrophoblasts. Several gelatin-degrading proteinases were uniquely expressed by early gestation, invasive cytotrophoblasts, and all these activities could be abolished by inhibitors of metalloproteinases. By early second trimester, the time when cytotrophoblast invasion rapidly diminishes in vivo, the proteinase pattern of the cytotrophoblasts was identical to that of term, noninvasive cells. These results are the first evidence suggesting that specialized, temporally regulated metalloproteinases are involved in trophoblast invasion of the uterus. Since the cytotrophoblasts from first trimester and later gestation placentas maintain for several days the temporally regulated degradative behavior displayed in vivo, the short-term cytotrophoblast outgrowth culture system described here should be useful in studying some of the early events in human place

Summarizing First-Person Videos from Third Persons' Points of Views
Video highlight or summarization is among interesting topics in computer
vision, which benefits a variety of applications like viewing, searching, or
storage. However, most existing studies rely on training data of third-person
videos, which cannot easily generalize to highlight the first-person ones. With
the goal of deriving an effective model to summarize first-person videos, we
propose a novel deep neural network architecture for describing and
discriminating vital spatiotemporal information across videos with different
points of view. Our proposed model is realized in a semi-supervised setting, in
which fully annotated third-person videos, unlabeled first-person videos, and a
small number of annotated first-person ones are presented during training. In
our experiments, qualitative and quantitative evaluations on both benchmarks

Quantitative trait loci analysis for chlorophyll content of cucumber (Cucumis sativus L.) seedlings under low-light stress
An increase in chlorophyll content is an adaptive response to low-light stress and can be used to evaluate low-light tolerance. The effects of low-light stress (100 ìmol·m-2.s-1) on the chlorophyll content of cucumber (Cucumis sativus L.) were investigated in a set of 123 F2:3 lines in the seedling stage in the autumn of 2008 and spring of 2009. Quantitative trait loci (QTL) analysis was undertaken on the basis of a genetic linkage map of the corresponding F2 population that was constructed using composite interval mapping. F2:3-based QTL analysis of the chlorophyll-a (chl.a), chlorophyll-b (chl.b) and chlorophyll-a+b (chl.a+b) content in the 2 environments revealed 21 QTLs located on the linkage groups 1, 2, 3, 4, 6 and 7, which accounted for 4.8 - 17.3% of the phenotypic variation. In the spring of 2009, the total phenotypic variation among the F2:3 lines accounted for by the QTLs for chl.a, chl.b and chl.a+b were 44.5, 29.4 and39.0%, respectively. In the autumn of 2008, 11 QTLs were identified, which accounted for 4.8 - 14.9% of the observed phenotypic variation and an additive effect of -8.10 to 20.85. Four major-effect QTLs (chla2.1, chlb2.2, chlb3.1 and chla+b2.2) were detected under both conditions. The QTL information presented in this research, together with the data from our previous study on heredity of low-light tolerant traits, will facilitate the breeding of low-light-stress-resistant cucumbers
Weakening of the stratospheric polar vortex by Arctic sea-ice loss
Successive cold winters of severely low temperatures in recent years have had critical social and economic impacts on the mid-latitude continents in the Northern Hemisphere. Although these cold winters are thought to be partly driven by dramatic losses of Arctic sea-ice, the mechanism that links sea-ice loss to cold winters remains a subject of debate. Here, by conducting observational analyses and model experiments, we show how Arctic sea-ice loss and cold winters in extra-polar regions are dynamically connected through the polar stratosphere. We find that decreased sea-ice cover during early winter months (November-December), especially over the Barents-Kara seas, enhances the upward propagation of planetary-scale waves with wavenumbers of 1 and 2, subsequently weakening the stratospheric polar vortex in mid-winter (January-February). The weakened polar vortex preferentially induces a negative phase of Arctic Oscillation at the surface, resulting in low temperatures in mid-latitudes.open11167174Ysciescopu
Holonomic surface codes for fault-tolerant quantum computation
© 2018 American Physical Society. Surface codes can protect quantum information stored in qubits from local errors as long as the per-operation error rate is below a certain threshold. Here we propose holonomic surface codes by harnessing the quantum holonomy of the system. In our scheme, the holonomic gates are built via auxiliary qubits rather than the auxiliary levels in multilevel systems used in conventional holonomic quantum computation. The key advantage of our approach is that the auxiliary qubits are in their ground state before and after each gate operation, so they are not involved in the operation cycles of surface codes. This provides an advantageous way to implement surface codes for fault-tolerant quantum computation
RIPEx: Extracting malicious IP addresses from security forums using cross-forum learning
Is it possible to extract malicious IP addresses reported in security forums
in an automatic way? This is the question at the heart of our work. We focus on
security forums, where security professionals and hackers share knowledge and
information, and often report misbehaving IP addresses. So far, there have only
been a few efforts to extract information from such security forums. We propose
RIPEx, a systematic approach to identify and label IP addresses in security
forums by utilizing a cross-forum learning method. In more detail, the
challenge is twofold: (a) identifying IP addresses from other numerical
entities, such as software version numbers, and (b) classifying the IP address
as benign or malicious. We propose an integrated solution that tackles both
these problems. A novelty of our approach is that it does not require training
data for each new forum. Our approach does knowledge transfer across forums: we
use a classifier from our source forums to identify seed information for
training a classifier on the target forum. We evaluate our method using data
collected from five security forums with a total of 31K users and 542K posts.
First, RIPEx can distinguish IP address from other numeric expressions with 95%
precision and above 93% recall on average. Second, RIPEx identifies malicious
IP addresses with an average precision of 88% and over 78% recall, using our
cross-forum learning. Our work is a first step towards harnessing the wealth of
