Phylogeny, ultrastructure, histopathology and prevalence of Myxobolus oliveirai sp. nov., a parasite of Brycon hilarii (Characidae) in the Pantanal wetland, Brazil
This paper presents the morphological, histological and ultrastructural characteristics of Myxobolus oliveirai sp. nov., a parasite of the gill filaments in Brycon hilarii from the Brazilian Pantanal. Out of 216 B. hilariispecimens examined (126 wild and 90 cultivated), 38.1% of wild specimens (n = 48) were infected. The parasites form elongated plasmodia primarily in the tip of gill filaments, reaching about 3 mm in length. A thorough comparison with all the Myxobolus species described from South American hosts, as well as nearly all the Myxobolus species described so far is provided. Partial sequencing of the 18S rDNA gene revealed a total of 1,527 bp. The Myxobolus species parasite of B. hilarii did not match any of the Myxozoa available in GenBank. In the phylogenetic analysis, M. oliveirai sp. nov. composed a monophyletic group with eight other species: five species of Myxobolus parasites of mugilid fishes, two parasites of pangasiid and one of centrarchid. Expression of tumor suppressors miR-195 and let-7a as potential biomarkers of invasive breast cancer
MicroRNAs (miRNAs) are small non-coding RNAs that regulate gene expression at the posttranscriptional level. Some miRNAs, including let-7a and miR-195, have been described as tumor suppressors. However, the roles of these microRNAs in breast cancer progression remain controversial. DIAGNÓSTICO E PROAGNÓSTICO DOS RSU PARA MUNICÍPIO DE SANANDUVA/RS
A falta de uma gestão adequada dos resíduos sólidos urbanos (RSU) tem provocado ao longo dos anos nos Municípios com menos de vinte mil habitantes, diversos prejuízos nas áreas da saúde pública, equidade social e conservação ambiental, além de impactos significativos na ordem econômica em diversos segmentos de atividade. Dentro deste contexto, é necessária a efetiva implantação das diretrizes que preconizam a Política Nacional de Resíduos Sólidos. Com isso, o objetivo deste trabalho foi realizar o diagnóstico e o prognóstico para a gestão de resíduos sólidos urbanos (RSU) do município de Sananduva/RS, e como a proposta de novos métodos de gerenciamento de RSU pode contribuir na redução e adequada destinação de RSU. A metodologia deste trabalho foi dividida em três etapas: revisão literária do tema, avaliação da situação atual de gerenciamento de resíduos sólidos e a análise e cruzamento dos dados obtidos. Os resultados obtidos demonstram que a população urbana do município de Sananduva/RS, no período de set/2012 a ago/2013 teve uma geração de 3.577.440 kg de RSU, o que representa uma produção per capita 0,919 kg.hab-1.d-1 de RSU. Com isto, verifica-se a necessidade da inserção de novas técnicas de tratamento final dos RSU, como a compostagem, coleta seletiva/logística reversa e incineração, aliadas a efetiva conscientização ambiental da população, buscando mudanças de hábitos sustentávei
Proteomic profile of culture filtrate from the Brazilian vaccine strain Mycobacterium bovis BCG Moreau compared to M. bovis BCG Pasteur
<p>Abstract</p> <p>Background</p> <p>Bacille Calmette-Guerin (BCG) is currently the only available vaccine against tuberculosis (TB) and comprises a heterogeneous family of sub-strains with genotypic and phenotypic differences. The World Health Organization (WHO) affirms that the characterization of BCG sub-strains, both on genomic and proteomic levels, is crucial for a better comprehension of the vaccine. In addition, these studies can contribute in the development of a more efficient vaccine against TB. Here, we combine two-dimensional electrophoresis (2DE) and mass spectrometry to analyse the proteomic profile of culture filtrate proteins (CFPs) from <it>M. bovis </it>BCG Moreau, the Brazilian vaccine strain, comparing it to that of BCG Pasteur. CFPs are considered of great importance given their dominant immunogenicity and role in pathogenesis, being available for interaction with host cells since early infection.</p> <p>Results</p> <p>The 2DE proteomic map of <it>M. bovis </it>BCG Moreau CFPs in the pH range 3 - 8 allowed the identification of 158 spots corresponding to 101 different proteins, identified by MS/MS. Comparison to BCG Pasteur highlights the great similarity between these BCG strains. However, quantitative analysis shows a higher expression of immunogenic proteins such as Rv1860 (BCG1896, Apa), Rv1926c (BCG1965c, Mpb63) and Rv1886c (BCG1923c, Ag85B) in BCG Moreau when compared to BCG Pasteur, while some heat shock proteins, such as Rv0440 (BCG0479, GroEL2) and Rv0350 (BCG0389, DnaK), show the opposite pattern.</p> <p>Conclusions</p> <p>Here we report the detailed 2DE profile of CFPs from <it>M. bovis </it>BCG Moreau and its comparison to BCG Pasteur, identifying differences that may provide relevant information on vaccine efficacy. These findings contribute to the detailed characterization of the Brazilian vaccine strain against TB, revealing aspects that may lead to a better understanding of the factors leading to BCG's variable protective efficacy against TB.</p
A magnanimidade da teoria: interpretar a ética em teoria da literatura
Tese de doutoramento, Teoria da Literatura, Universidade de Lisboa, Faculdade de Letras, 2003Um dos pressupostos desta tese consiste na ideia segundo a qual o pensamento ético só adquire propriedade conceptual se preliminarmente pudermos definir ‘acção’. Como se procura demonstrar no capítulo I, saber o que é uma acção não se constitui, no entanto, como tarefa que descobre e estabelece propriedades intrínsecas e estáveis, mas como descrição de certas ocorrências, nos termos das pessoas que procuram explicar e descrever acções. Assim, o pensamento ético é algo que se constitui a partir da coerência e racionalidade de certas descrições de acções particulares e não um sistema de regras ou prescrições. Defende-se que a coerência estrutural destas descrições determina não apenas a consideração moral das acções, mas também a possibilidade do conhecimento entre as pessoas e a certeza, mais poética do que epistemológica, da sua existência.
Se uma certa estabilidade do texto ético e uma certeza suficiente acerca do conhecimento humano dependem da configuração mais ou menos padronizada das acções humanas e do funcionamento mental, assumem especial relevância cognitiva e ética os casos e fenómenos de irracionalidade na realização de acções e formação de crenças. De facto, como se sugere no capítulo II, estes casos suscitam perplexidades éticas e cognitivas e assinalam o carácter provisório e conceptualmente circunscrito das nossas descrições sobre acções e pessoas, no contexto restrito do pensamento filosófico e ético.
Importa por isso considerar outros textos em que a acção se constitui como motivo de estrutura argumentativa e objecto de análise. A consideração da tragédia e, sobretudo, a análise da teorização poética de Aristóteles acerca do texto trágico formam o capítulo III desta tese. A determinação ética da argumentação técnica da Poética é tão relevante para o entendimento da tragédia quanto, para a ética, conduta moral e compreensão da irracionalidade, é de absoluta pertinência uma particular descrição da acção trágica e da peculiaridade de certas actividades interpretativas exigidas não só por esse entendimento trágico de acção, mas por qualquer texto.ABSTRACT - One of the central ideas of this thesis is that ethical thinking acquires conceptual accuracy only when we can define ‘action’. As it is suggested in chapter I, to know what an action is is not, nevertheless, an activity which finds and establishes intrinsic and stable properties, but a description of some phenomena, in the vocabulary of those who seek to explain and to describe actions. Thus, ethical thinking is derived from the coherence and rationality of descriptions of particular actions and is not, therefore, a system of rules and prescriptions. It is argued that the structural coherence of these descriptions determines not only the moral apprehension of actions, but also the possibility of knowledge of persons and the assurance, more poetical than epistemological, of their existence.
If a certain stability of the ethical text and a sufficient certainty about human knowledge depend on a more or less standard configuration of human actions and of mental functioning, then irrational episodes and phenomena in action and belief assume a particular cognitive and ethical relevance. As proposed in chapter II, these cases excite some ethical and cognitive perplexities and show the provisional and conceptually circumscribed character of our descriptions of actions and persons, in the strict realm of philosophy and ethics.
It is necessary, therefore, to consider other texts in which action is constituted as the cause for the argumentative structure and object of analysis. The consideration of tragedy and, mostly, of the treatment of the tragic text in Aristotle’s Poetics constitutes an important part of this task and is sketched in chapter III. The ethical influence of the technical arguments of the Poetics is therefore relevant to the understanding of tragedy. A particular description of tragic action and of the specificity of interpretative activities is required not only by the understanding of action in tragedy as by the description of ethical patterns of moral behaviour and the comprehension of irrationality in any text.Fundação Calouste Gulbenkian: Bolsa de Curta Duraçã
MicroRNA Hsa-miR-134 is a circulating biomarker for mesial temporal lobe epilepsy

Recently, circulating microRNAs have emerged as potential biomarkers in a number of clinical scenarios. The purpose of this study was to identify and to validate circulating microRNAs that could be used as biomarkers in the diagnosis of epilepsy. Quantitative real-time PCR was used to measure plasma levels of three candidate microRNAs in two phases of study: an initial discovery phase with 14 patients with mesial temporal lobe epilepsy (MTLE), 13 with focal cortical dysplasia (FCD) and 16 controls; and a validation cohort constituted of an independent cohort of 65 patients with MTLE and 83 controls. We found hsa-miR-134 downregulated in patients with MTLE (p = 0.018) but not in patients with FCD, when compared to controls. Furthermore, hsa-miR-134 expression could be used to discriminate MTLE patients with an area under the curve (AUC) of 0.75. To further assess the robustness of hsa-miR-134 as a biomarker for MTLE, we studied an independent cohort of 65 patients with MTLE, 27 of whom MTLE patients were responsive to pharmacotherapy, and 38 patients were pharmacoresistant and 83 controls. We confirmed that hsa-miR-134 was significantly downregulated in the plasma of patients with MTLE when compared with controls (p < 0.001). In addition, hsa-miR-134 identified patients with MTLE regardless of their response to pharmacotherapy or the presence of MRI signs of hippocampal sclerosis. Career self: a longitudinal study with college students
O self de carreira constituí um subconjunto organizado do universo cognitivo de uma pessoa, responsável pelo carácter subjetivo que a mesma confere à carreira. Este estudo pretende avaliar mudanças no conteúdo do self de carreira
de estudantes universitários, do início para o final do último ano de graduação. Para tal, recorreu-se a medidas repetidas dos
índices da Grelha de Repertório da Carreira (Silva & Taveira, 2005; Silva, 2008). Na investigação, participaram 80 estudantes, dos quais 49 são mulheres (61,25%) e 31 são homens (38,75%), com idades entre os 21 e os 45 anos (M= 23,9, DP= 4,31).
Os resultados indicam que, no final da licenciatura, os estudantes diminuem a distância como se constrõem em relação aos outros e mantêm uma construção positiva do self de carreira.Fundação para a Ciência e a Tecnologia (FCT
Synthesis, spectroscopic characterization, photochemical and photophysical properties and biological activities of ruthenium complexes with mono- and bi-dentate histamine ligand
The monodentate cis-[Ru(phen)(2)(hist)(2)](2+) 1R and the bidentate cis-[Ru(phen)(2)(hist)](2+) 2A complexes were prepared and characterized using spectroscopic (H-1, (H-1-H-1) COSY and (H-1-C-13) HSQC NMR, UV-vis, luminescence) techniques. The complexes presented absorption and emission in the visible region, as well as a tri-exponential emission decay. The complexes are soluble in aqueous and non-aqueous solution with solubility in a buffer solution of pH 7.4 of 1.14 x 10(-3) mol L-1 for (1R + 2A) and 6.43 x 10(-4) mol L-1 for 2A and lipophilicity measured in an aqueous-octanol solution of -1.14 and -0.96, respectively. Photolysis in the visible region in CH3CN converted the starting complexes into cis-[Ru(phen)(2)(CH3CN)(2)](2+). Histamine photorelease was also observed in pure water and in the presence of BSA (1.0 x 10(-6) mol L-1). The bidentate coordination of the histamine to the ruthenium center in relation to the monodentate coordination increased the photosubstitution quantum yield by a factor of 3. Pharmacological studies showed that the complexes present a moderate inhibition of AChE with an IC50 of 21 mu mol L-1 (referred to risvagtini, IC50 181 mu mol L-1 and galantamine IC50 0.006 mu mol L-1) with no appreciable cytotoxicity toward to the HeLa cells (50% cell viability at 925 mu mol L-1). Cell uptake of the complexes into HeLa cells was detected by fluorescence confocal microscopy. Overall, the observation of a luminescent complex that penetrates the cell wall and has low cytotoxicity, but is reactive photochemically, releasing histamine when irradiated with visible light, are interesting features for application of these complexes as phototherapeutic agents.FAPESP [Proc 2009/08218-0]FAPESPCNPq [Universal 470890/2010-0]CNPqCAPESCAPE
