The structural organization and protein composition of lens fiber junctions.
The structural organization and protein composition of lens fiber junctions isolated from adult bovine and calf lenses were studied using combined electron microscopy, immunolocalization with monoclonal and polyclonal anti-MIP and anti-MP70 (two putative gap junction-forming proteins), and freeze-fracture and label-fracture methods. The major intrinsic protein of lens plasma membranes (MIP) was localized in single membranes and in an extensive network of junctions having flat and undulating surface topologies. In wavy junctions, polyclonal and monoclonal anti-MIPs labeled only the cytoplasmic surface of the convex membrane of the junction. Label-fracture experiments demonstrated that the convex membrane contained MIP arranged in tetragonal arrays 6-7 nm in unit cell dimension. The apposing concave membrane of the junction displayed fracture faces without intramembrane particles or pits. Therefore, wavy junctions are asymmetric structures composed of MIP crystals abutted against particle-free membranes. In thin junctions, anti-MIP labeled the cytoplasmic surfaces of both apposing membranes with varying degrees of asymmetry. In thin junctions, MIP was found organized in both small clusters and single membranes. These small clusters also abut against particle-free apposing membranes, probably in a staggered or checkerboard pattern. Thus, the structure of thin and wavy junctions differed only in the extent of crystallization of MIP, a property that can explain why this protein can produce two different antibody-labeling patterns. A conclusion of this study is that wavy and thin junctions do not contain coaxially aligned channels, and, in these junctions, MIP is unlikely to form gap junction-like channels. We suggest MIP may behave as an intercellular adhesion protein which can also act as a volume-regulating channel to collapse the lens extracellular space. Junctions constructed of MP70 have a wider overall thickness (18-20 nm) and are abundant in the cortical regions of the lens. A monoclonal antibody raised against this protein labeled these thicker junctions on the cytoplasmic surfaces of both apposing membranes. Thick junctions also contained isolated clusters of MIP inside the plaques of MP70. The role of thick junctions in lens physiology remains to be determined
Duration of shedding of respiratory syncytial virus in a community study of Kenyan children
Background: Our understanding of the transmission dynamics of respiratory syncytial virus (RSV) infection will be better informed with improved data on the patterns of shedding in cases not limited only to hospital admissions.
Methods: In a household study, children testing RSV positive by direct immunofluorescent antibody test (DFA) were enrolled. Nasal washings were scheduled right away, then every three days until day 14, every 7 days until day 28 and every 2 weeks until a maximum of 16 weeks, or until the first DFA negative RSV specimen. The relationship between host factors, illness severity and viral shedding was investigated using Cox regression methods.
Results: From 151 families a total of 193 children were enrolled with a median age of 21 months (range 1-164 months), 10% infants and 46% male. The rate of recovery from infection was 0.22/person/day (95% CI 0.19-0.25) equivalent to a mean duration of shedding of 4.5 days (95%CI 4.0-5.3), with a median duration of shedding of 4 days (IQR 2-6, range 1-14). Children with a history of RSV infection had a 40% increased rate of recovery i.e. shorter duration of viral shedding (hazard ratio 1.4, 95% CI 1.01-1.86). The rate of cessation of shedding did not differ significantly between males and females, by severity of infection or by age.
Conclusion: We provide evidence of a relationship between the duration of shedding and history of infection, which may have a bearing on the relative role of primary versus re-infections in RSV transmission in the community
Assessment of body composition and association with clinical outcomes in patients with lung and colorectal cancer
OBJECTIVES:
To assess body composition in patients with non-small cell lung cancer (NSCLC) and colorectal cancer using whole-body MRI and relate this to clinical outcomes.
METHODS:
53 patients with NSCLC (28 males, 25 females; mean age 66.9) and 74 patients with colorectal cancer (42 males, 32 females; mean age 62.9) underwent staging whole-body MRI scans, which were post-processed to derive fat mass (FM), fat free mass (FFM) and skeletal muscle (SM) indices and SM fat fraction (FF). These were compared between the two cancer cohorts using two-sided t-tests and the chi-squared test. Measurements of body composition were correlated with outcomes including length of hospital stay, metastatic status and mortality.
RESULTS:
Patients with NSCLC had significantly lower FFM (p = 0.0071) and SM (p = 0.0084) indices. Mean SM FF was greater in patients with NSCLC (p = 0.0124) and was associated with longer hospital stay (p = 0.035). There was no significant relationship between FM, FFM and SM indices and length of hospital stay, metastatic status or mortality.
CONCLUSIONS:
Patients with NSCLC had lower FFM and SM indices than patients with colorectal cancer and greater SMFF, indicating lower SM mass with fatty infiltration. These findings reflect differences in the phenotype of the two groups and suggest patients with lung cancer are more likely to require additional nutritional support.

A matter of words: NLP for quality evaluation of Wikipedia medical articles
Automatic quality evaluation of Web information is a task with many fields of
applications and of great relevance, especially in critical domains like the
medical one. We move from the intuition that the quality of content of medical
Web documents is affected by features related with the specific domain. First,
the usage of a specific vocabulary (Domain Informativeness); then, the adoption
of specific codes (like those used in the infoboxes of Wikipedia articles) and
the type of document (e.g., historical and technical ones). In this paper, we
propose to leverage specific domain features to improve the results of the
evaluation of Wikipedia medical articles. In particular, we evaluate the
articles adopting an "actionable" model, whose features are related to the
content of the articles, so that the model can also directly suggest strategies
for improving a given article quality. We rely on Natural Language Processing
(NLP) and dictionaries-based techniques in order to extract the bio-medical
concepts in a text. We prove the effectiveness of our approach by classifying
the medical articles of the Wikipedia Medicine Portal, which have been
previously manually labeled by the Wiki Project team. The results of our
experiments confirm that, by considering domain-oriented features, it is
possible to obtain sensible improvements with respect to existing solutions,
mainly for those articles that other approaches have less correctly classified.
Other than being interesting by their own, the results call for further
research in the area of domain specific features suitable for Web data quality
assessment
»I am a virgin woman and a virgin woman’s child« Critical Plant Theory and the Maiden Mother Conceit in Early Medieval Riddles
While early medieval riddles in Old English and, to a lesser extent, Latin, have been studied extensively from ecocritical perspectives in recent years, the large corpora of riddles in other languages of western Eurasia have yet to benefit from or feed back into these methodological developments. Meanwhile, ecocritical research generally has focused on animals at the expense of plants. We respond to both problems by providing the first extensive study of riddles whose solutions are plants, through the lens of one recurrent conceit in ancient and medieval verse riddles in Arabic, Greek, Latin, Old Norse and, we argue, Hebrew. The conceit is that a plant is a virgin woman who nevertheless reproduces. By examining different permutations of this motif, we show how these riddles use plants to comment on human gendering, and how, while usually fundamentally patriarchal in their world-views, they register patriarchal anxiety at women’s reproductive capabilities, acknowledge critiques of patriarchal constraints on women, and queer gender norms in other ways; inter alia we note that the Old Norse riddle studied here may be the only explicit (albeit metaphorical) representation of female homosexual eroticism in the Old Norse corpus. However, we also draw on critical plant theory to explore how the riddles situate plants in medieval Abrahamicate cultures, uncovering implicit recognitions of the dynamic and reciprocal relationships between human farmers and their family structures, the plants that domesticate them, people’s and plants’ mutual shaping of the ecosystems they inhabit or colonise, and the economies that these interactions constitute
The logic of comparative life history studies for estimating key parameters, with a focus on natural mortality rate
There are a number of key parameters in population dynamics that are difficult to estimate, such as natural mortality rate, intrinsic rate of population growth, and stock-recruitment relationships. Often, these parameters of a stock are, or can be, estimated indirectly on the basis of comparative life history studies. That is, the relationship between a difficult to estimate parameter and life history correlates is examined over a wide variety of species in order to develop predictive equations. The form of these equations may be derived from life history theory or simply be suggested by exploratory data analysis. Similarly, population characteristics such as potential yield can be estimated by making use of a relationship between the population parameter and bio-chemico-physical characteristics of the ecosystem. Surprisingly, little work has been done to evaluate how well these indirect estimators work and, in fact, there is little guidance on how to conduct comparative life history studies and how to evaluate them. We consider five issues arising in such studies: (i) the parameters of interest may be ill-defined idealizations of the real world, (ii) true values of the parameters are not known for any species, (iii) selecting data based on the quality of the estimates can introduce a host of problems, (iv) the estimates that are available for comparison constitute a non-random sample of species from an ill-defined population of species of interest, and (v) the hierarchical nature of the data (e.g. stocks within species within genera within families, etc., with multiple observations at each level) warrants consideration. We discuss how these issues can be handled and how they shape the kinds of questions that can be asked of a database of life history studies
Perceived impacts and residents\u27 support for tourism development in Port Dickson, Malaysia
This study evaluates the image that residents perceive of their location and its influences on their understanding of tourism impacts, and their support for the development of the tourism. The data was collected from 422 residents of Port Dickson in Malaysia and were examined by applying PLS-SEM. Results displayed a positive image of place will lead to positive perceptions of tourism development impacts leadings to residents\u27 support for tourism development. Practical implications of these outcomes are also discussed relative to tourism planning and development
4D analysis of the microstructural evolution of Si-based electrodes during lithiation: Time-lapse X-ray imaging and digital volume correlation
Silicon is a promising candidate to substitute or complement graphite as anode material in Li-ion batteries due, mainly, to its high energy density. However, the lithiation/delithiation processes of silicon particles are inherently related to drastic volume changes which, within a battery's physically constrained case, can induce significant deformation of the fundamental components of the battery that can eventually cause it to fail.
In this work, we use non-destructive time-lapse X-ray imaging techniques to study the coupled electrochemo-mechanical phenomena in Li-ion batteries. We present X-ray computed tomography data acquired at different times during the first lithiation of custom-built silicon-lithium battery cells. Microstructural volume changes have been quantified using full 3D strain field measurements from digital volume correlation analysis. Furthermore, the extent of lithiation of silicon particles has been quantified in 3D from the grey-scale of the tomography images. Correlation of the volume expansion and grey-scale changes over the silicon-based electrode volume indicates that the process of lithiation is kinetically affected by the reaction at the Si/LixSi interface
