    Evaluation of cognition enhancing activity of pioglitazone in albino mice

    Background: Worldwide, 47.5 million people have dementia, with just over half (58%) living in low- and middle-income countries. Every year, there are 7.7 million new cases. The estimated proportion of the general population aged 60 and over with dementia at a given time is between 5 to 8 per 100 people. The total number of people with dementia is projected to be 75.6 million in 2030 and almost triple by 2050 to 135.5 million.Methods: All animals were allowed to acclimatize with laboratory conditions at least two weeks before starting the experiment and they were maintained under the same condition throughout the experiment. They were given food and water ad libitum. The experiments were performed as per the Committee for the Purpose of Control and Supervision on Experiments on Animals (CPCSEA) guidelines. The animals were subjected to experimentation between 0900-1600 hours in noise free atmosphere with ambient temperature 23-300ºC.Results: The Pioglitazone treated groups showed statistically significant results in the SDL when compared to the control group (p<0.01 on both days) and the scopolamine group (p<0.01 on both days) on both acquisition day and retention day.Conclusions: There was no significant difference in SDL in standard and Pioglitazone treated groups on both acquisition day and retention day

    Background: Pioglitazone (PIO), a Peroxisome Proliferator Activated Receptor γ (PPAR-γ) agonist, is an oral anti-diabetic agent belonging to the group of thiazolidinediones-TZDs used for the treatment of diabetes mellitus type 2 in monotherapy and in combination with a sulfonylurea, metformin, or insulin.Methods: All animals were allowed to acclimatize with laboratory conditions at least two weeks before starting the experiment and they were maintained under the same condition throughout the experiment.     A linked data representation for summary statistics and grouping criteria

    Summary statistics are fundamental to data science, and are the buidling blocks of statistical reasoning. Most of the data and statistics made available on government web sites are aggregate, however, until now, we have not had a suitable linked data representation available. We propose a way to express summary statistics across aggregate groups as linked data using Web Ontology Language (OWL) Class based sets, where members of the set contribute to the overall aggregate value. Additionally, many clinical studies in the biomedical field rely on demographic summaries of their study cohorts and the patients assigned to each arm. While most data query languages, including SPARQL, allow for computation of summary statistics, they do not provide a way to integrate those values back into the RDF graphs they were computed from. We represent this knowledge, that would otherwise be lost, through the use of OWL 2 punning semantics, the expression of aggregate grouping criteria as OWL classes with variables, and constructs from the Semanticscience Integrated Ontology (SIO), and the World Wide Web Consortium’s provenance ontology, PROV-O, providing interoperable representations that are well supported across the web of Linked Data. We evaluate these semantics using a Resource Description Framework (RDF) representation of patient case information from the Genomic Data Commons, a data portal from the National Cancer Institute

    Genetic Divergence for Yield, Physiological and Quality Traits in Super-Early Pigeon pea (Cajanus cajan. (l.) Millsp.)

    The present investigation aimed to study genetic divergence and clustering pattern of 37super-early pigeon pea genotypes. Analysis of variance and hierarchical cluster analysis of tocher’s method revealed significant differences among the genotypes for all the traits under study. Based on genetic distance (D2 value), the 37 genotypes were grouped into 9 distinctive clusters, of which cluster I and II formed the largest clusters with 10 genotypes in each. Among all the characters understudy, leaf area index(LAI) at 60 DAS contributed more to the divergence followed by leaf area (17.02) and leaf area index (12.71) at maturity. Based on the average inter-cluster distance, the cluster III and IX (66.93) tailed by cluster III and VIII (64.86) and cluster VI and VIII (64.06) showed higher inter-cluster distance depicting the wider divergence. Trait-wise selection of diverse parents from the above clusters aids in exploitation of heterosis in superearly pigeon pea

    Genetic Variability for Yield, Physiological and Quality Traits in Novel Super-Early Pigeonpea (Cajanus cajan (L.) Millsp.)

    Super-early pigeonpea are novel genotypes that are reported to be photoperiod insensitive making it possible to grow it in non-traditional regions. Estimation of genetic parameters would be useful in developing appropriate selection and breeding strategies. A study was conducted to evaluate 37 super-early pigeonpea genotypes to access the magnitude of variability and to study heritable component of variation present in the yield, physiological and quality traits. The results revealed that traits leaf area duration between 60 DAS & maturity followed by leaf area & leaf area index at maturity, net assimilation between 60 DAS & maturity, leaf area index & leaf area at 60 DAS, leaf area duration between 60 DAS & maturity and plant height had high had higher PCV and GCV values. In general, phenotypic coefficients of variation (PCV) estimates were higher than genotypic coefficients of variation (GCV) estimates for all the characters under study, but the difference was relatively small indicating that these characters were less influenced by the environment and selection to improve those traits might be effective. High heritability combined with high genetic advance as a percent of mean was noted for all the traits except protein content conveying the governance of additive gene on trait expression. Anticipating these traits as selection index reaps competent improvement in yield, physiological and quality traits in early maturing pigeonpea

    Multi-environment testing for G×E interactions and identification of high-yielding, stable, medium-duration pigeonpea genotypes employing AMMI, GGE biplot, and YREM analyses

    Pigeonpea [Cajanus cajan (L.) Millspaugh] is a widely grown pulse with high seed protein content that contributes to food and nutritional security in the Indian subcontinent. The majority of pigeonpea varieties cultivated in India are of medium duration (<180 days to maturity), which makes it essential for breeders to focus on the development of stable high-yielding varieties. The diverse agroecological regime in the Indian subcontinent necessitates an efficient multi-environment study by taking into consideration genotype (G) × environment (E) interaction (GEI) that has a significant impact on traits like grain yield (GY) in developing high-yielding and widely adaptable varieties. In the present study, 37 pigeonpea genotypes were evaluated during the 2021 rainy season at ARS Badnapur, ARS Tandur, BAU Ranchi, GKVK Bengaluru, and ICRISAT Patancheru. The GEI was significant on the grain yield (p < 0.01), and hence, genotype + genotype × environment (GGE) and additive main effects and multiplicative interaction (AMMI) biplots along with AMMI stability value (ASV) and yield relative to environmental maximum (YREM) statistics were used to identify stable high-yielding genotypes. The interaction principal component analysis 1 and 2 (IPC1 and IPC2) explained 40.6% and 23.3% variations, respectively. Based on the rankings of genotypes, G37 (ICPL 20205), G35 (ICPL 20203), G8 (ICPL 19404), G17 (ICPL 19415), and G9 (ICPL 19405) were identified as ideal genotypes. Discriminativeness vs. representativeness identified GKVK Bengaluru as an ideal environment for comprehensive evaluation of test genotypes. However, ICPL 19405 was identified as the potentially stable high-yielding genotype for further testing and release across the test environments based on its mean grain yield (1,469.30 kg/ha), least ASV (3.82), and low yield stability index (YSI) of 13

    Recognition of microbial viability via TLR8 drives TFH cell differentiation and vaccine responses

    Live attenuated vaccines are generally highly efficacious and often superior to inactivated vaccines, yet the underlying mechanisms of this remain largely unclear. Here we identify recognition of microbial viability as a potent stimulus for follicular helper T cell (TFH cell) differentiation and vaccine responses. Antigen-presenting cells (APCs) distinguished viable bacteria from dead bacteria through Toll-like receptor 8 (TLR8)-dependent detection of bacterial RNA. In contrast to dead bacteria and other TLR ligands, live bacteria, bacterial RNA and synthetic TLR8 agonists induced a specific cytokine profile in human and porcine APCs, thereby promoting TFH cell differentiation. In domestic pigs, immunization with a live bacterial vaccine induced robust TFH cell and antibody responses, but immunization with its heat-killed counterpart did not. Finally, a hypermorphic TLR8 polymorphism was associated with protective immunity elicited by vaccination with bacillus Calmette-Guérin (BCG) in a human cohort. We have thus identified TLR8 as an important driver of TFH cell differentiation and a promising target for TFH cell–skewing vaccine adjuvants
