    Is the plant Bolboschoenus maritimus an adequate biomonitor for trace metal contamination in saltmarshes? A field study from the Óbidos lagoon (Portugal)

    Monitoring the negative impacts of trace metals is crucial to assess the health and stability of ecosystems. In salt marshes, halophyte plants were reported as possible bioaccumulators of these elements. The aim of this work was to explore the bioaccumulation potential of Bolboschoenus maritimus as a tool for monitoring the presence of metals in coastal environments. Bolboschoenus maritimus were collected from a brackish water lagoon, and the presence of the trace metals lead, cadmium, and nickel were seasonally evaluated in distinct parts of the plants, and in water and sediment samples. Lead was the trace metal with the highest concentration detected in water and sediments of the sampling site. The highest lead concentrations in B. maritimus were recorded in the spring season. The transport index indicated an accumulation of lead in the leaves of around 70% in the spring of 2009. Cadmium in leaves in spring and summer of 2009 reached values above 5 mg Cd. kg−1. Nickel was not detected in most samples collected. Bolboschoenus maritimus was considered an adequate biomonitor for lead and cadmium, since it bioaccumulates both metals with seasonally distinct results, as the bioaccumulation factor results indicated.info:eu-repo/semantics/publishedVersio

    Evaluation of different strains of Saccharomyces cerevisiae for ethanol production from high-amylopectin BRS AG rice (Oryza sativa L.)

    Ethanol is the main biofuel produced by fermentation route and the search for new feedstocks to produce fuel ethanol is still a great challenge. This work aims to compare the ethanol production from a new irrigated rice cultivar BRS AG to the conventional cultivar BRS PAMPA applied in Brazil. Six different commercial strains of Saccharomyces cerevisiae (BG-1, CAT-1, FT-858, JP-1, PE-2, and SA-1) were applied in fermentation reactions. Fermentations performed with BRS PAMPA rice revealed that the highest yields were achieved with strain SA-1, corresponding to 93.0% of the theoretical maximum and final ethanol concentration of 58.92 g L−1, and with CAT-1, a yield of 92.7% and final ethanol concentration of 58.93 g L−1. For the fermentations with BRS AG rice, the highest yields were obtained with strain FT-858, exhibiting a 89.6% yield and final ethanol concentration of 62.45 g L−1, and with CAT-1, 87.9% yield and final ethanol concentration of 61.25 g L−1 were achieved. The most appropriate microorganism for ethanol production using BRS PAMPA rice and BRS AG rice was CAT-1. Comparatively, the ethanol yield and productivity using BRS AG were higher than those observed for BRS PAMPA for all strains, except for PE-2 and SA-1 that led to very similar results. The experimental results showed that the giant rice BRS AG is an excellent feedstock for fuel ethanol production in lowland fields

    Bioconversion of glycerol into lactic acid by a new bacterial strain from the Brazilian cerrado soil

    A lactic-acid-producing strain was isolated from the Brazilian Cerrado soil (Brazilian savanna). Glycerol, a byproduct of the biodiesel industry, can be converted into various chemical intermediates of industrial value by biotechnological routes. Klebsiella pneumoniae can metabolize glycerol in environments with or without oxygen and bioconvert it into several chemicals with high value-added, such as lactic acid, 3-hydroxypropionic acid and 1,3 propanediol. The wild-type bacterial strain (2GPP) isolated from a soil sample from the Brazilian Cerrado was determined to be a K. pneumoniae complex that was capable of successfully metabolizing glycerol. Fermentations were performed with different temperatures, pH, and inoculum concentrations to evaluate the best lactic acid production. At first, 1,3-propanediol and L-(+)-lactic acid were produced in mini reactors. A lactic acid production of 3.8 g·L −1 and a decrease in 1,3-propanediol output were observed. Thus, by adjusting process variables such as pH and temperature during fermentation, it was possible to maximize the production of lactic acid and decrease the formation of 1,3-propanediol by utilizing experimental design strategies

    This paper reports the creation of the Laboratory of Popular Education and Health at the Psychology Department of University of Santa Catarina, Brazil. Ten years ago some teachers of this Department started to develop isolated actions in order to extend psychological services to lower class people, traditionally excluded from this kind of professional care. In the course of this work the inefficacy of theoretical and methodological framework of traditional Clinical Psychology became evident. They summed up their experiences in the Laboratory, where two urgent tasks were in need: the search for new psychogical theories and practices and the reexamination of graduate courses of Psychology, in order to cope with the Brazilian social problems.O presente artigo relata a constituição do Laboratório de Educação e Saúde Popular do Departamento de Psicologia da Universidade Federal de Santa Catarina. Há mais de dez anos, alguns docentes do referido departamento começaram, de forma isolada, a desenvolver atividades voltadas a estender o atendimento psicológico a camadas de baixa renda, camadas estas historicamente marginalizadas deste tipo de serviço. O contato com esta clientela evidenciou a ineficácia do referencial teórico-metodológico da Psicologia Clínica tradicional, apontando a necessidade de repensar e questionar os conceitos e práticas existentes. Impôs-se a urgência de construir novas práticas e modalidades de atendimento, bem como de redirecionar a atividade acadêmica e a formação dos futuros profissionais de Psicologia, de forma a instrumentá-los para o trabalho com as realidades estudadas e, por conseqüência, com os graves problemas que afligem a sociedade brasileira

    Expression of histone methyltransferases as novel biomarkers for renal cell tumor diagnosis and prognostication

    Renal cell tumors (RCTs) are the most lethal of the common urological cancers. The widespread use of imaging entailed an increased detection of small renal masses, emphasizing the need for accurate distinction between benign and malignant RCTs, which is critical for adequate therapeutic management. Histone methylation has been implicated in renal tumorigenesis, but its potential clinical value as RCT biomarker remains mostly unexplored. Hence, the main goal of this study was to identify differentially expressed histone methyltransferases (HMTs) and histone demethylases (HDMs) that might prove useful for RCT diagnosis and prognostication, emphasizing the discrimination between oncocytoma (a benign tumor) and renal cell carcinoma (RCC), especially the chromophobe subtype (chRCC). We found that the expression levels of three genes-SMYD2, SETD3, and NO66-was significantly altered in a set of RCTs, which was further validated in a large independent cohort. Higher expression levels were found in RCTs compared to normal renal tissues (RNTs) and in chRCCs comparatively to oncocytomas. SMYD2 and SETD3 mRNA levels correlated with protein expression assessed by immunohistochemistry. SMYD2 transcript levels discriminated RCTs from RNT, with 82.1% sensitivity and 100% specificity (AUC=0.959), and distinguished chRCCs from oncocytomas, with 71.0% sensitivity and 73.3% specificity (AUC: 0.784). Low expression levels of SMYD2, SETD3, and NO66 were significantly associated with shorter disease-specific and disease-free survival, especially in patients with non-organ confined tumors.     SETDB2 and RIOX2 are differentially expressed among renal cell tumor subtypes, associating with prognosis and metastization

    Increasing detection of small renal masses by imaging techniques entails the need for accurate discrimination between benign and malignant renal cell tumors (RCTs) as well as among malignant RCTs, owing to differential risk of progression through metastization. Although histone methylation has been implicated in renal tumorigenesis, its potential as biomarker for renal cell carcinoma (RCC) progression remains largely unexplored. Thus, we aimed to characterize the differential expression of histone methyltransferases (HMTs) and histone demethylases (HDMs) in RCTs to assess their potential as metastasis biomarkers. We found that SETDB2 and RIOX2 (encoding for an HMT and an HDM, respectively) expression levels was significantly altered in RCTs; these genes were further selected for validation by quantitative RT-PCR in 160 RCTs. Moreover, SETDB2, RIOX2, and three genes encoding for enzymes involved in histone methylation (NO66, SETD3, and SMYD2), previously reported by our group, were quantified (RT-PCR) in an independent series of 62 clear cell renal cell carcinoma (ccRCC) to assess its potential role in ccRCC metastasis development. Additional validation was performed using TCGA dataset. SETDB2 and RIOX2 transcripts were overexpressed in RCTs compared to renal normal tissues (RNTs) and in oncocytomas vs. RCCs, with ccRCC and papillary renal cell carcinoma (pRCC) displaying the lowest levels. Low SETDB2 expression levels and higher stage independently predicted shorter disease-free survival. In our 62 ccRCC cohort, significantly higher RIOX2, but not SETDB2, expression levels were depicted in cases that developed metastasis during follow-up. These findings were not apparent in TCGA dataset. We concluded that SETDB2 and RIOX2 might be involved in renal tumorigenesis and RCC progression, especially in metastatic spread. Moreover, SETDB2 expression levels might independently discriminate among RCC subgroups with distinct outcome, whereas higher RIOX2 transcript levels might identify ccRCC cases with more propensity to endure metastatic dissemination.This study was funded by research grants from Research Center of Portuguese Oncology Institute - Porto (CI-IPOP 4-2012 and CI-IPOP 27) and from Associacao Portuguesa de Urologia (APU-2010).     Conservation Biogeography of the Sahara‐Sahel: additional protected areas are needed to secure unique biodiversity

    Aim Identification of priority conservation areas and evaluation of coverage of the current protected areas are urgently needed to halt the biodiversity loss. Identifying regions combining similar environmental traits (climate regions) and species assemblages (biogroups) is needed for conserving the biodiversity patterns and processes. We identify climate regions and biogroups and map species diversity across the Sahara-Sahel, a large geographical area that exhibits wide environmental heterogeneity and multiple species groups with distinct biogeographical affinities, and evaluate the coverage level of current network of protected areas for biodiversity conservation. Location Sahara-Sahel, Africa. Methods We use spatially explicit climate data with the principal component analysis and model-based clustering techniques to identify climate regions. We use distributions of 1147 terrestrial vertebrates (and of 125 Sahara-Sahel endemics) and apply distance clustering methods to identify biogroups for both species groups. We apply reserve selection algorithms targeting 17% of species distribution, climate regions and biogroups to identify priority areas and gap analysis to assess their representation within the current protected areas. Results Seven climate regions were identified, mostly arranged as latitudinal belts. Concentrations of high species richness were found in the Sahel, but the central Sahara gathers most endemic and threatened species. Ten biogroups (five for endemics) were identified. A wide range of biogroups tend to overlap in specific climate regions. Identified priority areas are inadequately represented in protected areas, and six new top conservation areas are needed to achieve conservation targets. Main conclusions Biodiversity distribution in Sahara-Sahel is spatially structured and apparently related to environmental variation. Although the majority of priority conservation areas are located outside the areas of intense human activities, many cross multiple political borders and require internationally coordinated efforts for implementation and management. Optimized biodiversity conservation solutions at regional scale are needed. Our work contradicts the general idea that deserts are uniform areas and provide options for the conservation of endangered species.info:eu-repo/semantics/publishedVersio

    Bagaza virus emerged in Spain in 2010 and was not reported in other countries in Europe until 2021, when the virus was detected by molecular methods in a corn bunting and several red-legged partridges in Portugal.     Differential sensitivities to lactate transport inhibitors of breast cancer cell lines

Monocarboxylate transporters (MCTs) play a role in extracellular acidification, which is widely known to be involved in tumour progression. Recently, we have described the upregulation of MCT1 in breast carcinomas and its association with poor prognostic variables. Thus, we aimed to evaluate the effect of lactate transport inhibition in human breast cancer cell lines. The effects of a-cyano-4-hydroxycinnamate, quercetin and lonidamine on cell viability, metabolism, proliferation, apoptosis, migration and invasion were assessed in a panel of different breast cancer cell lines. MCT1, MCT4 and CD147 were differently expressed among the breast cancer cell lines and, as expected, different sensitivities were observed for the three inhibitors. Interestingly, in the most sensitive cell lines, lactate transport inhibition induced a decrease in cell proliferation, migration and invasion, as well as an increase in cell death. Results were validated by silencing MCT1 expression using siRNA. The results obtained here support targeting of lactate transport as a strategy to treat breast cancer, with a special emphasis on the basal-like subtype, which so far does not have a specific molecular therapy.This work was supported by the Fundação para a Ciência e a Tecnologia (FCT) grant ref.
