For the assessment of the gentoxic potential of mineral fibers physical data alone are not sufficient. As biological endpoints micronuclei and chromosomal aberrations can be readily analyzed qualitatively and quantitatively. In the course of the project such fiber-induced changes were investigated by means of the tandem-labeling-technique which facilitates the analysis of interphase nuclei. Our results show that mineral fibers induce micronuclei also in human cells. Furthermore, additional information was obtained concerning chromosomal aberrations in specific regions. In this way, a more detailed characterization of fiber action at the chromosomal level was achieved. In summary we have shown that primary genotoxic effects of fibers can be readily investigated in detail taking into account the observed specific changes. Thus, the requirements for primary test applications of the developed fiber test system are fulfilled. Validation and further technical improvement studies are under way.
