    Extreme Value Statistics Analysis of Process Defects in Additive Manufacturing Materials

    Fatigue and fracture studies focused on process defects that occur in Additive Manufacturing (AM) materials have shown that defect populations possess features which are better measured with extreme value statistics (EVS). In AM alloys, defect occurrences increase with material volume. This situation facilitates the need to model process defects in the path of fatigue crack growth with suitable statistical tools, such as EVS, which is more cost-effective when compared to destructive experiments. The application of EVS on defect space features helps determine the difference in defects present on fracture surfaces. As the fatigue quality of any material depends on its extreme value flaws, we use the Block Maxima and Peak over Threshold methodologies to study the distribution of the features of the defects in AM Ti-6Al-4V and make recommendations for the distributions of best fit based on different scenarios with different ranges of complexity of different defect types

    Streptococcus Pneumoniae Serotype 19A in Hospitalized Children With invasive Pneumococcal Disease after the introduction of Conjugated Vaccines in Lima, Peru

    BACKGROUND: The Pneumococcal conjugate vaccine (PCV) has decreased cases of invasive pneumococcal disease (IPD) worldwide. However, the impact of PCVs introduction may be affected by the serotype distribution in a specific context. METHODS: Cross-sectional multicenter passive surveillance study of IPD cases in pediatric patients hospitalized in Lima, Peru between 2016 and 2019 (after PCV13 introduction) to determine the serotype distribution and antimicrobial resistance of Streptococcus pneumoniae. Serotyping was performed by a sequential multiplex PCR and confirmed by whole genome sequencing. RESULTS: Eighty-five S. pneumoniae isolates were recovered (4.07/100,000 among childrenage). Serotype 19A was the most common (49.4%). Children infected with serotype 19A in comparison with children infected with other serotypes were younger, had a lower rate of meningitis and higher rates of pneumonia, complicated pneumonia and antimicrobial resistance; 28.6% of patients with serotype 19A have received at least one dose of PCV13 vs. 62.8% of patients with other serotypes. Using MIC-breakpoints, 81.2% (56/69) of non-meningitis strains and 31.2% (5/16) of meningitis strains were susceptible to penicillin; 18.8% (3/16) of meningitis strains had intermediate resistance to ceftriaxone. Resistance to azithromycin was 78.8% (67/85). Serotype 19A frequency increased over time in the same study population, from 4.2% (4/96) in 2006-2008, to 8.6% (5/58) in 2009-2011, to 49.4% (42/85) in the current study (2016-2019) (p \u3c 0.001). CONCLUSIONS: After PCV13 introduction in Peru, serotype 19A remains the most prevalent; however, the vaccination coverage is still not optimal. Therefore, additonal surveillance studies are needed to determine the remaining IPD burden

    Fumarate Hydratase Loss Causes Combined Respiratory Chain Defects.

    Fumarate hydratase (FH) is an enzyme of the tricarboxylic acid (TCA) cycle mutated in hereditary and sporadic cancers. Despite recent advances in understanding its role in tumorigenesis, the effects of FH loss on mitochondrial metabolism are still unclear. Here, we used mouse and human cell lines to assess mitochondrial function of FH-deficient cells. We found that human and mouse FH-deficient cells exhibit decreased respiration, accompanied by a varying degree of dysfunction of respiratory chain (RC) complex I and II. Moreover, we show that fumarate induces succination of key components of the iron-sulfur cluster biogenesis family of proteins, leading to defects in the biogenesis of iron-sulfur clusters that affect complex I function. We also demonstrate that suppression of complex II activity is caused by product inhibition due to fumarate accumulation. Overall, our work provides evidence that the loss of a single TCA cycle enzyme is sufficient to cause combined RC activity dysfunction

    Open loop amplitudes and causality to all orders and powers from the loop-tree duality

    Multiloop scattering amplitudes describing the quantum fluctuations at high-energy scattering processes are the main bottleneck in perturbative quantum field theory. The loop-tree duality is a novel method aimed at overcoming this bottleneck by opening the loop amplitudes into trees and combining them at integrand level with the real-emission matrix elements. In this Letter, we generalize the loop-tree duality to all orders in the perturbative expansion by using the complex Lorentz-covariant prescription of the original one-loop formulation. We introduce a series of mutiloop topologies with arbitrary internal configurations and derive very compact and factorizable expressions of their open-to-trees representation in the loop-tree duality formalism. Furthermore, these expressions are entirely independent at integrand level of the initial assignments of momentum flows in the Feynman representation and remarkably free of noncausal singularities. These properties, that we conjecture to hold to other topologies at all orders, provide integrand representations of scattering amplitudes that exhibit manifest causal singular structures and better numerical stability than in other representations.Comment: Final version to appear in Physical Review Letter

    Nuclear rupture at sites of high curvature compromises retention of DNA repair factors.

    The nucleus is physically linked to the cytoskeleton, adhesions, and extracellular matrix-all of which sustain forces, but their relationships to DNA damage are obscure. We show that nuclear rupture with cytoplasmic mislocalization of multiple DNA repair factors correlates with high nuclear curvature imposed by an external probe or by cell attachment to either aligned collagen fibers or stiff matrix. Mislocalization is greatly enhanced by lamin A depletion, requires hours for nuclear reentry, and correlates with an increase in pan-nucleoplasmic foci of the DNA damage marker ÎłH2AX. Excess DNA damage is rescued in ruptured nuclei by cooverexpression of multiple DNA repair factors as well as by soft matrix or inhibition of actomyosin tension. Increased contractility has the opposite effect, and stiff tumors with low lamin A indeed exhibit increased nuclear curvature, more frequent nuclear rupture, and excess DNA damage. Additional stresses likely play a role, but the data suggest high curvature promotes nuclear rupture, which compromises retention of DNA repair factors and favors sustained damage

    The CERN Neutrino beam to Gran Sasso (NGS)

    The conceptual technical design of the NGS (CERN neutrino beam to Gran Sasso) facility has been presented in the report CERN 98-02 / INFN-AE/98-05. Additional information, in particular an update on various neutrino beam options for the NGS facility, has been provided in a memorandum to the CERN-SPSC Committee (CERN-SPSC/98-35). In the present report, further improvements on the NGS design and performance, in particular new scenarios for SPS proton cycles for NGS operation and a new version of the NGS "high energy" neutrino beam for nt appearance experiments, are described. This new NGS reference beam is estimated to provide three times more nt events per year than the beam presented in the 1998 report. The radiological aspects of the NGS facility have been re-examined with the new beam design. An updated version of the construction schedule is also presented

    Subarachnoidal Neurocysticercosis non-responsive to cysticidal drugs: a case series

    <p>Abstract</p> <p>Background</p> <p>Neurocysticercosis (NC) is one of the most frequent parasitic diseases of the central nervous system. Cysticidal drugs, albendazole and praziquantel, are generally effective when parasites localize in the parenchyma. In contrast, parasites lodged in the subarachnoid basal cisterns are less responsive to treatment.</p> <p>Case Presentation</p> <p>The clinical and radiological pictures of six Mexican patients non-respondent to cysticidal treatment are presented.</p> <p>Conclusions</p> <p>The possible factors involved in the cysticidal non-response are discussed and hints are provided of potentially useful changes to therapeutic protocols.</p

    LSST: from Science Drivers to Reference Design and Anticipated Data Products

    (Abridged) We describe here the most ambitious survey currently planned in the optical, the Large Synoptic Survey Telescope (LSST). A vast array of science will be enabled by a single wide-deep-fast sky survey, and LSST will have unique survey capability in the faint time domain. The LSST design is driven by four main science themes: probing dark energy and dark matter, taking an inventory of the Solar System, exploring the transient optical sky, and mapping the Milky Way. LSST will be a wide-field ground-based system sited at Cerro Pach\'{o}n in northern Chile. The telescope will have an 8.4 m (6.5 m effective) primary mirror, a 9.6 deg2^2 field of view, and a 3.2 Gigapixel camera. The standard observing sequence will consist of pairs of 15-second exposures in a given field, with two such visits in each pointing in a given night. With these repeats, the LSST system is capable of imaging about 10,000 square degrees of sky in a single filter in three nights. The typical 5σ\sigma point-source depth in a single visit in rr will be ∌24.5\sim 24.5 (AB). The project is in the construction phase and will begin regular survey operations by 2022. The survey area will be contained within 30,000 deg2^2 with ÎŽ<+34.5∘\delta<+34.5^\circ, and will be imaged multiple times in six bands, ugrizyugrizy, covering the wavelength range 320--1050 nm. About 90\% of the observing time will be devoted to a deep-wide-fast survey mode which will uniformly observe a 18,000 deg2^2 region about 800 times (summed over all six bands) during the anticipated 10 years of operations, and yield a coadded map to r∌27.5r\sim27.5. The remaining 10\% of the observing time will be allocated to projects such as a Very Deep and Fast time domain survey.     Salmonella enterica serotype Virchow associated with human infections in Switzerland: 2004-2009

    BACKGROUND: Salmonellosis is one of the most important foodborne diseases and a major threat to public health. Salmonella serotype Virchow ranks among the top five serovars in Europe. METHOD: A total of 153 strains isolated from different patients from 2004 through 2009 in Switzerland were further characterized by (i) assessing phenotypic antibiotic resistance profiles using the disk diffusion method and (ii) by genotyping using pulsed-field gel electrophoresis (PFGE) after macrorestriction with XbaI in order to evaluate strain relationship. RESULTS: The relative frequency of S. Virchow among other Salmonella serovars varied between 4th to 8th rank. The annual incidence ranged from 0.45/100'000 in 2004 to 0.40/100'000 in 2009. A total of 48 strains (32%) were resistant to one to 3 antimicrobials, 54 strains (36%) displayed resistance patterns to more than three antibiotics. No trend was identifiable over the years 2004 to 2009. We found a high prevalence (62%) of nalidixic acid resistant strains, suggesting an equally high rate of decreased fluoroqionolone susceptibility, whereas intermediate resistance to ciprofloxacin was negligible. Two strains were extended spectrum ÎČ-lactamase (ESBL) producers. Analysis of PFGE patterns uncovered a predominant cluster (similarity coefficient above 80%) consisting of 104 of the 153 strains. CONCLUSION: The worldwide increase of antibiotic resistances in Salmonella is an emerging public health problem. For Switzerland, no clear trend is identifiable over the years 2004 to 2009 for S. Virchow. Antimicrobial susceptibility and resistance profiles varied considerably within this period. Nevertheless, the situation in Switzerland coincided with findings in other European countries. Genotyping results of this strain collection revealed no evidence for an undetected outbreak within this time period
