    One-year timeline kinetics of cytokine-mediated cellular immunity in dogs vaccinated against visceral leishmaniasis

    BACKGROUND: The main control strategy for visceral leishmaniasis in Brazil has been based on the elimination of seropositive dogs, although this is not widely accepted. In this context, the use of a long-lasting protective vaccine against canine visceral leishmaniasis (CVL) has been highly expected. The aim of this work was to determine the timeline kinetics of the cytokine microenvironment derived from circulating leukocytes as supportive immunological biomarkers triggered by Leishmune® vaccine. Cross-sectional kinetic analysis of cellular immunity cytokines was carried out at three times (1, 6 and 12 months) after primovaccination with Leishmune®. In vitro short-term whole blood cultures were stimulated with Leishmania infantum soluble antigen (SLAg). The secreted cytokine signatures and their major sources were determined. RESULTS: At six months after vaccination, Leishmune® induced an increase in IL-8, IFN-γ, IL-17a and TNF-α levels and a decrease in IL-10. Cytokine signature analysis revealed a shift in the microenvironment towards a pro-inflammatory profile mediated by IL-8 and IFN-γ. Both, CD4(+) (↑TNF-α(+) and ↑IFN-γ (+)) and CD8(+) (↑IL-17a and ↓IL-4) T-cells contributed to the acquired immune responses observed after stimulation with SLAg. CONCLUSIONS: The changes observed in the cytokine profile suggested that Leishmune® was able to induce an effective response at six months after primovaccination. After one year, it returned to baseline suggesting the need of additional boosting

    Variabilidad genética de lotes de brycon orbignyanus utilizados en programas de repoblamiento: manejo y conservación

    Alteraciones ambientales causadas por el calentamiento global y principalmente causa-das por la acción del hombre, han reducido poblaciones naturales de peces. Como forma de conservación, programas de repoblamiento han sido utilizados; sin embargo, sin una debida orientación científica, estas medidas pueden generar disturbios genéticos sobre la diversidad genética de poblaciones de peces naturales y sobre el ecosistema. El objetivo de este estudio fue estimar y analizar la variabilidad genética de dos lotes y una progenie de Brycon orbignyanus utilizados en programas de repoblamiento, utilizando el marcador molecular RAPD (Random Amplified Polymorphic DNA). Cincuenta y ocho reproductores de dos lotes (A y C) y 30 larvas de la progenie del lote A (B) pertenecientes a la Estação de Aqüicultura e Hidrologia da Duke Energy Internacional (Geração Parana-panema; São Paulo, Brasil) fueron analizados. Los resultados de variabilidad genética estimados por el índice de diversidad de Shannon (A: 0,3184; B: 0,3433 y C: 0,3687) y por el porcentaje de fragmentos polimórficos (A: 54,02%; B: 57,47% y C: 58,62%) mostraron que la variabilidad genética fue mantenida en la progenie, debido posiblemente al adecuado manejo reproductivo y al efecto fundador. Por el contrario, la variabilidad encontrada entre los dos lotes de reproductores indica una similaridad genética, a pesar de ser originarios de diferentes pisciculturas. Este resultado es comprobado en el valor moderado de diferenciación genética encontrado (0,0968), en el alto Nm (4,67) y en el dendrograma, que sugieren que los lotes poseen un pool genético simila

    Alimentação popular em São Paulo (1920 a 1950): políticas públicas, discursos técnicos e práticas profissionais

    This article discusses how the concept of lower-class eating habits came about and developed in the intellectual circles of São Paulo during the first half of the 20th century. It starts by reconstructing the elements of the debate around the income and ignorance of the underprivileged as the main reasons behind their bad eating habits. Then, it looks at the focal points for interventions and public policies proposed by the government to deal with the problem thus identified, namely: training methods to produce sanitation counselors capable of offering dietary guidance as well; popular educational campaigns and new learning sites in addition to schools (e.g. healthcare centers and households); lunch and other means of offering food at schools; and diagnostic studies about food intake and eating habits among laborers. Because they were translated into technical and scientific language, the proposals and policies implemented in São Paulo left traces in a variety of supporting documents and media (photographs, primers, posters, inquiry notebooks, and academic literature).O artigo discute a construção da idéia de alimentação popular nos meios intelectuais em São Paulo, na primeira metade do século XX. Para isso, reconstitui, como motivos da má alimentação, elementos do debate em torno da renda e da ignorância dos mais pobres. Identificado o problema, as propostas de intervenção e as políticas públicas concentraram-se em alguns setores, abordados neste trabalho: métodos para a formação de educadores sanitários aptos a atuar também na educação alimentar; campanhas de instrução popular e criação de novos lugares de aprendizado (além das escolas, os centros de saúde e os lares); merenda escolar e outras alternativas de alimentação nas escolas; e diagnósticos referentes ao conteúdo e à forma da alimentação dos operários.     Iron Deficiency Generates Oxidative Stress and Activation of the SOS Response in Caulobacter crescentus

    In C. crescentus, iron metabolism is mainly controlled by the transcription factor Fur (ferric uptake regulator). Iron-bound Fur represses genes related to iron uptake and can directly activate the expression of genes for iron-containing proteins. In this work, we used total RNA sequencing (RNA-seq) of wild type C. crescentus growing in minimal medium under iron limitation and a fur mutant strain to expand the known Fur regulon, and to identify novel iron-regulated genes. The RNA-seq of cultures treated with the iron chelator 2-2-dypiridyl (DP) allowed identifying 256 upregulated genes and 236 downregulated genes, being 176 and 204 newly identified, respectively. Sixteen transcription factors and seven sRNAs were upregulated in iron limitation, suggesting that the response to low iron triggers a complex regulatory network. Notably, lexA along with most of its target genes were upregulated, suggesting that DP treatment caused DNA damage, and the SOS DNA repair response was activated in a RecA-dependent manner, as confirmed by RT-qPCR. Fluorescence microscopy assays using an oxidation-sensitive dye showed that wild type cells in iron limitation and the fur mutant were under endogenous oxidative stress, and a direct measurement of cellular H2O2 showed that cells in iron-limited media present a higher amount of endogenous H2O2. A mutagenesis assay using the rpoB gene as a reporter showed that iron limitation led to an increase in the mutagenesis rate. These results showed that iron deficiency causes C. crescentus cells to suffer oxidative stress and to activate the SOS response, indicating an increase in DNA damage

    Serological reactivity against T. cruzi-derived antigens: Evaluation of their suitability for the assessment of response to treatment in chronic Chagas disease.

    Chagas disease, caused by the protozoan Trypanosoma cruzi, affects more than 6 million people worldwide. Following a mostly asymptomatic acute phase, the disease progresses to a long-lasting chronic phase throughout which life-threatening disorders to the heart and/or gastrointestinal tract will manifest in about 30% of those chronically infected. During the chronic phase, the parasitemia is low and intermittent, while a high level of anti-T. cruzi antibodies persist for years. These two features hamper post-chemotherapeutic follow-up of patients with the tools available. The lack of biomarkers for timely assessment of therapeutic response discourages a greater use of the two available anti-parasitic drugs, and complicates the evaluation of new drugs in clinical trials. Herein, we investigated in a blinded case-control study the serological reactivity over time of a group of parasite-derived antigens to potentially address follow up of T. cruzi chronically infected subjects after treatment. We tested PFR2, KMP11, HSP70, 3973, F29 and the InfYnity multiplexed antigenic array, by means of serological assays on a multi-national retrospective collection of samples. Some of the antigens exhibited promising results, underscoring the need for further studies to determine their potential role as treatment response biomarkers.We thank Dr. A. Egui, Dr A.     ANTHELMINTIC EFFECTS OF DRIED GROUND BANANA PLANT LEAVES (MUSA SPP.) FED TO SHEEP ARTIFICIALLY INFECTED WITH HAEMONCHUS CONTORTUS AND TRICHOSTRONGYLUS COLUBRIFORMIS

    Background: Helminths is a endoparasites that cause the major losses for profitable sheep production in Brazil. The increased development of resistant strains of endoparasites have enforced the search for sustainable alternatives. The aim of this paper was to provide information about endoparasites control with banana leaves in infected sheep as alternative control strategies and see its viability. Materials and Methods: In this study, we performed two trials to investigate the anthelmintic properties of banana leaves on endoparasites in sheep. In Trial 1, twelve sheep were artificially infected with Trichostrongylus colubriformis; in Trial 2, eleven sheep were artificially infected with Haemonchus contortus. Clinical examinations, packed cell volume, total protein, faecal egg counts (FECs) and egg hatchability tests (EHTs) were performed. At the end of the trials, the sheep were humanely slaughtered, and total worm counts were performed. Results: In Trial 1 and 2, no significant FEC decreases were note but significant diference in EHTs were observed. Total worm counts, clinical and haematological parameters did not reveal significant changes between the treatment and control groups. These results suggest that feeding dried ground banana plant leaves to sheep may reduce the viability of Trichostrongylus colubriformis eggs, and this anthelmintic activity is potentially exploitable as part of an integrated parasite management programme. Conclusion: However, further investigation is needed to establish the optimal dosage, develop a convenient delivery form and confirm the economic feasibility of using banana plantation byproducts as feed for ruminant species

    Seasonal variation of water uptake of a Quercus suber tree in Central Portugal

    Hydraulic redistribution (HR) is the phenomenon where plant roots transfer water between soil horizons of different water potential. When dry soil is a stronger sink for water loss from the plant than transpiration, water absorbed by roots in wetter soil horizons is transferred toward, and exuded into dry soil via flow reversals through the roots. Reverse flow is a good marker of HR and can serve as a useful tool to study it over the long-term. Seasonal variation of water uptake of a Quercus suber tree was studied from late winter through autumn 2003 at Rio Frio near Lisbon, Portugal. Sap flow was measured in five small shallow roots (diameter of 3–4 cm), 1 to 2 m from the tree trunk and in four azimuths and at different xylem depths at the trunk base, using the heat field deformation method (HFD). The pattern of sap flow differed among lateral roots as soil dried with constant positive flow in three roots and reverse flow in two other roots during the night when transpiration ceased. Rain modified the pattern of flow in these two roots by eliminating reverse flow and substantially increasing water uptake for transpiration during the day. The increase in water uptake in three other roots following rain was not so substantial. In addition, the flux in individual roots was correlated to different degrees with the flux at different radial depths and azimuthal directions in trunk xylem. The flow in outer trunk xylem seemed to be mostly consistent with water movement from surface soil horizons, whereas deep roots seemed to supply water to the whole cross-section of sapwood. When water flow substantially decreased in shallow lateral roots and the outer stem xylem during drought, water flow in the inner sapwood was maintained, presumably due to its direct connection to deep roots. Results also suggest the importance of the sap flow sensor placement, in relation to sinker roots, as to whether lateral roots might be found to exhibit reverse flow during drought. This study is consistent with the dimorphic rooting habit of Quercus suber trees in which deep roots access groundwater to supply superficial roots and the whole tree, when shallow soil layers were dry
