    Removal of disinfection by-product precursors by coagulation and an innovative suspended ion exchange process

    This investigation aimed to compare the disinfection by-product formation potentials (DBPFPs) of three UK surface waters (1 upland reservoir and 2 lowland rivers) with differing characteristics treated by (a) a full scale conventional process and (b) pilot scale processes using a novel suspended ion exchange (SIX) process and inline coagulation (ILCA) followed by ceramic membrane filtration (CMF). Liquid chromatography-organic carbon detection analysis highlighted clear differences between the organic fractions removed by coagulation and suspended ion exchange. Pretreatments which combined SIX and coagulation resulted in significant reductions in dissolved organic carbon (DOC), UV absorbance (UVA), trihalomethane and haloacetic acid formation potential (THMFP, HAAFP), in comparison with the SIX or coagulation process alone. Further experiments showed that in addition to greater overall DOC removal, the processes also reduced the concentration of brominated DBPs and selectively removed organic compounds with high DBPFP. The SIX/ILCA/CMF process resulted in additional removals of DOC, UVA, THMFP, HAAFP and brominated DBPs of 50, 62, 62, 62% and 47% respectively compared with conventional treatment

    The Impact of Lateral Gene Transfer in Chlamydia

    Lateral gene transfer (LGT) facilitates many processes in bacterial ecology and pathogenesis, especially regarding pathogen evolution and the spread of antibiotic resistance across species. The obligate intracellular chlamydiae, which cause a range of diseases in humans and animals, were historically thought to be highly deficient in this process. However, research over the past few decades has demonstrated that this was not the case. The first reports of homologous recombination in the Chlamydiaceae family were published in the early 1990s. Later, the advent of whole-genome sequencing uncovered clear evidence for LGT in the evolution of the Chlamydiaceae, although the acquisition of tetracycline resistance in Chlamydia (C.) suis is the only recent instance of interphylum LGT. In contrast, genome and in vitro studies have shown that intraspecies DNA exchange occurs frequently and can even cross species barriers between closely related chlamydiae, such as between C. trachomatis, C. muridarum, and C. suis. Additionally, whole-genome analysis led to the identification of various DNA repair and recombination systems in C. trachomatis, but the exact machinery of DNA uptake and homologous recombination in the chlamydiae has yet to be fully elucidated.     Pre-treatment of surface waters for ceramic microfiltration

    The influence of pre-treatment on the suppression of irreversible (IR) fouling of ceramic membranes challenged with three UK surface waters has been studied at pilot scale. An initial scoping study compared the efficacy of suspended ion exchange (SIX) and clarification (coagulation followed by sludge blanket clarification) individually and in combination. Direct membrane filtration following in-line coagulation (ILCA) was also investigated with and without SIX. The impact on the various organic fractions, specifically high molecular weight (HMW) biopolymers (BPs) and humic substances (HSs), and lower molecular weight (LMW) building blocks (BBs) and neutrals, was studied using liquid chromatography-organic carbon detection (LC-OCD). Results revealed SIX and coagulation to preferentially remove the LMW and HMW organic fractions respectively. Residual HMW organic matter (primarily BPs) following SIX pre-treatment were retained by the membrane which led to rapid irreversible fouling. Coagulation pre-treatment provided stable membrane operation and the residual LMW organics were not significantly retained by the membrane. Combining clarification and SIX resulted in significantly increased removal of organics and lower membrane fouling rates. Tests performed using SIX and ILCA revealed high dissolved organic carbon (DOC) removal compared to SIX with clarification. However, unlike the case for clarification with SIX, the addition of SIX to optimised ILCA dosing offered no additional suppression of membrane fouling compared to ILCA alone. Optimised ILCA pretreatment led to very low IR fouling rates of <0.3 kPa/day trans-membrane pressure, despite highly challenging operating conditions of elevated fluxes (185 L m−2 h−1) and highly variable feedwater dissolved organic carbon concentrations

    Zoomshock : the geography and local labour market consequences of working from home

    The Covid-19 health crisis has led to a substantial increase in work done from home, which shifts economic activity across geographic space. We refer to this shift as a Zoomshock. The Zoomshock has implications for locally consumed services; much of the clientele of restaurants, coffee bars, pubs, hair stylists, health clubs, and the like located near workplaces is transferred to establishments located near where people live. In this paper we measure the Zoomshock at a very granular level for UK neighbourhoods. We establish three important empirical facts. First, the Zoomshock is large; many workers can work-from-home and live in a different neighbourhood than they work. Second, the Zoomshock is very heterogeneous; economic activity is decreasing in productive city centres and increasing residential suburbs. Third, the Zoomshock moves workers away from neighbourhoods with a large supply of locally consumed services to neighbourhoods where the supply of these services is relatively scarce. We discuss the implications for aggregate employment and local economic recovery following the Covid-19 health crisis

    Do remote workers deter neighborhood crime? Evidence from the rise of working from home

    We examine the impact of the working from home (WFH) shift on neighborhood-level burglary rates, employing detailed street-level crime data and a neighborhood WFH measure. We find a one standard deviation increase in WFH (9.5pp) leads to a persistent 4% drop in burglaries. A spatial search model identifies two deterrence channels: occupancy, as burglars avoid occupied houses, and “eyes on the street”. We provide evidence supporting both channels. Despite crime displacement to low WFH areas offseting 30% of the burglary eduction, a hedonic pricing model reveals significant willingness to pay for high WFH areas, especially those with high ex-ante burglary risk

    Amoxicillin-Clavulanate-Induced Liver Injury

    Background and Aims Amoxicillin–clavulanate (AC) is the most frequent cause of idiosyncratic drug-induced injury (DILI) in the US DILI Network (DILIN) registry. Here, we examined a large cohort of AC-DILI cases and compared features of AC-DILI to those of other drugs. Methods Subjects with suspected DILI were enrolled prospectively, and cases were adjudicated as previously described. Clinical variables and outcomes of patients with AC-DILI were compared to the overall DILIN cohort and to DILI caused by other antimicrobials. Results One hundred and seventeen subjects with AC-DILI were identified from the cohort (n = 1038) representing 11 % of all cases and 24 % of those due to antimicrobial agents (n = 479). Those with AC-DILI were older (60 vs. 48 years, P < 0.001). AC-DILI was more frequent in men than women (62 vs. 39 %) compared to the overall cohort (40 vs. 60 %, P < 0.001). The mean time to symptom onset was 31 days. The Tb, ALT, and ALP were 7 mg/dL, 478, and 325 U/L at onset. Nearly all liver biopsies showed prominent cholestatic features. Resolution of AC-DILI, defined by return of Tb to <2.5 mg/dL, occurred on average 55 days after the peak value. Three female subjects required liver transplantation, and none died due to DILI. Conclusion AC-DILI causes a moderately severe, mixed hepatocellular–cholestatic injury, particularly in older men, unlike DILI in general, which predominates in women. Although often protracted, eventual apparent recovery is typical, particularly for men and usually in women, but three women required liver transplantation

    Covid reallocation of spending : the effect of remote working on the retail and hospitality sector

    A defining economic outcome from the Covid-19 pandemic is the unprecedented shift towards remote working from home. The extent and duration of this shift will have important consequences for local economies and especially the retail and hospitality sectors which depend on business around the workplace. Using a new bespoke, nationally representative survey of UK working age adults we analyse their ability and willingness to work remotely, and the consequences for spending on food, beverages, retail and entertainment around the workplace. We establish five key facts. (i) The post-pandemic change will be large: the fraction of work done from home will increase by 20 percentage points over its pre-pandemic level. (ii) The Dingel-Neiman (2020) assessment of remote working potential by occupation are reasonably predictive of what workers and employers expect to do, with a correlation coefficient of over 0.7. (iii) Relocation will be higher for better paid professional occupations, which will skew spending toward the most socio-economically affluent geographical areas. (iv) The corresponding geographical shift in annual retail and hospitality spending will be ÂŁ3.0 billion with more remote working shifting demand away from urban areas. (v) On average, a 1% change in neighbourhood workforce changes local spending by 0.25%

    Reliability of causality assessment for drug, herbal and dietary supplement hepatotoxicity in the Drug‐Induced Liver Injury Network (DILIN)

    Background & AimsBecause of the lack of objective tests to diagnose drug‐induced liver injury (DILI), causality assessment is a matter of debate. Expert opinion is often used in research and industry, but its test–retest reliability is unknown. To determine the test–retest reliability of the expert opinion process used by the Drug‐Induced Liver Injury Network (DILIN).MethodsThree DILIN hepatologists adjudicate suspected hepatotoxicity cases to one of five categories representing levels of likelihood of DILI. Adjudication is based on retrospective assessment of gathered case data that include prospective follow‐up information. One hundred randomly selected DILIN cases were re‐assessed using the same processes for initial assessment but by three different reviewers in 92% of cases.ResultsThe median time between assessments was 938 days (range 140–2352). Thirty‐one cases involved >1 agent. Weighted kappa statistics for overall case and individual agent category agreement were 0.60 (95% CI: 0.50–0.71) and 0.60 (0.52–0.68) respectively. Overall case adjudications were within one category of each other 93% of the time, while 5% differed by two categories and 2% differed by three categories. Fourteen per cent crossed the 50% threshold of likelihood owing to competing diagnoses or atypical timing between drug exposure and injury.ConclusionsThe DILIN expert opinion causality assessment method has moderate interobserver reliability but very good agreement within one category.
