Reducing the burden of rural water supply through greywater reuse: A case study from northern Malawi
Greywater reuse has potential for non-potable applications that conserve freshwater resources in water-stressed areas especially in sub-Saharan Africa. The feasibility of reusing greywater for domestic activities in a rural area of Malawi, Africa, was evaluated from microbiological and public acceptance perspectives. Median Escherichia coli concentrations for eight domestic greywater sources (handwashing, laundry, runoff from a tap apron, bathing, cleaning a home/kitchen, cleaning a water collection container, washing plates and soaking vegetables) ranged from 100 to \u3e20, 000 colony forming units (cfu)/100 ml. Twenty-four of 47 greywater samples tested (51%) met the World Health Organization guideline for unrestricted use of greywater for irrigation. Pertinently, 80% (4/5) and 60% (3/5) of greywater samples from handwashing stations and bathing had E. coli less than the WHO guideline. Users reported greatest acceptance of reusing greywater for growing food and washing clothes, especially when the greywater source was bathing. Acceptance was closely tied to a household\u27s economic standing, geographic location, and first-hand knowledge of reusing greywater. Greywater reuse practices in rural areas, especially targeting bathing water as suitable from bacteriological and user perception criteria, can help mitigate the impacts of water stress in sub-Saharan Africa
Early Increase in Extrasynaptic NMDA Receptor Signaling and Expression Contributes to Phenotype Onset in Huntington's Disease Mice
SummaryN-methyl-D-aspartate receptor (NMDAR) excitotoxicity is implicated in the pathogenesis of Huntington's disease (HD), a late-onset neurodegenerative disorder. However, NMDARs are poor therapeutic targets, due to their essential physiological role. Recent studies demonstrate that synaptic NMDAR transmission drives neuroprotective gene transcription, whereas extrasynaptic NMDAR activation promotes cell death. We report specifically increased extrasynaptic NMDAR expression, current, and associated reductions in nuclear CREB activation in HD mouse striatum. The changes are observed in the absence of dendritic morphological alterations, before and after phenotype onset, correlate with mutation severity, and require caspase-6 cleavage of mutant huntingtin. Moreover, pharmacological block of extrasynaptic NMDARs with memantine reversed signaling and motor learning deficits. Our data demonstrate elevated extrasynaptic NMDAR activity in an animal model of neurodegenerative disease. We provide a candidate mechanism linking several pathways previously implicated in HD pathogenesis and demonstrate successful early therapeutic intervention in mice
CUMYL-4CN-BINACA Is an Efficacious and Potent Pro-Convulsant Synthetic Cannabinoid Receptor Agonist
Synthetic cannabinoid receptor agonists (SCRAs) are the largest class of new psychoactive substances (NPS). New examples are detected constantly, and some are associated with a series of adverse effects, including seizures. CUMYL-4CN-BINACA (1-(4-cyanobutyl)-N-(2-phenylpropan-2-yl)indazole-3-carboxamide) is structurally related to potent, cumylamine-derived SCRAs such as 5F-CUMYL-PINACA, but is unusual due to a terminal aliphatic nitrile group not frequently encountered in SCRAs or pharmaceuticals. We report here that CUMYL-4CN-BINACA is a potent CB1 receptor agonist (Ki = 2.6ânM; EC50 = 0.58ânM) that produces pro-convulsant effects in mice at a lower dose than reported for any SCRA to date (0.3âmg/kg, i.p). Hypothermic and pro-convulsant effects in mice could be reduced or blocked, respectively, by pretreatment with CB1 receptor antagonist SR141716, pointing to at least partial involvement of CB1 receptors in vivo. Pretreatment with CB2 receptor antagonist AM-630 had no effect on pro-convulsant activity. The pro-convulsant properties and potency of CUMYL-4CN-BINACA may underpin the toxicity associated with this compound in humans
Eutrophication and acidification: Do they induce changes in the dissolvedorganic matter dynamics in the coastal Mediterranean Sea?
Original research paperTwo mesocosms experiments were conducted in winter 2010 and summer 2011 to examine how increased pCO2and/or nutrient concentrations potentially perturbate dissolved organic matter dynamics in natural microbialassemblages. Thefluorescence signals of protein- and humic-like compounds were used as a proxy for labileand non-labile material, respectively, while the evolution of bacterial populations, chlorophylla(Chla) anddissolved organic carbon (DOC) concentrations were used as a proxy for biological activity. For both seasons,the presence of elevated pCO2did not cause any significant change in the DOC dynamics (p-valueb0.05). Theconditions that showed the greatest changes in prokaryote abundances and Chlacontent were those amendedwith nutrients, regardless of the change in pH. The temporal evolution offluorophores and optical indices re-vealed that the degree of humification of the organic molecules and their molecular weight changed significantlyin the nutrient-amended treatment. The generation of protein-like compounds was paired to increases in theprokaryote abundance, being higher in the nutrient-amended tanks than in the control. Different patterns inthe magnitude and direction of the generation of humic-like molecules suggested that these changes dependedon initial microbial populations and the availability of extra nutrient inputs. Gene and cell therapy for cystic fibrosis: From bench to bedside
Clinical trials in cystic fibrosis (CF) patients established proof-of-principle for transfer of the wild-type cystic fibrosis transmembrane conductance regulator (CFTR) gene to airway epithelial cells. However, the limited efficacy of gene transfer vectors as well as extra- and intracellular barriers have prevented the development of a gene therapy-based treatment for CF. Here, we review the use of new viral and nonviral gene therapy vectors, as well as human artificial chromosomes, to overcome barriers to successful CFTR expression. Pre-clinical studies will surely benefit from novel animal models, such as CF pigs and ferrets. Prenatal gene therapy is a potential alternative to gene transfer to fully developed lungs. However, unresolved issues, including the possibility of adverse effects on pre- and postnatal development, the risk of initiating oncogenic or degenerative processes and germ line transmission require further investigation. Loneliness of Older Immigrant Groups in Canada: Effects of Ethnic-Cultural Background
This study aimed to explore the loneliness of several groups of older immigrants in Canadacompared to native-born older adults. Data from the Canadian General Social Survey, Cycle 22 (Nolder adults = 3,692) were used. The dependent variable is the 6 item De Jong Gierveld lonelinessscale. Determinants of loneliness included country of birth, ethnic background (cultural context);belongingness (community context) and social networks (social context). Results showed that onlysome immigrant groups are significantly lonelier than older adults born in Canada. Immigrants withsimilar language and culture are not lonelier; while those from countries that differ in nativelanguage/culture are significantly higher on loneliness. Multivariate analyses showed the importanceof cultural background, of composition of the network of relatives and friends, and of localparticipation and feelings of belonging to the Canadian society in explaining loneliness of olderimmigrants
Negation and the functional sequence
There exists a general restriction on admissible functional sequences which prevents adjacent identical heads. We investigate a particular instantiation of this restriction in the domain of negation. Empirically, it manifests itself as a restriction the stacking of multiple negative morphemes. We propose a principled account of this restriction in terms of the general ban on immediately consecutive identical heads in the functional sequence on the one hand, and the presence of a Neg feature inside negative morphemes on the other hand. The account predicts that the stacking of multiple negative morphemes should be possible provided they are separated by intervening levels of structure. We show that this prediction is borne out

Proceedings of the 3rd Biennial Conference of the Society for Implementation Research Collaboration (SIRC) 2015: advancing efficient methodologies through community partnerships and team science
It is well documented that the majority of adults, children and families in need of evidence-based behavioral health interventionsi do not receive them [1, 2] and that few robust empirically supported methods for implementing evidence-based practices (EBPs) exist. The Society for Implementation Research Collaboration (SIRC) represents a burgeoning effort to advance the innovation and rigor of implementation research and is uniquely focused on bringing together researchers and stakeholders committed to evaluating the implementation of complex evidence-based behavioral health interventions. Through its diverse activities and membership, SIRC aims to foster the promise of implementation research to better serve the behavioral health needs of the population by identifying rigorous, relevant, and efficient strategies that successfully transfer scientific evidence to clinical knowledge for use in real world settings [3]. SIRC began as a National Institute of Mental Health (NIMH)-funded conference series in 2010 (previously titled the âSeattle Implementation Research Conferenceâ; $150,000 USD for 3 conferences in 2011, 2013, and 2015) with the recognition that there were multiple researchers and stakeholdersi working in parallel on innovative implementation science projects in behavioral health, but that formal channels for communicating and collaborating with one another were relatively unavailable. There was a significant need for a forum within which implementation researchers and stakeholders could learn from one another, refine approaches to science and practice, and develop an implementation research agenda using common measures, methods, and research principles to improve both the frequency and quality with which behavioral health treatment implementation is evaluated. SIRCâs membership growth is a testament to this identified need with more than 1000 members from 2011 to the present.ii SIRCâs primary objectives are to: (1) foster communication and collaboration across diverse groups, including implementation researchers, intermediariesi, as well as community stakeholders (SIRC uses the term âEBP championsâ for these groups) â and to do so across multiple career levels (e.g., students, early career faculty, established investigators); and (2) enhance and disseminate rigorous measures and methodologies for implementing EBPs and evaluating EBP implementation efforts. These objectives are well aligned with Glasgow and colleaguesâ [4] five core tenets deemed critical for advancing implementation science: collaboration, efficiency and speed, rigor and relevance, improved capacity, and cumulative knowledge. SIRC advances these objectives and tenets through in-person conferences, which bring together multidisciplinary implementation researchers and those implementing evidence-based behavioral health interventions in the community to share their work and create professional connections and collaborations
