    A reversed-phase high-performance liquid chromatographic (HPLC) method was developed and validated for the determination of encapsulation efficiency of zidovudine in nanoparticules. The method was carried out in isocratic mode using 0.040M sodium acetate: methanol: acetonitrile: glacial acetic acid (880:100:20:2) as mobile phase, a C8 column at 25ºC and UV detection at 240 nm. The method was linear (r2 ˃ 0.99) over the range of 25.0-150.0 μg/mL, precise (RSD ˂ 5%), accurate (recovery = 100.5%), robust and selective.     Anxiety and depression symptoms after pulmonary rehabilitation in people with interstitial lung disease: responders and non-responders

    Pulmonary rehabilitation (PR) is an effective intervention for people with interstitial lung disease (ILD), as it improvesexercise capacity, quality of life and dyspnoea. Less is, however, known about other important and frequentsymptoms, such as anxiety and depression. This study explored the response of anxiety and depression symptomsto PR in people with ILD. Participants undertook a 12-week community-based PR programme. Anxiety and depression symptoms wereevaluated before and after PR with the hospital anxiety and depression scale (HADS). The sample was stratified into2 groups according to the HADS score: ≥8 probable anxiety (PA) or depression (PD) and score <8 normal symptoms(NS). Responders were classified using the HADS minimal clinical important difference (≥1.5 points). 46 people with ILD (62±13 years; 63%♀; 75±20 FVC%; 56±18 DLCO%) participated. Number of responders inanxiety symptoms (73% PA; HADS-A pre: 12±3; mean change: 4±2; p<0.001 vs 25% NS; HADS-A pre: 6±2; meanchange: 2±0; p<0.001) was much higher than in depression symptoms (27% PD; HADS-D pre: 11±3; mean change:3±1; p=0.014 vs 13% NS; HADS-D pre: 5±2; mean change: 2±1; p=0.003). PR seems to improve anxiety symptoms, but it might not be as effective for depression symptoms in people with ILD.Future studies are needed to investigate the role of PR in the psychological health of people with ILD.publishe

    Antidepressant-like effects of chronic guanosine in the olfactory bulbectomy mouse model

    Major depressive disorder (MDD) leads to pervasive changes in the health of afflicted patients. Despite advances in the understanding of MDD and its treatment, profound innovation is needed to develop fast-onset antidepressants with higher effectiveness. When acutely administered, the endogenous nucleoside guanosine (GUO) shows fast-onset antidepressant-like effects in several mouse models, including the olfactory bulbectomy (OBX) rodent model. OBX is advocated to possess translational value and be suitable to assess the time course of depressive-like behavior in rodents. This study aimed at investigating the long-term behavioral and neurochemical effects of GUO in a mouse model of depression induced by bilateral bulbectomy (OBX). Mice were submitted to OBX and, after 14 days of recovery, received daily (ip) administration of 7.5 mg/kg GUO or 40 mg/kg imipramine (IMI) for 45 days. GUO and IMI reversed the OBX-induced hyperlocomotion and recognition memory impairment, hippocampal BDNF increase, and redox imbalance (ROS, NO, and GSH levels). GUO also mitigated the OBX-induced hippocampal neuroinflammation (IL-1, IL-6, TNF-α, INF-γ, and IL-10). Brain microPET imaging ([18F]FDG) shows that GUO also prevented the OBX-induced increase in hippocampal FDG metabolism. These results provide additional evidence for GUO antidepressant-like effects, associated with beneficial neurochemical outcomes relevant to counteract depression

    Casos confirmados de Esquistossomose no Pará no período de 2007 a 2017 / Confirmed cases of Schistosomiasis in Pará in the period 2007 to 2017

    Introdução: A esquistossomose é uma doença crônica causada pelos parasitas multicelulares platelmintos do gênero Schistosoma. A esquistossomose possui o homem como hospedeiro definitivo, modificações ambientais e condições inadequadas de saneamento básico são as principais responsáveis pelo aumento do número de transmissores e de seus focos de transmissão. Sendo uma doença relacionada fortemente com as condições socioeconômicas da população.Objetivo: Analisar os casos confirmados de Esquistossomose de 2007 a 2017 no Pará.Método: Trata-se de um estudo descritivo, feito com base em dados recolhidos por meio de fichas de notificação compulsórias do SINAN presentes no DATASUS. Foram recolhidas informações condizentes com o ano da notificação, cidades e escolaridade durante o período de 2007 a 2017.Resultados: Foram encontrados 174 casos notificados, sendo que a maioria ocorreu no ano de 2009 (22,9%), enquanto que minoria ocorreu no ano de 2007 (2,2%). Além disso a cidade mais afetada foi a cidade de Altamira (23,5%), porém acredita-se que possam existir subnotificações em outras cidades do estado. No que diz respeito a escolaridade, os mais afetados foram pacientes com escolaridade da 1ª à 4ª série incompleta do ensino fundamental (20,6%), seguidos pelos analfabetos (17,8%).Conclusão: Foi visto um acréscimo aos casos de notificação compulsória de Esquistossomose no Pará em um comparativo dos anos de 2007 e 2017. No entanto, acredita-se que ainda há muitas subnotificações, principalmente nas cidades mais interioranas.

    MIXED MODELS FOR NUTRIENTS PREDICTION IN SPECIES OF THE BRAZILIAN CAATINGA BIOME

    ABSTRACT Nutrient prediction models applied to tree species from Brazilian Caatinga can be a crucial tool in understanding this biome. The study aimed to fit a mixed model to predict nitrogen (N), phosphorus (P), and potassium (K) content in tree species native to the Caatinga biome located in Floresta municipality, Pernambuco State – PE, Brazil. The following species were considered the area’s most important and evaluated in the present study: Poincianella bracteosa (Tul.) L.P.Queiroz, Mimosa ophtalmocentra Mart. ex Benth, Aspidosperma pyrifolium Mart, Cnidoscolus quercifolius (Mull. Arg.) Pax. & Hoffm, and Anadenanthera colubrina var. cebil (Griseb.) Altschul. Four trees, representing the average circumference in each diameter class, were harvested for NPK quantification. The Spurr model was evaluated for NPK prediction, and species inclusion as a random effect was significant (p > 0.05) in all models. The Spurr model with fixed and random effects presented better statistics than fixed-effect models in all parameters for all nutrients. Generated NPK predicting equations can be a handy tool to understand the impact of wood extraction over Caatinga’s biogeochemical cycles and guide forest management strategies in semi-arid regions of the world

    Role of rutin in 5-Fluorouracil-induced intestinal mucositis : prevention of histological damage and reduction of inflammation and oxidative stress

    Intestinal mucositis, characterized by inflammatory and/or ulcerative processes in the gastrointestinal tract, occurs due to cellular and tissue damage following treatment with 5-fluorouracil (5-FU). Rutin (RUT), a natural flavonoid extracted from Dimorphandra gardneriana, exhibits antioxidant, anti-inflammatory, cytoprotective, and gastroprotective properties. However, the effect of RUT on inflammatory processes in the intestine, especially on mucositis promoted by antineoplastic agents, has not yet been reported. In this study, we investigated the role of RUT on 5-FU-induced experimental intestinal mucositis. Swiss mice were randomly divided into seven groups: Saline, 5-FU, RUT-50, RUT-100, RUT-200, Celecoxib (CLX), and CLX + RUT-200 groups. The mice were weighed daily. After treatment, the animals were euthanized and segments of the small intestine were collected to evaluate histopathological alterations (morphometric analysis); malondialdehyde (MDA), myeloperoxidase (MPO), and glutathione (GSH) concentrations; mast and goblet cell counts; and cyclooxygenase-2 (COX-2) activity, as well as to perform immunohistochemical analyses. RUT treatment (200 mg/kg) prevented 5-FU-induced histopathological changes and reduced oxidative stress by decreasing MDA concentrations and increasing GSH concentrations. RUT attenuated the inflammatory response by decreasing MPO activity, intestinal mastocytosis, and COX-2 expression. These results suggest that the COX-2 pathway is one of the underlying protective mechanisms of RUT against 5-FU-induced intestinal mucositis

    Pervasive gaps in Amazonian ecological research

    Biodiversity loss is one of the main challenges of our time,1,2 and attempts to address it require a clear un derstanding of how ecological communities respond to environmental change across time and space.3,4 While the increasing availability of global databases on ecological communities has advanced our knowledge of biodiversity sensitivity to environmental changes,5–7 vast areas of the tropics remain understudied.8–11 In the American tropics, Amazonia stands out as the world’s most diverse rainforest and the primary source of Neotropical biodiversity,12 but it remains among the least known forests in America and is often underrepre sented in biodiversity databases.13–15 To worsen this situation, human-induced modifications16,17 may elim inate pieces of the Amazon’s biodiversity puzzle before we can use them to understand how ecological com munities are responding. To increase generalization and applicability of biodiversity knowledge,18,19 it is thus crucial to reduce biases in ecological research, particularly in regions projected to face the most pronounced environmental changes. We integrate ecological community metadata of 7,694 sampling sites for multiple or ganism groups in a machine learning model framework to map the research probability across the Brazilian Amazonia, while identifying the region’s vulnerability to environmental change. 15%–18% of the most ne glected areas in ecological research are expected to experience severe climate or land use changes by 2050.     Impacto clínico, terapêutico e social do canabidiol na epilepsia recorrente / Clinical, therapeutic and social impact of cannabidiol on recurrent epilepsy

    Introdução: Estima-se que mais de 50 milhões de pessoas no mundo tenham diagnóstico de epilepsia, no entanto, um terço dessas pessoas não possuem a patologia controlada, apresentando quadros de convulsão e epilepsia recorrente. Desta forma, buscar novas terapêuticas antiepilépticas são necessárias, a atividade antiepiléptica dos canabinóides têm-se mostrado com grande poder terapêutico. Objetivo: Responder quais os impactos clínicos, terapêuticos e sociais do uso do canabidiol (CBD) em pacientes com epilepsia recorrente. Metodologia: Trata-se de uma revisão bibliográfica no método de revisão de integrativa de literatura, realizado entre os meses de abril e maio de 2021, nas bases de dados da MEDLINE/Pubmed e LILACS, por meio dos descritores e operador booleano “Canabidiol” AND “Epilepsia”. Localizando-se artigos publicados entre os anos de 2016 e maio de 2021. Resultados e discussões: O mecanismo de ação ainda não é bem esclarecido, mas é reconhecido sua atuação sobre as manifestações psiquiátricas e neurológicas nas fendas pré-sinápticas. A terapia associada entre CBD e as terapias convencionais para epilepsia apresentaram melhora significativa nos sintomas epilépticos, havendo relatos de melhora completa dos sintomas em até 87,5% dos pacientes. No entanto, cerca de 10% dos pacientes queixam-se de efeitos indesejáveis e desagradáveis como aumento da ansiedade, angústia, medo, tremor e sudorese excessiva durante o tratamento, dado que seu efeito é dose dependente, necessitando de atenção do prescritor para os efeitos indesejados. O impacto social é relatado pelos pacientes como melhora na qualidade de vida e laboral, dada a diminuição da frequência de eventos convulsivos. Conclusão: A terapêutica associada do CBD com a terapia convencional na epilepsia recorrente mostra-se com grandes impactos clínicos e sociais para os pacientes acometidos. No entanto, requer atenção dos prescritores para manifestações clínicas de efeitos colaterais para readequação terapêutica e seguimento. Diante disto, a importância de novos estudos terapêuticos com o CBD é estabelecida pela necessidade de compreender mecanismos de ação e atuação em demais áreas complexas cerebrais que a epilepsia atua para ajudar no tratamento de diversas manifestações desta patologia neurológica
