    Conflict is an unavoidable fact of life and often creative. Conflicts occured when the purpose of society is not the same way. Various of disagreements and conflicts can usually be resolved without violence, and often by results in better situation for most or all of the parties involved. The purpose of this study to know perceptions and expectations of Tani Saiyo Cooperative towards conflict by PT. Citra Riau Sarana in Perhentian Luas District of Logas Tanah Darat Regency of Kuantan Singingi. Determination of the respondents used is key informan. Determination of sample conducted census sampling by thorough sampling techniques. Respondent was taken only 30 members of Tani Saiyo Cooperative as sample. The techniques of data collection used the method of survey. The data has beenob tained qualitative descriptive. The tabulation has done by making distribution of respondents from each of the variables studied. To describe perception variables conflict towards Tani Saiyo cooperative was used 5W+1H. While, expectation variable Scale of Liker's Summated Rating (LSR) was used. The results showed that:(1) Perceptions of Tani Saiyo Cooperative in Perhentian Luas Village has understanding of conflict issue by measured of perception towards of the conflict understanding, perceptions of understanding of the perpetrators purposes of conflict and perception of conflict perpetrators. (2) Expectations of Tani Saiyo cooperative towards of conflict in the category of high expectations with the score 3,60 as measured from goal, pathway thinking and agency thingking

    Remodeling of the neuromuscular junction precedes sarcopenia related alterations in myofibers

    Several mechanisms contributing to the etiology of sarcopenia (age-related loss of muscle size) have been postulated. One of these attributes the loss of muscle mass to a preceding age-related denervation of myofibers. The aim of this study was to determine if signs of denervation were apparent at the neuromuscular Junction (NMJ) before fiber atrophy, or fiber type conversion could be documented, and to reveal if a muscle\u27s activity level impacts its sensitivity to age-related denervation. Plantaris and soleus muscles were obtained from young adult (10 months) and early aged (21 months) rats. Pre- and post-synaptic NMJ morphology was quantified with cytofluorescent staining of nerve terminal branches and endplate regions, respectively Myofiber profiles (fiber size and fiber type composition) were assessed with histochemical procedures Results show that in the lightly recruited plantaris, significant (P \u3c 0.05) signs of denervation were noted in aged rats, while the same muscles displayed no change in myofiber profile. In the heavily recruited soleus, however, there was little evidence of denervation, and again no alterations in myofiber profile These results indicate that age-related denervation occurs before myofiber atrophy, and that high amounts of neuromuscular activity may delay the onset of age-related denervation and sarcopenia (C) 2010 Elsevier Inc All rights reserved

    Kegiatan pengabdian pada masyarakat ini merupakan kegiatan yang dilakukan sebagai wujud pembinaan kepada masyarakat. Pengabdian ini dilaksanakan pada anggota pramuka di SMA Negeri 5 Mataram. Kegiatan pengabdian ini dilaksanakan dalam rangka melakukan pembinaan materi berbicara di depan umum pada anggota pramuka. Pelatihan berbicara di depan umum ini dimaksudkan agar para anggota pramuka di gugus depan 06.101-06.102 Ambalan Udayana-Ratu Sima, mampu berbicara di depan umum dengan bahasa yang baik dan benar serta menjunjung kesantunan berbahasa, hal ini dimaksudkan untuk membentuk karakter anggota pramuka sejak dini, sehingga di kemudian hari mereka menjadi unggul di masyarakat dalam hal positif. Kegiatan tersebut dilaksanakan sebanyak 4 kali pertemuan dengan rincian: (1) Pertemuan I: Penyampaian materi tentang keterampilan berbicara di depan umum; (2) Pertemuan II: Pelatihan berpidato dan memandu acara; (3) Pertemuan III: Pelatihan berdiskusi dan memimpin rapat; (4) Pertemuan IV: Evaluasi kegiatan

    Epigenetic patterns in blood associated with lipid traits predict incident coronary heart disease events and are enriched for results from genome-wide association studies

    Background- Genome-wide association studies have identified loci influencing circulating lipid concentrations in humans; further information on novel contributing genes, pathways, and biology may be gained through studies of epigenetic modifications. Methods and Results- To identify epigenetic changes associated with lipid concentrations, we assayed genome-wide DNA methylation at cytosine-guanine dinucleotides (CpGs) in whole blood from 2306 individuals from 2 population-based cohorts, with replication of findings in 2025 additional individuals. We identified 193 CpGs associated with lipid levels in the discovery stage (P < 1.08E-07) and replicated 33 (at Bonferroni-corrected P < 0.05), including 25 novel CpGs not previously associated with lipids. Genes at lipid-associated CpGs were enriched in lipid and amino acid metabolism processes. A differentially methylated locus associated with triglyceridesand high-density lipoprotein cholesterol (HDL- C; cg27243685; P= 8.1E-26 and 9.3E-19) was associated with cis-expression of a reverse cholesterol transporter (ABCG1; P= 7.2E-28) and incident cardiovascular disease events (hazard ratio per SD increment, 1.38; 95% confidence interval, 1.15-1.66; P= 0.0007). We found significant cis-methylation quantitative trait loci at 64% of the 193 CpGs with an enrichment of signals from genome-wide association studies of lipid levels (P-TC = 0.004, PHDL-C = 0.008 and P-triglycerides = 0.00003) and coronary heart disease ( P= 0.0007). For example, genome-wide significant variants associated with low-density lipoprotein cholesterol and coronary heart disease at APOB were cis-methylation quantitative trait loci for a low-density lipoprotein cholesterol-related differentially methylated locus. Conclusions-We report novel associations of DNA methylation with lipid levels, describe epigenetic mechanisms related to previous genome-wide association studies discoveries, and provide evidence implicating epigenetic regulation of reverse cholesterol transport in blood in relation to occurrence of cardiovascular disease events

    No major flaws in "Identification of individuals by trait prediction using whole-genome sequencing data"

    In a recently published PNAS article, we studied the identifiability of genomic samples using machine learning methods [Lippert et al., 2017]. In a response, Erlich [2017] argued that our work contained major flaws. The main technical critique of Erlich [2017] builds on a simulation experiment that shows that our proposed algorithm, which uses only a genomic sample for identification, performed no better than a strategy that uses demographic variables. Below, we show why this comparison is misleading and provide a detailed discussion of the key critical points in our analyses that have been brought up in Erlich [2017] and in the media. Further, not only faces may be derived from DNA, but a wide range of phenotypes and demographic variables. In this light, the main contribution of Lippert et al. [2017] is an algorithm that identifies genomes of individuals by combining multiple DNA-based predictive models for a myriad of traits

    Association of Body Mass Index with DNA Methylation and Gene Expression in Blood Cells and Relations to Cardiometabolic Disease: A Mendelian Randomization Approach

    Background The link between DNA methylation, obesity, and adiposity-related diseases in the general population remains uncertain. Methods and Findings We conducted an association study of body mass index (BMI) and differential methylation for over 400,000 CpGs assayed by microarray in whole-blood-derived DNA from 3,743 participants in the Framingham Heart Study and the Lothian Birth Cohorts, with independent replication in three external cohorts of 4,055 participants. We examined variations in whole blood gene expression and conducted Mendelian randomization analyses to investigate the functional and clinical relevance of the findings. We identified novel and previously reported BMI-related differential methylation at 83 CpGs that replicated across cohorts; BMI-related differential methylation was associated with concurrent changes in the expression of genes in lipid metabolism pathways. Genetic instrumental variable analysis of alterations in methylation at one of the 83 replicated CpGs, cg11024682 (intronic to sterol regulatory element binding transcription factor 1 [SREBF1]), demonstrated links to BMI, adiposity-related traits, and coronary artery disease. Independent genetic instruments for expression of SREBF1 supported the findings linking methylation to adiposity and cardiometabolic disease. Methylation at a substantial proportion (16 of 83) of the identified loci was found to be secondary to differences in BMI. However, the cross-sectional nature of the data limits definitive causal determination. Conclusions We present robust associations of BMI with differential DNA methylation at numerous loci in blood cells. BMI-related DNA methylation and gene expression provide mechanistic insights into the relationship between DNA methylation, obesity, and adiposity-related diseases
