    Proof of concept of a method that assesses the spread of microbial infections with spatially explicit and non-spatially explicit data

    <p>Abstract</p> <p>Background</p> <p>A method that assesses bacterial spatial dissemination was explored. It measures microbial genotypes (defined by electrophoretic patterns or EP), host, location (farm), interfarm Euclidean distance, and time. Its proof of concept (construct and internal validity) was evaluated using a dataset that included 113 <it>Staphylococcus aureus </it>EPs from 1126 bovine milk isolates collected on 23 farms between 1988 and 2005.</p> <p>Results</p> <p>Construct validity was assessed by comparing results based on the interfarm Euclidean distance (a spatially explicit measure) and those produced by the (non-spatial) interfarm number of isolates reporting the same EP. The distance associated with EP spread correlated with the interfarm number of isolates/EP (<it>r </it>= .59, <it>P </it>< 0.02). Internal validity was estimated by comparing results obtained with different versions of the same indices. Concordance was observed between: (a) EP distance (estimated microbial dispersal over space) and EP speed (distance/year, <it>r </it>= .72, <it>P </it>< 0.01), and (b) the interfarm number of isolates/EP (when measured on the basis of non-repeated cow testing) and the same measure as expressed by repeated testing of the same animals (<it>r </it>= .87, <it>P </it>< 0.01). Three EPs (2.6% of all EPs) appeared to be super-spreaders: they were found in 26.75% of all isolates. Various indices differentiated local from spatially disseminated infections and, within the local type, infections suspected to be farm-related were distinguished from cow-related ones.</p> <p>Conclusion</p> <p>Findings supported both construct and internal validity. Because 3 EPs explained 12 times more isolates than expected and at least twice as many isolates as other EPs did, false negative results associated with the remaining EPs (those erroneously identified as lacking spatial dispersal when, in fact, they disseminated spatially), if they occurred, seemed to have negligible effects. Spatial analysis of laboratory data may support disease surveillance systems by generating hypotheses on microbial dispersal ability.</p

    Transmission of MRSA between Companion Animals and Infected Human Patients Presenting to Outpatient Medical Care Facilities

    Methicillin-resistant Staphylococcus aureus (MRSA) is a significant pathogen in both human and veterinary medicine. The importance of companion animals as reservoirs of human infections is currently unknown. The companion animals of 49 MRSA-infected outpatients (cases) were screened for MRSA carriage, and their bacterial isolates were compared with those of the infected patients using Pulsed-Field Gel Electrophoresis (PFGE). Rates of MRSA among the companion animals of MRSA-infected patients were compared to rates of MRSA among companion animals of pet guardians attending a “veterinary wellness clinic” (controls). MRSA was isolated from at least one companion animal in 4/49 (8.2%) households of MRSA-infected outpatients vs. none of the pets of the 50 uninfected human controls. Using PFGE, patient-pets MRSA isolates were identical for three pairs and discordant for one pair (suggested MRSA inter-specie transmission p-value = 0.1175). These results suggest that companion animals of MRSA-infected patients can be culture-positive for MRSA, representing a potential source of infection or re-infection for humans. Further studies are required to better understand the epidemiology of MRSA human-animal inter-specie transmission

    Genetic Diversity and Population Parameters of Sea Otters, Enhydra lutris, before Fur Trade Extirpation from 1741–1911

    All existing sea otter, Enhydra lutris, populations have suffered at least one historic population bottleneck stemming from the fur trade extirpations of the eighteenth and nineteenth centuries. We examined genetic variation, gene flow, and population structure at five microsatellite loci in samples from five pre-fur trade populations throughout the sea otter's historical range: California, Oregon, Washington, Alaska, and Russia. We then compared those values to genetic diversity and population structure found within five modern sea otter populations throughout their current range: California, Prince William Sound, Amchitka Island, Southeast Alaska and Washington. We found twice the genetic diversity in the pre-fur trade populations when compared to modern sea otters, a level of diversity that was similar to levels that are found in other mammal populations that have not experienced population bottlenecks. Even with the significant loss in genetic diversity modern sea otters have retained historical structure. There was greater gene flow before extirpation than that found among modern sea otter populations but the difference was not statistically significant. The most dramatic effect of pre fur trade population extirpation was the loss of genetic diversity. For long term conservation of these populations increasing gene flow and the maintenance of remnant genetic diversity should be encouraged

    Unveiling the enigma of ATLAS17aeu

    Aims: The unusual transient ATLAS17aeu was serendipitously detected within the sky localisation of the gravitational wave trigger GW 170104. The importance of a possible association with gravitational waves coming from a binary black hole merger led to an extensive follow-up campaign, with the aim of assessing a possible connection with GW 170104. Methods: With several telescopes, we carried out both photometric and spectroscopic observations of ATLAS17aeu, for several epochs, between ∼3 and ∼230 days after the first detection. Results: We studied in detail the temporal and spectroscopic properties of ATLAS17aeu and its host galaxy. Although at low significance and not conclusive, we found similarities to the spectral features of a broad-line supernova superposed onto an otherwise typical long-GRB afterglow. Based on analysis of the optical light curve, spectrum, and host galaxy spectral energy distribution, we conclude that the redshift of the source is probably z ≃ 0.5 ± 0.2. Conclusions: While the redshift range we have determined is marginally compatible with that of the gravitational wave event, the presence of a supernova component and the consistency of this transient with the Ep-Eiso correlation support the conclusion that ATLAS17aeu was associated with the long gamma-ray burst GRB 170105A. This rules out the association of the GRB 170105A/ATLAS17aeu transient with the gravitational wave event GW 170104, which was due to a binary black hole merger.
