    The role of autophagy in osteoclast differentiation and bone resorption function

    Autophagy is an evolutionary conserved and highly regulated recycling process of cellular wastes. Having a housekeeping role, autophagy through the digestion of domestic cytosolic organelles, proteins, macromolecules, and pathogens, eliminates unnecessary materials and provides nutrients and energy for cell survival and maintenance. The critical role of autophagy and autophagy-related proteins in osteoclast differentiation, bone resorption, and maintenance of bone homeostasis has previously been reported. Increasing evidence reveals that autophagy dysregulation leads to alteration of osteoclast function and enhanced bone loss, which is associated with the onset and progression of osteoporosis. In this review, we briefly consolidate the current state-of-the-art technology regarding the role of autophagy in osteoclast function in both physiologic and pathologic conditions to have a more general view on this issue

    Probabilistic classification models for the in situ authentication of iberian pig carcasses using near infrared spectroscopy

    Iberian pig ham is one of several high value European food products that are the subject of significant attempts at fraud because of the high price differences between commercial categories. Iberian pig products are classified by the Spanish regulations into different categories, mainly depending on the feeding regime during the fattening phase and the race involved, being of Premium quality those products obtained from the animals fed with acorns and other natural resources. Most of the previous NIRS studies related to the Iberian pig have involved the use of at-line instruments to predict quantitative quality parameters. This paper explores the use of the NIR spectra (369 for training and 199 for validation) to classify samples according to the categories Premium (animals fed with acorn) and Non Premium (animals fed with compound feeds), using a MicroNIRℱ Pro1700 microspectrometer to analyse individual carcasses in situ at the slaughterhouse line. Four discriminant methods were explored: linear discriminant analysis (LDA), quadratic discriminant analysis (QDA), Kernel Bayes and Logistic Regression. These are all discriminant methods that naturally produce classification probabilities to quantify the uncertainty of the results. Rules were tuned and methods compared using both classification error rates and a probability scoring rule. LDA gave the best results, attaining an overall accuracy of 93% and providing well-calibrated classification probabilities

    The hidden value of nontimber forest products: income contribution of the Basilicata wild truffle

    The Basilicata region (South of Italy) is land of truffles where the gastronomic, economic and cultural awareness has developed for this non-timber forest product only in the past decade. Little is known about truffle production and its social, economic and environmental implications. In this article we investigate the Basilicata truffle sector by devoting particular attention to the truffle hunters who gather the truffles from the forests. The data for the analysis were collected through a survey with the aim of describing the gathering activity (people involved, specie and quantities collected, etc.) and assessing its significance as a source of income. Results show that truffles can provide local communities with earning opportunities. However, the truffle sector needs to be protected and enhanced

    Stem-like and highly invasive prostate cancer cells expressing CD44v8-10 marker originate from CD44-negative cells

    In human prostate cancer (PCa), the neuroendocrine cells, expressing the prostate cancer stem cell (CSC) marker CD44, may be resistant to androgen ablation and promote tumor recurrence. During the study of heterogeneity of the highly aggressive neuroendocrine PCa cell lines PC3 and DU-145, we isolated and expanded in vitro a minor subpopulation of very small cells lacking CD44 (CD44neg). Unexpectedly, these sorted CD44neg cells rapidly and spontaneously converted to a stable CD44high phenotype specifically expressing the CD44v8-10 isoform which the sorted CD44high subpopulation failed to express. Surprisingly and potentially interesting, in these cells expression of CD44v8-10 was found to be induced in stem cell medium. CD44 variant isoforms are known to be more expressed in CSC and metastatic cells than CD44 standard isoform. In agreement, functional analysis of the two sorted and cultured subpopulations has shown that the CD44v8-10pos PC3 cells, resulting from the conversion of the CD44neg subpopulation, were more invasive in vitro and had a higher clonogenic potential than the sorted CD44high cells, in that they produced mainly holoclones, known to be enriched in stem-like cells. Of interest, the CD44v8-10 is more expressed in human PCa biopsies than in normal gland. The discovery of CD44v8-10pos cells with stem-like and invasive features, derived from a minoritarian CD44neg cell population in PCa, alerts on the high plasticity of stem-like markers and urges for prudency on the approaches to targeting the putative CSC

    Acorn Iberian ham (JamĂłn IbĂ©rico de Bellota) is one of the most expensive luxury foodstuffs produced in Europe, with a highly appreciated smell and flavour. Its recognized high-sensorial quality and health properties are mainly due to the traditional outdoor feeding system (Montanera) of Iberian pigs (IP), which provides high standards of animal welfare. Nowadays, one of the frauds affecting this product is the use of “special compound feeds” to simulate the fat composition of the acorns through the inclusion of sources of oleic acid like the ones found in pigs fed outdoors. The high prices paid for a cured leg of Iberian ham –ranging from hundreds to thousands of euros- leads to many opportunities for mislabelling and fraud. Fatty acid content of the adipose tissue could provide evidence of the feeding system. Gas chromatography (GC) is used at industry level for production control purposes. However, it is costly and time-consuming, and it is only applied to batches of animals rather than individual pigs. The main goal of this study was to use spectra belonging to a portable NIRS instrument (MicroNIR Onsite Lite, Viavi Solutions Inc.) for on–site quantitative (fatty acid content) analysis of individual Iberian pork carcasses at the slaughterhouse. Performance of this portable instrument was compared with an at-line NIRS monochromator. PLS models were built and optimized resulting in standard errors of cross validation ranging from 0.83 to 0.84 for palmitic acid, 0.94 to 0.99 for stearic acid, 1.47 to 1.56 for oleic acid and 0.53 to 0.58 for linoleic acid

    Short Communication: The potential of portable near infrared spectroscopy for assuring quality and authenticity in the food chain, using Iberian hams as an example

    This communication assesses the use of a portable near infrared (NIR) instrument to measure quantitative (fatty acid profile) properties and qualitative (‘Premium’ and ‘Non-premium’) categories of individual Iberian pork carcasses at the slaughterhouse. Acorn-fed Iberian pigs have more unsaturated fats than pigs fed conventional compound feed. Recent advances in miniaturisation have led to a number of handheld NIR devices being developed, allowing processing decisions to be made earlier, significantly reducing time and costs. The most common methods used for assessing quality and authenticity of Iberian hams are analysis of the fatty acid composition of subcutaneous fat using gas chromatography and DNA analysis. In this study, NIR calibrations for fatty acids and classification as premium or non-premium ham, based on carcass fat measured in situ, were developed using a portable NIR spectrometer. The accuracy of the quantitative equations was evaluated through the standard error of cross validation or standard error of prediction of 0.84 for palmitic acid (C16:0), 0.94 for stearic acid (C18:0), 1.47 for oleic acid (C18:1) and 0.58 for linoleic acid (C18:2). Qualitative calibrations provided acceptable results, with up to 98% of samples (n = 234) correctly classified with probabilities â©Ÿ0.9. Results indicated a portable NIR instrument has the potential to be used to measure quality and authenticity of Iberian pork carcasses

    Toll-iike receptor-3 activation enhances malignant traits in human breast cancer cells through hypoxia-inducible factor-1α

    Background/Aim: Hypoxia-inducible factor 1 (HIF1) inhibitors have been proposed as therapeutic agents for several tumor types. HIF1α is induced by hypoxia and by pathogens in normoxia through toll-like receptors (TLRs). The TLR3 activator polyinosinic:polycytidylic acid [poly(I:C)] induces apoptosis in various types of cancer but not in the most aggressive breast cancer cell lines. We hypothesized that the failure of TLR3 stimulation to induce apoptosis in these cells might be due to an elevated HIF1α level and this link might be exploited. Materials and Methods: Poly(I:C)-induced signaling pathway and expression of HIF1α and HIF1α targets were studied in MDA MB-231 and MCF-7 breast cancer cell lines by western blot. Flow cytometry was used for apoptotic responses and vasculogenic mimicry as bioassay. Results: Poly(I:C) increased expression of HIF1α and its targets BCL2 apoptosis regulator and c-MYC. Moreover, using pharmacological or genetic HIF1 inhibition, reduction of poly(I:C)-induced expression of HIF1α was paralleled by lowering of c-MYC and increased sensitivity to poly(I:C)-induced apoptosis, demonstrating the crucial role of this factor. We provide the first evidence in breast cancer cells that TLR3 stimulation induces HIF1αdependent vasculogenic mimicry. By using specific inhibitors, we identified a signaling cascade upstream of HIF1α induction. Conclusion: Combined treatment with poly(I:C) and HIF1 inhibitors deserves consideration as an effective strategy in breast cancer therapy

    Thinning in black pine (Pinus nigra J.F. Arnold) forests: The economic sustainability of the wood-energy supply chain in a case study in Italy

    In Italy, black pine has been largely used in reforestation projects in the past. Most of these reforestations are characterized by a high instability, vulnerability, and a limited resistance to atmospheric agents. In this situation, it is crucial to define silvicultural interventions able to increase the ecological stability of black pine stands and at the same time to guarantee the economic sustainability of the wood products obtained. Thinning in black pine forests can provide wood material for energy use. The main aim of the present study was to investigate the economic sustainability of a local wood-energy supply chain applying three different forest management options. The case study was Monte Morello forest, a degraded black pine forest located in Central Italy. The results show that the long-term economic sustainability of the wood-energy supply chain is ensured only when the use of bio-fuel is characterized by high energy efficiency. In addition, the results show that public contributions are fundamental to ensure that silvicultural interventions are realized with a positive economic balance and that to surmount this situation many loggings companies are organizing. Finally, the results highlighted the importance of the quantities of thermal energy sold to ensure the economic and environmental efficiency of the wood-energy supply chain

    Urban consumer trust and food certifications in China

    China has experienced frequent food safety incidents that have undermined consumer trust in the food supply chain. To overcome this problem, China requalified the legislative framework and adopted a comprehensive food certification system over the years. Here, we investigated the influences of food traceability and Chinese certifications (QS/SC—food quality safety market access/production system, hazard-free, green, and organic) on Chinese consumer trust of food safety for different types of products: fish, meat, milk, eggs, and rice. Data were collected through face-to-face surveys conducted in rural and urban Chinese areas. With a sample of 757 questionnaires, we ran a logit model. The results show consumers’ uncertainty and skepticism of certifications guaranteeing food safety attributes, especially for animal-based products. We found that price is used as a cue of safety by Chinese consumers. Individuals with higher education seem less influenced by certifications and other cues included in the analysis. The findings demonstrate that Chinese policy makers should implement new strategies to enhance consumer food safety trust, and design policies by considering different categories (e.g., vegetables, meat, fish, etc.) of food
