    Heat stress affects tassel development and reduces the kernel number of summer maize

    Maize grain yield is drastically reduced by heat stress (HTS) during anthesis and early grain filling. However, the mechanism of HTS in reproductive organs and kernel numbers remains poorly understood. From 2018 to 2020, two maize varieties (ND372, heat tolerant; and XY335, heat sensitive) and two temperature regimens (HTS, heat stress; and CK, natural control) were evaluated, resulting in four treatments (372CK, 372HTS, 335CK, and 335HTS). HTS was applied from the nine-leaf stage (V9) to the anthesis stage. Various morphological traits and physiological activities of the tassels, anthers, and pollen from the two varieties were evaluated to determine their correlation with kernel count. The results showed that HTS reduced the number of florets, tassel volume, and tassel length, but increased the number of tassel branches. HTS accelerates tassel degradation and reduces pollen weight, quantity, and viability. Deformation and reduction in length and volume due to HTS were observed in both the Nongda 372 (ND372) and Xianyu 335 (XY335) varieties, with the average reductions being 22.9% and 35.2%, respectively. The morphology of the anthers changed more conspicuously in XY335 maize. The number of kernels per spike was reduced in the HTS group compared with the CK group, with the ND372 and XY335 varieties showing reductions of 47.3% and 59.3%, respectively. The main factors underlying the decrease in yield caused by HTS were reductions in pollen quantity and weight, tassel rachis, and branch length. HTS had a greater effect on the anther shape, pollen viability, and phenotype of XY335 than on those of ND372. HTS had a greater impact on anther morphology, pollen viability, and the phenotype of XY335 but had no influence on the appearance or dissemination of pollen from tassel

    Effect of Sintering Temperature, Sm3+ Concentration and Excitation Wavelength on Luminescence Properties in Eu3+ Doped Strontium Tungstate Phosphors

    The color-tunable SrWO4:0.20Eu3+, Sm3+ phosphors are successfully prepared by co-precipitation method at a low temperature (800 °C). These phosphors can be efficiently excited by near-ultraviolet and blue light. The phase formation, luminescence properties, energy transfer between Eu3+ and Sm3+, and the critical distance were studied. The Sm3+ ions as the sensitizer could extend the excitation spectrum and enhance emission. The CIE chromaticity coordinate was also presented. The emission hues covered the regions from pink, orange, to reddish-orange, and eventually to red, which can be controlled by adjusting excitation energy, calcination temperature, and doping rare earth ions ratio via the energy transfer. The high efficiency and high color purity red emitting SrWO4:0.20Eu3+, xSm3+ (x = 0.005 ¸ 0.01) phosphors have higher color saturation than the commercially available Y2O2S:Eu3+ red phosphor, which is in coincidence with the National Television Standard Committee system standard for red chromaticity (0.67, 0.33). The obtained phosphors exhibit an excellent light emitting efficiency, color-purity and lower correlated color temperature of the comfortable warm white LEDs.</p

    BmILF and I-motif Structure Are Involved in Transcriptional Regulation of \u3cem\u3eBmPOUM2\u3c/em\u3e in \u3cem\u3eBombyx mori\u3c/em\u3e

    Guanine-rich and cytosine-rich DNA can form four-stranded DNA secondary structures called G-quadruplex (G4) and i-motif, respectively. These structures widely exist in genomes and play important roles in transcription, replication, translation and protection of telomeres. In this study, G4 and i-motif structures were identified in the promoter of the transcription factor gene BmPOUM2, which regulates the expression of the wing disc cuticle protein gene (BmWCP4) during metamorphosis. Disruption of the i-motif structure by base mutation, anti-sense oligonucleotides (ASOs) or inhibitory ligands resulted in significant decrease in the activity of the BmPOUM2 promoter. A novel i-motif binding protein (BmILF) was identified by pull-down experiment. BmILF specifically bound to the i-motif and activated the transcription of BmPOUM2. The promoter activity of BmPOUM2 was enhanced when BmILF was over-expressed and decreased when BmILF was knocked-down by RNA interference. This study for the first time demonstrated that BmILF and the i-motif structure participated in the regulation of gene transcription in insect metamorphosis and provides new insights into the molecular mechanism of the secondary structures in epigenetic regulation of gene transcription

    Derivedness index for estimating degree of phenotypic evolution of embryos: a study of comparative transcriptomic analyses of chordates and echinoderms

    Species retaining ancestral features, such as species called living fossils, are often regarded as less derived than their sister groups, but such discussions are usually based on qualitative enumeration of conserved traits. This approach creates a major barrier, especially when quantifying the degree of phenotypic evolution or degree of derivedness, since it focuses only on commonly shared traits, and newly acquired or lost traits are often overlooked. To provide a potential solution to this problem, especially for inter-species comparison of gene expression profiles, we propose a new method named "derivedness index" to quantify the degree of derivedness. In contrast to the conservation-based approach, which deals with expressions of commonly shared genes among species being compared, the derivedness index also considers those that were potentially lost or duplicated during evolution. By applying our method, we found that the gene expression profiles of penta-radial phases in echinoderm tended to be more highly derived than those of the bilateral phase. However, our results suggest that echinoderms may not have experienced much larger modifications to their developmental systems than chordates, at least at the transcriptomic level. In vertebrates, we found that the mid-embryonic and organogenesis stages were generally less derived than the earlier or later stages, indicating that the conserved phylotypic period is also less derived. We also found genes that potentially explain less derivedness, such as Hox genes. Finally, we highlight technical concerns that may influence the measured transcriptomic derivedness, such as read depth and library preparation protocols, for further improvement of our method through future studies. We anticipate that this index will serve as a quantitative guide in the search for constrained developmental phases or processes.Published versio

    Pre-anesthetic use of butorphanol for the prevention of emergence agitation in thoracic surgery: A multicenter, randomized controlled trial

    BackgroundEmergence agitation (EA) is common in patients after general anesthesia (GA) and is associated with poor outcomes. Patients with thoracic surgery have a higher incidence of EA compared with other surgery. This study aimed to investigate the impact of pre-anesthetic butorphanol infusion on the incidence of EA in patients undergoing thoracic surgery with GA.Materials and methodsThis prospective randomized controlled trial (RCT) was conducted in 20 tertiary hospitals in China. A total of 668 patients undergoing elective video-assisted thoracoscopic lobectomy/segmentectomy for lung cancer were assessed for eligibility, and 620 patients were enrolled. In total, 296 patients who received butorphanol and 306 control patients were included in the intention-to-treat analysis. Patients in the intervention group received butorphanol 0.02 mg/kg 15 min before induction of anesthesia. Patients in the control group received volume-matched normal saline in the same schedule. The primary outcome was the incidence of EA after 5 min of extubation, and EA was evaluated using the Riker Sedation-Agitation Scale (RSAS). The incidence of EA was determined by the chi-square test, with a significance of P &lt; 0.05.ResultsIn total, 296 patients who received butorphanol and 306 control patients were included in the intention-to-treat analysis. The incidence of EA 5 min after extubation was lower with butorphanol treatment: 9.8% (29 of 296) vs. 24.5% (75 of 306) in the control group (P = 0.0001). Patients who received butorphanol had a lower incidence of drug-related complications (including injecting propofol pain and coughing with sufentanil): 112 of 296 vs. 199 of 306 in the control group (P = 0.001) and 3 of 296 vs. 35 of 306 in the control group (P = 0.0001).ConclusionThe pre-anesthetic administration of butorphanol reduced the incidence of EA after thoracic surgery under GA.Clinical trial registration[http://www.chictr.org.cn/showproj.aspx?proj=42684], identifier [ChiCTR1900025705]

    Goss Texture Evolution of Grain Oriented Silicon Steel by High-Energy X-ray Diffraction

    International audienceHigh energy synchrotron diffraction offers great potential to study the recrystallization kinetics of metallic materials. To study the formation of Goss texture (\110\) of grain oriented (GO) silicon steel during secondary recrystallization process, an in situ experiment using high energy X-ray diffraction was designed. The results showed that the secondary recrystallization began when the heating temperature was 1,494 K, and the grains grew rapidly above this temperature. With an increase in annealing temperature, the large grains with gamma orientation [//normal direction] formed and gradually occupied the dominant position. As the annealing temperature increased even further, the grains with Goss orientation to a very large size by devouring the gamma orientation grains that formed in the early annealing stage. A single crystal with a Goss orientation was observed in the GO silicon steel when the annealing temperature was 1,540 K
