    Robust model calibration of a wind turbine with load uncertainties

    International audienceThe goal of this work is to explore a model calibration strategy for an industrial problem consisting in a MW class geared wind turbine power train subjected to uncertain loads. Lack of knowledge is commonplace in this kind of engineering system and a realistic model calibration cannot be performed without taking into account this type of uncertainty. The question at stake in this study is how to perform a more robust model of a dynamic system given that the excitations are poorly known. The uncertainty in the latter will be represented with an info-gap model. This methodology is illustrated on a Megawat class wind turbine power train torsional model

    A simple, low-cost conductive composite material for 3D printing of electronic sensors

    3D printing technology can produce complex objects directly from computer aided digital designs. The technology has traditionally been used by large companies to produce fit and form concept prototypes (‘rapid prototyping’) before production. In recent years however there has been a move to adopt the technology as full-scale manufacturing solution. The advent of low-cost, desktop 3D printers such as the RepRap and Fab@Home has meant a wider user base are now able to have access to desktop manufacturing platforms enabling them to produce highly customised products for personal use and sale. This uptake in usage has been coupled with a demand for printing technology and materials able to print functional elements such as electronic sensors. Here we present formulation of a simple conductive thermoplastic composite we term ‘carbomorph’ and demonstrate how it can be used in an unmodified low-cost 3D printer to print electronic sensors able to sense mechanical flexing and capacitance changes. We show how this capability can be used to produce custom sensing devices and user interface devices along with printed objects with embedded sensing capability. This advance in low-cost 3D printing with offer a new paradigm in the 3D printing field with printed sensors and electronics embedded inside 3D printed objects in a single build process without requiring complex or expensive materials incorporating additives such as carbon nanotubes

    Identifying and explaining the farming system composition of agricultural landscapes: The role of socioeconomic drivers under strong biophysical gradients

    In mountain landscapes, agricultural abandonment is taking place in the most vulnerable areas, while intensification increases in the most productive lands. These contrasting processes, which have different impacts on biodiversity and ecosystem services (BES), are related to changes in the farming system component of these landscapes. Farming systems are identified based on farmer’s decisions on, for example, type of crop and level of fertilizers, which represent the descriptors of farming systems and can be grouped into several dimensions (e.g. land use and intensity). Since obtaining this data at farm-level is often difficult, an alternative is to study the spatial combinations of farming systems at parish-level, i.e., Farming System Mixes (FSM), relying on agricultural census data. Other biophysical (e.g. climate, soil) and socioeconomic (e.g. labour, farmer’s age) variables, independent of farmers' decisions, represent the exogenous drivers of these decisions. The separation between descriptors and drivers is important to improve knowledge about what drives farmers' decisions regarding farming system choice, as these choices are often the focus of policies aiming the support of BES. In this study, we explored the underlying drivers of FSM and assessed the role of socioeconomic drivers, main target for policy makers, in a context of strong biophysical gradients. Biophysical drivers emerge as those that primarily discriminate between the FSM located in different topographic positions (valleys, mountains and plateau).     Crystallographic and Molecular Dynamics Analysis of Loop Motions Unmasking the Peptidoglycan-Binding Site in Stator Protein MotB of Flagellar Motor

    Background: The C-terminal domain of MotB (MotB-C) shows high sequence similarity to outer membrane protein A and related peptidoglycan (PG)-binding proteins. It is believed to anchor the power-generating MotA/MotB stator unit of the bacterial flagellar motor to the peptidoglycan layer of the cell wall. We previously reported the first crystal structure of this domain and made a puzzling observation that all conserved residues that are thought to be essential for PG recognition are buried and inaccessible in the crystal structure. In this study, we tested a hypothesis that peptidoglycan binding is preceded by, or accompanied by, some structural reorganization that exposes the key conserved residues. Methodology/Principal Findings: We determined the structure of a new crystalline form (Form B) of Helicobacter pylori MotB-C. Comparisons with the existing Form A revealed conformational variations in the petal-like loops around the carbohydrate binding site near one end of the b-sheet. These variations are thought to reflect natural flexibility at this site required for insertion into the peptidoglycan mesh. In order to understand the nature of this flexibility we have performed molecular dynamics simulations of the MotB-C dimer. The results are consistent with the crystallographic data and provide evidence that the three loops move in a concerted fashion, exposing conserved MotB residues that have previously been implicated in binding of the peptide moiety of peptidoglycan.
