Continuidad y cambios en la vida cotidiana de familias indígenas p’urhépechas de la zmg
Este capítulo examina, desde una perspectiva histórica–cultural siguiendo
el paradigma de Aprendizaje por Medio de la Observación y la Participación (lopi)
(Rogoff, 2014), los procesos de continuidad y de adaptación de un grupo de familias indígenas
asentadas en la Zona Metropolitana de Guadalajara (zmg). El análisis cualitativo
de entrevistas con padres de familia y niños p’urhépechas residentes de la colonia La
Noria, así como las observaciones etnográficas realizadas, reflejan la complejidad de las
experiencias de estas familias mientras se adaptan al contexto urbano. En general, las
madres de familia reconocen beneficios centrales de migrar a la zmg como el acceso a
educación y al empleo, y también señalan las dificultades económicas y otros factores que
les afectan en su identidad cultural y en el desarrollo de los hijos. Se identifican varios
aspectos tanto de continuidad como trasformaciones en algunas prácticas y tradiciones
culturales, en particular en la organización social comunitaria con el sistema de mayordomía; y a nivel intrafamiliar en la organización de la interacción familiar que refleja
prácticas indígenas que apoyan a la vez la autonomía de los niños y su contribución a las
actividades de la familia con aprendizajes por medio de la observación y participación.Se discuten las implicaciones con relación al papel de las instituciones y su escasa labor

Long-term etanercept survival in patients with psoriatic arthritis: a multicenter retrospective analysis in daily clinical practice in Spain
Although several randomized clinical trials and observational studies have evaluated the effectiveness, safety and drug survival of etanercept (ETN) in the treatment of psoriatic arthritis (PsA), long-term data regarding these aspects are currently scarce. For this reason, we sought to investigate the long-term survival and safety of ETN in PsA patients in 4 tertiary care Spanish hospitals over a 13-year observation period (from 2004 to 2017). The records of 85 PsA patients were reviewed. ETN showed an excellent survival profile, with rates of treatment discontinuation at 1, 3, 5 and 10 years of 15, 37, 46 and 59%, respectively. In our cohort, a trend toward longer drug survival in patients with shorter disease duration and those who were treated with ETN as their first biologic agent was observed. On the other hand, combination therapy with conventional disease-modifying antirheumatic drugs did not provide greater improvement on the long-term drug survival. Only 12% of the patients reported adverse events (AEs) during therapy, being most of them of mild to moderate intensity, and in only 7% AEs led to drug discontinuation. To the best of our knowledge, the present study shows the largest follow-up period of ETN-treated population analyzed in a real-life setting, and these results demonstrate the positive safety profile and long-term effectiveness of this biologic agent in the management of PsA patients
An Aptamer against MNK1 for Non-Small Cell Lung Cancer Treatment
Lung cancer is the leading cause of cancer-related death worldwide. Its late diagnosis and consequently poor survival make necessary the search for new therapeutic targets. The mitogen-activated protein kinase (MAPK)-interacting kinase 1 (MNK1) is overexpressed in lung cancer and correlates with poor overall survival in non-small cell lung cancer (NSCLC) patients. The previously identified and optimized aptamer from our laboratory against MNK1, apMNKQ2, showed promising results as an antitumor drug in breast cancer in vitro and in vivo. Thus, the present study shows the antitumor potential of apMNKQ2 in another type of cancer where MNK1 plays a significant role, such as NSCLC. The effect of apMNKQ2 in lung cancer was studied with viability, toxicity, clonogenic, migration, invasion, and in vivo efficacy assays. Our results show that apMNKQ2 arrests the cell cycle and reduces viability, colony formation, migration, invasion, and epithelial-mesenchymal transition (EMT) processes in NSCLC cells. In addition, apMNKQ2 reduces tumor growth in an A549-cell line NSCLC xenograft model. Incidence, Clinical Characteristics and Management of Inflammatory Bowel Disease in Spain : Large-Scale Epidemiological Study
(1) Aims: To assess the incidence of inflammatory bowel disease (IBD) in Spain, to describe the main epidemiological and clinical characteristics at diagnosis and the evolution of the disease, and to explore the use of drug treatments. (2) Methods: Prospective, population-based nationwide registry. Adult patients diagnosed with IBD-Crohn's disease (CD), ulcerative colitis (UC) or IBD unclassified (IBD-U)-during 2017 in Spain were included and were followed-up for 1 year. (3) Results: We identified 3611 incident cases of IBD diagnosed during 2017 in 108 hospitals covering over 22 million inhabitants. The overall incidence (cases/100,000 person-years) was 16 for IBD, 7.5 for CD, 8 for UC, and 0.5 for IBD-U; 53% of patients were male and median age was 43 years (interquartile range = 31-56 years). During a median 12-month follow-up, 34% of patients were treated with systemic steroids, 25% with immunomodulators, 15% with biologics and 5.6% underwent surgery. The percentage of patients under these treatments was significantly higher in CD than UC and IBD-U. Use of systemic steroids and biologics was significantly higher in hospitals with high resources. In total, 28% of patients were hospitalized (35% CD and 22% UC patients, p < 0.01). (4) Conclusion: The incidence of IBD in Spain is rather high and similar to that reported in Northern Europe. IBD patients require substantial therapeutic resources, which are greater in CD and in hospitals with high resources, and much higher than previously reported. One third of patients are hospitalized in the first year after diagnosis and a relevant proportion undergo surgery

Clonal chromosomal mosaicism and loss of chromosome Y in elderly men increase vulnerability for SARS-CoV-2
The pandemic caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2, COVID-19) had an estimated overall case fatality ratio of 1.38% (pre-vaccination), being 53% higher in males and increasing exponentially with age. Among 9578 individuals diagnosed with COVID-19 in the SCOURGE study, we found 133 cases (1.42%) with detectable clonal mosaicism for chromosome alterations (mCA) and 226 males (5.08%) with acquired loss of chromosome Y (LOY). Individuals with clonal mosaic events (mCA and/or LOY) showed a 54% increase in the risk of COVID-19 lethality. LOY is associated with transcriptomic biomarkers of immune dysfunction, pro-coagulation activity and cardiovascular risk. Interferon-induced genes involved in the initial immune response to SARS-CoV-2 are also down-regulated in LOY. Thus, mCA and LOY underlie at least part of the sex-biased severity and mortality of COVID-19 in aging patients. Given its potential therapeutic and prognostic relevance, evaluation of clonal mosaicism should be implemented as biomarker of COVID-19 severity in elderly people. El discurso de los gigantes corporativos de la minería recurren a falsear la realidad sobre los efectos de sus procesos productivos sobre cualquier expresión de vida, principalmente por la contaminación dinámica, pues se contaminan cuerpos de agua, tierras por dispersión de polvo por los vientos, se colapsa el medioambiente y con ello toda expresión de vida y tejido comunitario. Mientras esto sucede, el discurso oficial y empresarial se obstina en reiterar que todo está bajo control mientras las comunidades continúan padeciendo el desastre
The archaeology of the Middle Pleistocene deposits of Lake Eyasi, Tanzania
Ongoing archaeological research at North Lake Eyasi has produced a wealth of information, including a new hominid fossil and several archaeological sites dating to the end of the Middle Pleistocene. One of the sites (WB9) has been excavated and has produced evidence of multiple processes in its formation, including evidence of functional associations of stone tools and faunal remains which are scarce for this time period. The stone tool industry is based on a core and flake industry, which is not very diagnostic and attributed to MSA. Earlier heavy-duty tools classified as Sangoan may derive from the underlying Eyasi Beds. The stratigraphic provenience of previous fossil hominids is unknown. Surface collections from the Eyasi lake, thus, comprise two different sets of stone tools and fossils, which can only be clearly differentiated in the field. This advises against the use of previously curated collections as a homogeneous sample. Earlier definitions of the Njarasa industry should be revised. This work presents results on the paleoecology of the area and of its paleontological and archaeological information, with special reference to the excavation of WB9, the most complete site discovered in the area so far. This contributes to the limited information available about site functionality and hominid subsistential behaviour in East Africa during the end of the Middle Pleistocene. A technological study from WB9 also shows the variability of stone tool traditions at this time.Peer Reviewe
Comparative Transcriptome Analysis of <i>Babesia bigemina</i> Attenuated Vaccine and Virulent Strains of Mexican Origin
Bovine babesiosis, caused by the protozoan Babesia bigemina, is one of the most important hemoparasite diseases of cattle in Mexico and the world. An attenuated B. bigemina strain maintained under in vitro culture conditions has been used as a live attenuated vaccine; however, the biological mechanisms involved in attenuation are unknown. The objective of this study was to identify, through a comparative transcriptomics approach, the components of the B. bigemina virulent parasites that are differentially expressed in vivo, as opposed to those expressed by B. bigemina attenuated vaccine parasites when inoculated into naïve cattle. The biological material under study was obtained by inoculating spleen-intact cattle with infected erythrocytes containing either the attenuated strain or a virulent field strain. After RNA extraction, transcriptomic analysis (RNA-seq) was performed, followed by bioinformatic Differential Expression (DE) analysis and Gene Ontology (GO) term enrichment. The high-throughput sequencing results obtained by analyzing three biological replicates for each parasite strain ranged from 9,504,000 to 9,656,000, and 13,400,000 to 15,750,000 reads for the B. bigemina attenuated and virulent strains, respectively. At least 519 differentially expressed genes were identified in the analyzed strains. In addition, GO analysis revealed both similarities and differences across the three categories: cellular components, biological processes, and molecular functions. The attenuated strain of B. bigemina derived from in vitro culture presents global transcriptomic changes when compared to the virulent strain. Moreover, the obtained data provide insights into the potential molecular mechanisms associated with the attenuation or pathogenicity of each analyzed strain, offering molecular markers that might be associated with virulence or potential vaccine candidates
