Development and characterization of 16 microsatellite markers for the Louisiana pine snake, Pituophis ruthveni, and two congeners of conservation concern
We isolated and characterized 16 microsatellite loci from the Louisiana pine snake, Pituophis ruthveni. Loci were screened in 24 individuals from locations throughout its distribution in Louisiana and Texas. The number of alleles per locus ranged from 4 to 12, observed heterozygosity ranged from 0.200 to 0.875, and the probability of identity ranged from 0.043 to 0.298. We examined cross-species amplification at these loci in P. catenifer (bullsnakes and gopher snakes) and P. melanoleucus (pine snakes). These new markers provide tools for examining the conservation genetics of this species complex. Louisiana pine snakes face numerous threats: population densities are extremely low and their natural habitat has been severely altered and fragmented. In southern Canada, P. catenifer is at the northern extreme of its range and limited by the availability of suitable over-wintering sites. Hence, for these two species reduction of heterozygosity, potential for inbreeding, and increased effects of genetic drift are all of considerable conservation concern
Using fish models to investigate the links between microbiome and social behaviour: the next step for translational microbiome research?
Recent research has revealed surprisingly important connections between animalsâ microbiome and social behaviour. Social interactions can affect the composition and function of the microbiome; conversely, the microbiome affects social communication by influencing the hostsâ central nervous system and peripheral chemical communication. These discoveries set the stage for novel research focusing on the evolution and physiology of animal social behaviour in relation to microbial transmission strategies. Here, we discuss the emerging roles of teleost fish models and their potential for advancing research fields, linked to sociality and microbial regulation. We argue that fish models, such as the zebrafish (Danio rerio, Cyprinidae), sticklebacks (âGasterosteidae), guppies (Poeciliidae) and cleanerâclient dyads (e.g., obligate cleaner fish from the Labridae and Gobiidae families and their visiting clientele), will provide valuable insights into the roles of microbiome in shaping social behaviour and vice versa, while also being of direct relevance to the food and ornamental fish trades. The diversity of fish behaviour warrants more interdisciplinary research, including microbiome studies, which should have a strong ecological (fieldâderived) approach, together with laboratoryâbased cognitive and neurobiological experimentation. The implications of such integrated approaches may be of translational relevance, opening new avenues for future investigation using fish models
Spatial ecology and multi-scale habitat selection by Western Painted Turtles (Chrysemys picta bellii) in an urban area
As urban centres expand, knowledge on the habitat and space use of native wildlife, particularly long-lived species, is required for proper management. Our objective was to understand space requirements and key habitat features necessary for long-term persistence of Western Painted Turtles (Chrysemys picta bellii) living in a Canadian urban park. Using radio telemetry, we examined seasonal habitat selection and space use over two years, 2015â2016 (n = 23), and 2016â2017 (n = 29) in Regina, Saskatchewan. Daily movements and home ranges of males and females were smaller during emergence than during nesting or post-nesting phases of the active season. Turtles inhabiting marsh sites had 2- and 4-times larger daily movements and home ranges compared to turtles inhabiting the creek. Turtles selected the shoreline habitat over urban/parkland and open water. Turtles used marsh-shoreline habitats non-randomly, selecting accessible shoreline with large trees in the active season. In contrast, turtles used creek-shoreline habitat according to availability. Overwintering sites selected by turtles were warmer and deeper than random available sites, with no difference in dissolved oxygen level. However, water was hypoxic for most overwintering sites. Our results show that turtles range widely, requiring 20â60 ha throughout the year. Urban park areas should be managed to provide accessible shorelines with a combination of cover and open basking areas. Critically, careful attention needs to be paid to managing water depth so that over-wintering sites remain viable
A report of unusual aggregation behaviour in Bullsnakes (Pituophis catenifer sayi) near a nest site in Saskatchewan
We describe an unusual aggregation of Bullsnakes (Pituophis catenifer sayi) near a nest site in Saskatchewan. Bullsnake is a wide-ranging oviparous colubrid that reaches the northern tip of its continental range on the prairies and badlands of Canada. At that location, it is considered a species of Special Concern, but, until recently, has been the subject of relatively few natural history reports. This is significant, because, at the northern edge of their range, Bullsnakes may behave differently than elsewhere due to thermal limitations experienced at high latitudes. On 29 June 2019, we observed a mass of five adult Bullsnakes in a Silver Sagebrush (Artemisia cana) shrub on a slope in southwestern Saskatchewan. Aggregations of Bullsnakes are known to occur at hibernacula, during mating, and inside nest chambers before and after oviposition. However, we are unaware of the occurrence of surface aggregations in any other situation. We suggest that these Bullsnakes may have been exhibiting communal gestation, a behaviour thought to be rare in oviparous snakes whereby gravid females congregate before parturition or oviposition for thermal stability or protection from predators
Proximity to Hibernacula and Road Type Influence Potential Road Mortality of Snakes in Southwestern Saskatchewan
The behaviour of snakes makes them vulnerable to road mortality, which may have conservation implications for some species. We examined factors affecting road mortality of snakes around Grasslands National Park of Canada (GNP), Saskatchewan, an area inhabited by a unique snake community within Canada, including the threatened Eastern Yellow-bellied Racer (Coluber constrictor flaviventris). Systematic surveys (n = 45) of roads in the Grasslands National Park area in 2009 yielded 36 dead and 18 live snakes. Multivariate modelling revealed that proximity to hibernacula was positively correlated with presence of snakes on roads. Paved roads had disproportionately higher numbers of snake mortalities, suggesting that traffic patterns are a bigger risk factor than road use per se. Some radio-tracked Eastern Yellow-bellied Racers (2 of 17; 12%) and Bullsnakes (Pituophis catenifer sayi) (4 of 5; 80%) captured at emergence from hibernacula had road areas in their home ranges. These individuals equipped with radio-transmitters used roads and immediately adjacent areas significantly more than expected, based on their availability, suggesting possible selection for roads. Strategies to reduce road mortality of snakes should focus on key stretches of roads, such as those near winter hibernacula or riparian zones. The placement of paved roads in sensitive areas like those in and around Grasslands National Park should be carefully considered to minimize snake mortality
Variable habitat selection and movement patterns among Bullsnake (Pituophis catenifer sayi) populations in Saskatchewan
Pituophis catenifer sayi (Bullsnake) is a sparsely studied subspecies of conservation concern in Canada. Basic ecological information is lacking for P. c. sayi, which reaches its northern range limit in western Canada. To address this gap, we used radio-telemetry to examine space use and habitat selection in three populations of Bullsnakes in disjunct river valley systems (Frenchman, Big Muddy, and South Saskatchewan River Valleys) across their Saskatchewan range. Bullsnakes in two valleys used up to three times more space, travelled 2.5-times farther from overwintering sites, and had lower home range overlap than the third population. Landscape-level habitat selection was flexible, with snakes in all populations using both natural and human-modified habitats most frequently. Fine-scale habitat selection was also similar among populations, with Bullsnakes selecting sites within 1 m of refuges, regardless of whether they were natural or anthropogenic. Based on these results, Bullsnakes are flexible in their broad scale habitat use, as long as they are provided with fine scale refuge sites. The distribution of key seasonal resources appears to ultimately determine space use and habitat selection by Bullsnakes, regardless of the geographic location of the population
Thermoregulation by bullsnakes (Pituophis catenifer sayi): do burrows make life easier on the prairies?
Reptiles living in the grasslands of Western Canada cope with extremely variable environmental temperatures. We studied body temperatures (Tb) and operative environmental temperatures (Te) for bullsnakes (Pituophis catenifer sayi Schlegel, 1837) to address uncertainties regarding challenges posed by the thermal environments of northern grasslands, and to evaluate conflicting hypotheses regarding thermoregulatory strategies. Despite potentially extreme surface temperatures (xÌmin = 9.3, xÌmax = 31.4C), mammal burrows remained within voluntary limits (15 â 35C) for 93 % of the active season and created thermal gradients (up to 27.2C) which may facilitate thermoregulation by creating high thermal heterogeneity. This evidence suggests that grasslands may actually be less challenging for thermoregulation than comparable forests. 57.8 % of the variation in observed Tb (xÌmin = 20.1, xÌmax = 29.1C) was explained by 3 simple variables: time of day, day of year, and bare ground Te. Bullsnakes thermoregulated by conforming to Te near their preferred range (21 to 27C), selecting locations that enabled heating up below this range, and avoiding warmth above this range. Our results support broad hypotheses of reptilian thermoregulation that predict increased thermoregulation when a) environmental temperatures deviate further from preferred ranges and b) costs of thermoregulation are lower due to thermal heterogeneity.The accepted manuscript in pdf format is listed with the files at the bottom of this page.
