    Optimization of sowing dates in Indian mustard (Brassica juncea L.) to combat yield losses caused by high temperature at reproductive stage

    A field experiment was undertaken during the Rabi seasons of 2019-20 and 2021-22 at organic farm of Amity Institute of Organic Agriculture, Amity University, Noida, Uttar Pradesh, India, for the optimization of the sowing time period to reduce the effect of heat stress on Indian mustard (Brassica juncea L.) for increasing productivity. 32 Indian mustard germplasms were sown at three different time periods viz., the last week of September, the last fortnight of October, and the second week of November. Our results revealed that the mustard crop showed a substantial increase in yield supporting characters such as the number of siliqua and seeds per siliqua when sown at last week of September as compared to the second week of October, whereas the late sown crop resulted in less yield due to the heat stress faced by crop during the anthesis stage as compared to early and optimum sowing. The mean seed yield per plant showed a positive correlation with mean test weight. We established the positive effect of early sowing over optimal and late sowing in Indian mustard; further, we found a few mustard accessions were able to resist different climatic conditions. The mustard germplasm accessions IC296688, IC296703, IC296732, IC305130, IC401575, IC426385, and IC589669 were high yielding among all the accessions and can be considered as heat stress tolerant

    Sugar and Polyphenolic Diversity in Floral Nectar of Cherry

    Cherries (Prunus avium L. and Prunus cerasus L.) are economically important fruit species in the temperate region. Both are entomophilous fruit species, thus need pollinators to give high yields. Since cherry’s flower is easy-to-reach, bees and other pollinators can smoothly collect nectar as a reward for doing transfer of pollen to receptive stigma. Nectar in cherry is usually attractive for insects, especially to honey bee (Apis mellifera) who is the most common pollinator. Nectar is predominantly an aqueous solution of sugars, proteins, and free amino acids among which sugars are the most dominant. Trace amounts of lipids, organic acids, iridoid glycosides, minerals, vitamins, alkaloids, plant hormones, non-protein amino, terpenoids, glucosinolates, and cardenolides can be found in nectar too. Cherry flower may secrete nectar for 2–4 days and, depending on the cultivar, produces up to 10 mg nectar with sugar concentration from 28% to 55%. Detailed chemical analysis of cherry nectar described in this chapter is focused on sugar and phenolic profile in sour cherry. The most abounded sugars in cherry nectar was fructose, glucose, and sucrose, while arabinose, rhamnose, maltose, isomaltose, trehalose, gentiobiose, turanose, panose, melezitose, maltotriose, isomaltotriose, as well as the sugar alcohols glycerol, erythritol, arabitol, galactitol, and mannitol are present as minor constituents. Regarding polyphenolics, rutin was the most abundant phenolic compound followed by naringenin and chrysin. Cherry cultivars showed different chemical composition of nectar which implies that its content is cultivar dependent

    In Vitro Cultures of Schisandra chinensis (Turcz.) Baill. (Chinese Magnolia Vine)—a Potential Biotechnological Rich Source of Therapeutically Important Phenolic Acids

    The contents of free phenolic acids and cinnamic acid were determined using an HPLC method in methanolic extracts from biomass of Schisandra chinensis (Turcz.) Baill. (Chinese magnolia vine) at different stages of organogenesis, cultured in vitro on a few variants of Murashige and Skoog (MS) medium, containing different concentrations of plant growth regulators 6-benzylaminopurine (BAP) and 1-naphthaleneacetic acid (NAA) (from 0.1 to 3.0 mg/l) and in extracts from overground parts of plants growing in vivo. Six of 12 analysed compounds were detected in all extracts: chlorogenic, p-coumaric, p-hydroxybenzoic, protocatechuic, salicylic and syringic acids. Total contents of the examined metabolites in biomass of shoot-differentiating callus culture cultivated on six MS medium variants were dependent on concentrations of growth regulators in the media and ranged from 14.90 to 60.05 mg/100 g d.w. Total contents of the compounds in biomass extracts from undifferentiating callus culture maintained only on two of six MS medium variants were higher and amounted to 74.54 and 78.24 mg/100 g d.w. Maximum total contents of phenolic acids in both types of in vitro cultures were greater than in fruits (55.73 mg/100 g d.w.) and leaves (4.55 mg/100 g d.w.) of plants gowning in vivo. Chlorogenic acid and salicylic acid were the main compounds identified in biomass extracts of shoot-differentiating callus cultures (max 22.60 and 21.17 mg/100 g d.w., respectively), while chlorogenic acid (max 38.43 mg/100 g d.w.) and protocatechuic acid (max 20.95 mg/100 g d.w.) prevailed in the extracts from undifferentiating callus cultures. Other compounds dominated in fruits, namely p-coumaric acid (23.36 mg/100 g d.w.) and syringic acid (14.96 mg/100 g d.w.). This is the first report on biochemical potential of cells from S. chinensis in vitro cultures to produce the biologically active phenolic acids. These are the first results on the analysis of this group of metabolites in overground parts of plants growing in vivo, too

    Effect of CO2 Enrichment on Synthesis of Some Primary and Secondary Metabolites in Ginger (Zingiber officinale Roscoe)

    The effect of two different CO2 concentrations (400 and 800 μmol mol−1) on the photosynthesis rate, primary and secondary metabolite syntheses and the antioxidant activities of the leaves, stems and rhizomes of two Zingiber officinale varieties (Halia Bentong and Halia Bara) were assessed in an effort to compare and validate the medicinal potential of the subterranean part of the young ginger. High photosynthesis rate (10.05 μmol CO2 m−2s−1 in Halia Bara) and plant biomass (83.4 g in Halia Bentong) were observed at 800 μmol mol−1 CO2. Stomatal conductance decreased and water use efficiency increased with elevated CO2 concentration. Total flavonoids (TF), total phenolics (TP), total soluble carbohydrates (TSC), starch and plant biomass increased significantly (P ≤ 0.05) in all parts of the ginger varieties under elevated CO2 (800 μmol mol−1). The order of the TF and TP increment in the parts of the plant was rhizomes > stems > leaves. More specifically, Halia Bara had a greater increase of TF (2.05 mg/g dry weight) and TP (14.31 mg/g dry weight) compared to Halia Bentong (TF: 1.42 mg/g dry weight; TP: 9.11 mg/g dry weight) in average over the whole plant. Furthermore, plants with the highest rate of photosynthesis had the highest TSC and phenolics content. Significant differences between treatments and species were observed for TF and TP production. Correlation coefficient showed that TSC and TP content are positively correlated in both varieties. The antioxidant activity, as determined by the ferric reducing/antioxidant potential (FRAP) activity, increased in young ginger grown under elevated CO2. The FRAP values for the leaves, rhizomes and stems extracts of both varieties grown under two different CO2 concentrations (400 and 800 μmol mol−1) were significantly lower than those of vitamin C (3107.28 μmol Fe (II)/g) and α-tocopherol (953 μmol Fe (II)/g), but higher than that of BHT (74.31 μmol Fe (II)/g). These results indicate that the plant biomass, primary and secondary metabolite synthesis, and following that, antioxidant activities of Malaysian young ginger varieties can be enhanced through controlled environment (CE) and CO2 enrichment

    378-382Cell cultures of Pueraria tuberosa were established in modified Murashige and Skoog medium and challenged with yeast extract (YE), methyl jasmonate (MeJA) and salicylic acid (SA). Maximum isoflavonoids production was recorded at 48 h of YA incorporation at the stationary phase. 20 μM of MeJA and SA was most effective in isoflavonoids induction. Higher concentrations of these elicitors were negatively correlated with the isoflavonoids production. YE at 150 mg L⁻¹ was optimal for isoflavonoids production, yielding 10 mg L⁻¹ isoflavonoids, which was ~20% higher over the yields at optimal concentrations of MeJA and SA. YA incorporation can be used as a trigger to induce high yield of isoflavonoids
