    Deformation Response of Some Earth and Rock-fill Dams

    Vertical compressional data obtained from 10 earth and earth rock fill darns built in India has been analysed for during construction and post-construction response. The performance of these darns has been compared with some darns built in USA wherein the construction practices have been similar. The contributions of various factors, namely height of embankment, gradation and plasticity of soil, and placement conditions of moisture and density have been examined. A major portion of the total compression occurred by the end of construction. These compressions were found to be the functions of the height of the embankment. Both the compressions across and along the valley have been found to be small

The proposed model also creates outlines of users and brands or items used by them. These outlines may not be useful only for this particular prediction task, but could also be used for other important tasks in e-commerce, such as client segmentation, product recommendation and client base growth for brands

    Control and estimation of errors are important but difficult aspects of any analysis from which the numerical results are necessarily approximate. The order of difficulty is greater for local or distributed quantities like stresses and displacements than for global Of integrated parameter~ like eigenvalues and stiffnesses. To really bound a desired quantity between a pair of dose upper and lower bounds one should obtain either an oscillatory but clear convergence or, preferably, two rapidly converging sequences one from above and the other from below. Application of the two complementary variational principles of energy and complimentary energy, when both are possible to apply, do yield upper and lower bound approximations. But these or other alternate methods are generally expensive. On the other hand it would be advantageous if one basic procedure could be perturbed in a simple manner to provide both lower and upper bounds and to refine the solution and control the errors without undue effort. This paper discusses this concept and presents three powerful methods to closely bound any desired parameter in a problem. These are particularly valuable for eigenvalue problems

    QRDR mutations, efflux system & antimicrobial resistance genes in enterotoxigenic Escherichia coli isolated from an outbreak of diarrhoea in Ahmedabad, India

    Background & objectives: Diverse mechanisms have been identified in enteric bacteria for their adaptation and survival against multiple classes of antimicrobial agents. Resistance of bacteria to the most effective fluoroquinolones have increasingly been reported in many countries. We have identified that most of the enterotoxigenic Escherichia coli (ETEC) were resistant to several antimicrobials in a diarrhoea outbreak at Ahmedabad during 2000. The present study was done to identify several genes responsible for antimicrobial resistance and mobile genetic elements in the ETEC strains. Methods: Seventeen ETEC strains isolated from diarrhoeal patients were included in this study. The antimicrobial resistance was confirmed by conventional disc diffusion method. PCR and DNA sequencing were performed for the identification of mutation in the quinolone resistance-determining regions (QRDRs). Efflux pump was tested by inhibiting the proton-motive force. DNA hybridization assay was made for the detection of integrase genes and the resistance gene cassettes were identified by direct sequencing of the PCR amplicons. Results: Majority of the ETEC had GyrA mutations at codons 83 and 87 and in ParC at codon 80. Six strains had an additional mutation in ParC at codon 108 and two had at position 84. Plasmid-borne qnr gene alleles that encode quinolone resistance were not detected but the newly described aac(6')-Ib-cr gene encoding a fluoroquinolne-modifying enzyme was detected in 64.7 per cent of the ETEC. Class 1 (intI1) and class 2 (intI2) integrons were detected in six (35.3%) and three (17.6%) strains, respectively. Four strains (23.5%) had both the classes of integrons. Sequence analysis revealed presence of dfrA17, aadA1, aadA5 in class 1, and dfrA1, sat1, aadA1 in class 2 integrons. In addition, the other resistance genes such as tet gene alleles (94.1%), catAI (70.6%), strA (58.8%), bla TEM-1 (35.2%), and aphA1-Ia (29.4%) were detected in most of the strains. Interpretation & Conclusions: Innate gene mutations and acquisition of multidrug resistance genes through mobile genetic elements might have contributed to the emergence of multidrug resistance (MDR) in ETEC. This study reinforces the necessity of utilizing molecular techniques in the epidemiological studies to understand the nature of resistance responsible for antimicrobial resistance in different species of pathogenic bacteria

    Molecular evidence of clonal Vibrio parahaemolyticus pandemic strains.

    The upsurge in worldwide incidence of Vibrio parahaemolyticus infection in the last 5 years has been attributed to the recent appearance of three serotypes with pandemic potential: O3:K6, O4:K68, and O1:K untypeable (KUT). Thirty-five strains of these serotypes, isolated from different countries over 4 years, were characterized by ribotyping and pulsed-field gel electrophoresis to determine their origin. The ribotypes of the strains of these serotypes were indistinguishable, except for a Japanese tdh- negative O3:K6 strain and a U.S. clinical O3:K6 isolate, which had slightly different profiles. The migration patterns of the NotI-digest of the total DNA of the strains were similar, and only slight variations were observed between the serotypes. By contrast, the O3:K6 and O1:KUT strains isolated before 1995 and strains of other serotypes had markedly different profiles. The O4:K68 and O1:KUT strains most likely originated from the pandemic O3:K6 clone

    An indigenous cluster beam apparatus with a reflectron time-of-flight mass spectrometer

    The design and fabrication of a Smalley-type cluster source in combination with a reflectron based time-of-flight (TOF) mass spectrometer are reported. The generation of clusters is based on supersonic jet expansion of the sampling plume. Sample cells for both liquid and solid targets developed for this purpose are described. Two pulsed Nd-YAG lasers are used in tandem, one (532 nm) for target vapourization and the other (355 nm) for cluster ionization. Methanol clusters of nuclearity up to 14 (mass 500 amu) were produced from liquid methanol as the test sample. The clusters were detected with a mass resolution of ~2500 in the R-TOF geometry. Carbon clusters up to a nuclearity of 28 were obtained using a polyimide target. The utility of the instrument is demonstrated by carrying out experiments to generate mixed clusters from alcohol mixtures

    Vibrio cholerae O139 in Calcutta, 1992-1998: incidence, antibiograms, and genotypes.

    We report results of surveillance for cholera caused by Vibrio cholerae O139 from September 1992, when it was first identified, to December 1998. V. cholerae O139 dominated as the causative agent of cholera in Calcutta during 1992-93 and 1996- 97, while the O1 strains dominated during the rest of the period. Dramatic shifts in patterns of resistance to cotrimoxazole, neomycin, and streptomycin were observed. Molecular epidemiologic studies showed clonal diversity among the O139 strains and continuous emergence of new epidemic clones, reflected by changes in the structure, organization, and location of the CTX prophages in the V. cholerae O13

    Age for measles immunization seroconversion after measles vaccination at 6-8 months of age - A randomised controlled trial

    The, objective of the study was to compare the effectiveness of measles vaccine by seroconversion in vaccinated children with non-vaccinated children of 6 to 8 months age group in a city slam community so as to study the feasibility of advancing the age of immunization. Live attenuated lyophilized Schwartz strain of measles vaccine was used. Hemagglutination inhibition (HI) antibody was estimated. Seroconversion was defined as either the conversion of negative to positive or a two fold rise in titre. One hundred and thirty two children completed the study. There was no difference in the age, sex and nutritional status between vaccinated and non-vaccinated groups (p>0.7). The seroconversion rate in the vaccinated group was 65% and in the non-vaccinated group was 26%. The age, sex and nutritional status did not significantly affect the seroconversion. Oar data suggest that immunization with measles vaccine may be effective as early as 6 months of age. Immunization at 6 months may be needed at least for children in densely populated areas like cities and towns
