    Developmental changes in the histological structure of the testes, and testosterone profiles in male guinea fowls (Numida meleagris)

    Owing to the paucity of information on the reproductive biology of guinea fowls, a study involving a total of 66 males was conducted, and documented the developmental changes in histological structure of the testes of guinea cocks from hatching until adulthood. Changes in testosterone synthesis during sexual development were also determined. Age-related changes were analysed using univariate analysis for completely randomised design and means separated using Tukey's test/Kruskal-Wallis test and medians separated by Mann-Whitney U test. Total germ cell population per testis and testicular histological morphometric parameters increased significantly (p < 0.0001) from 12 weeks of age (WOA), and stabilized between 20 and 24 WOA. Peripheral testosterone concentrations increased gradually from 4 WOA, and peaked at 20 WOA. Correlations among all the testicular morphometric parameters were positive and highly significant (p < 0.01). Similarly, significant (p < 0.05) positive correlations existed between testicular weight and testicular sperm production, tubular diameter, Sertoli cell population, tubular length and peripheral testosterone concentration. Testicular sperm production was positively correlated with meiotic index (p < 0.01) and round spermatids population (p < 0.05). The correlations between peripheral testosterone concentrations, tubular diameter and Sertoli efficiency were also significant (p < 0.05) and positive. Testicular morphometric parameters stabilized between 20 and 24 WOA, while peripheral testosterone concentrations showed two patterns of secretion, initial and final phases of increasing and decreasing testosterone secretions, respectively, and may be implicated in the development of histological structures of the testes and spermatogenesis.     Monthly and Diurnal Variability of Rain Rate and Rain Attenuation during the Monsoon Period in Malaysia

    Rain is the major source of attenuation for microwave propagation above 10 GHz. In tropical and equatorial regions where the rain intensity is higher, designing a terrestrial and earth-to-satellite microwave links is very critical and challenging at these frequencies. This paper presents the preliminary results of rain effects in a 23 GHz terrestrial point-to-point communication link 1.3km long. The experimental test bed had been set up at Skudai, Johor Bahru, Malaysia. In this area, a monsoon equatorial climate prevails and the rainfall rate can reach values well above 100mm/h with significant monthly and diurnal variability. Hence, it is necessary to implement a mitigation technique for maintaining an adequate radio link performance for the action of very heavy rain. Since we now know that the ULPC (Up Link Power Control) cannot guarantee the desired performance, a solution based on frequency band diversity is proposed in this paper. Here, a secondary radio link operating in a frequency not affected by rain (C band for instance) is placed parallel with the main link. Under no rain or light rain conditions, the secondary link carries without priority radio signals. When there is an outage of the main link due to rain, the secondary link assumes the priority traffic. The outcome of the research shows a solution for higher operating frequencies during rainy events

    Antimicrobial Diterpenes: Recent Development From Natural Sources

    Antimicrobial resistance has been posing an alarming threat to the treatment of infectious diseases over the years. Ineffectiveness of the currently available synthetic and semisynthetic antibiotics has led the researchers to discover new molecules with potent antimicrobial activities. To overcome the emerging antimicrobial resistance, new antimicrobial compounds from natural sources might be appropriate. Secondary metabolites from natural sources could be prospective candidates in the development of new antimicrobial agents with high efficacy and less side effects. Among the natural secondary metabolites, diterpenoids are of crucial importance because of their broad spectrum of antimicrobial activity, which has put it in the center of research interest in recent years. The present work is aimed at reviewing recent literature regarding different classes of natural diterpenes and diterpenoids with significant antibacterial, antifungal, antiviral, and antiprotozoal activities along with their reported structure–activity relationships. This review has been carried out with a focus on relevant literature published in the last 5 years following the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines. A total of 229 diterpenoids from various sources like plants, marine species, and fungi are summarized in this systematic review, including their chemical structures, classification, and significant antimicrobial activities together with their reported mechanism of action and structure–activity relationships. The outcomes herein would provide researchers with new insights to find new credible leads and to work on their synthetic and semisynthetic derivatives to develop new antimicrobial agents

    High-Dimensional Stochastic Design Optimization by Adaptive-Sparse Polynomial Dimensional Decomposition

    This paper presents a novel adaptive-sparse polynomial dimensional decomposition (PDD) method for stochastic design optimization of complex systems. The method entails an adaptive-sparse PDD approximation of a high-dimensional stochastic response for statistical moment and reliability analyses; a novel integration of the adaptive-sparse PDD approximation and score functions for estimating the first-order design sensitivities of the statistical moments and failure probability; and standard gradient-based optimization algorithms. New analytical formulae are presented for the design sensitivities that are simultaneously determined along with the moments or the failure probability. Numerical results stemming from mathematical functions indicate that the new method provides more computationally efficient design solutions than the existing methods.     Arginine mutation alters binding of a human monoclonal antibody to antigens linked to systemic lupus erythematosus and the antiphospholipid syndrome

    Objective: Previous studies have shown the importance of somatic mutations and arginine residues in the complementarity-determining regions (CDRs) of pathogenic anti-double-stranded DNA (anti-dsDNA) antibodies in human and murine lupus, and in studies of murine antibodies, a role of mutations at position 53 in VH CDR2 has been demonstrated. We previously demonstrated in vitro expression and mutagenesis of the human IgG1 monoclonal antibody B3. The present study was undertaken to investigate, using this expression system, the importance of the arginine residue at position 53 (R53) in B3 VH. Methods: R53 was altered, by site-directed mutagenesis, to serine, asparagine, or lysine, to create 3 expressed variants of VH. In addition, the germline sequence of the VH3-23 gene (from which B3 VH is derived) was expressed either with or without arginine at position 53. These 5 new heavy chains, as well as wild-type B3 VH, were expressed with 4 different light chains, and the resulting antibodies were assessed for their ability to bind to nucleosomes, -actinin, cardiolipin, ovalbumin, 2-glycoprotein I (2GPI), and the N-terminal domain of 2GPI (domain I), using direct binding assays. Results: The presence of R53 was essential but not sufficient for binding to dsDNA and nucleosomes. Conversely, the presence of R53 reduced binding to -actinin, ovalbumin, 2GPI, and domain I of 2GPI. The combination B3 (R53S) VH/B3 VL bound human, but not bovine, 2GPI. Conclusion: The fact that the R53S substitution significantly alters binding of B3 to different clinically relevant antigens, but that the alteration is in opposite directions depending on the antigen, implies that this arginine residue plays a critical role in the affinity maturation of antibody B3

    Flat Cosmology with Coupled Matter and Dark Energies

    Three models of a flat universe of coupled matter and dark energies with different low-redshift parameterizations of the dark energy equation of state are considered. The dark energy is assumed to vary with time like the trace of the energy-momentum tensor of cosmic matter. In the radiation-dominated era the models reduce to standard cosmology. In the matter-dominated era they are, for modern values of the cosmological parameters, consistent with data from SNe Ia searches and with the data of Gurvits et al.(1999)for angular sizes of ultra compact radio sources. We find that the angular size-redshift tests for our models offer a higher statistical confidence than that based on SNe Ia data. A comparison of our results with a recent revised analysis of angular size-redshift legacy data is made,and the implications of our models with optimized relativistic beaming in the radio sources is discussed. In particular we find that relativistic beaming implies a Lorentz factor less than 6,in agreement with its values for powerful Active Galactic Nuclei.Comment: Version to appear in The Astronomical Journal, with a modified name- Flat Cosmology with Coupled Matter and Dark Energies.
