    Professionalising purchasing organisations : towards a purchasing development model

    How does purchasing develops as a discipline over time in large international organisations? What are the drivers and enablers behind the development of purchasing? Is there an ideal growthpath for purchasing in organisations? These subjects are discussed in this paper by, firstly, providing an overview of current literature and analysing the models which have been published in management literature. Based on our analysis we propose an integrated framework which may help practitioners in developing their future courses of action with regard to purchasing and supply strategy and organisation. Currently, this framework is tested through empirical research

    Application of In Situ Neutron and X-Ray Measurements at High Temperatures in the Development of Co-Re-Based Alloys for Gas Turbines

    Co-Re alloy development is prompted by the search for new materials for future gas turbines which can be used at temperatures considerably higher than the current day single crystal Ni-based superalloys. The Co-Re-based alloys have been designed to have very high melting range, and they are meant for application at +373 K (+100 °C) above Ni-superalloys. They are significantly different from the conventional Co-based alloys that are used in static components of today’s gas turbines, and the Co-Re alloys have never been used for structural applications before. The Co-Re-Cr system has complex microstructure with many different phases present. Phase transformations and stabilities of fine strengthening precipitates at high temperatures remain mostly unexplored in the Co-Re alloys, and to develop basic understanding, model ternary and quaternary compositions were studied within the alloy development program. In situ neutron and synchrotron measurements at high temperatures were extensively used for this purpose, and some recent results from the in situ measurements are presented. In particular, the effect of boron doping in Co-Re alloys and the stabilities of the fine TaC precipitates at high temperatures were investigated. A fine dispersion of TaC precipitates strengthens some Co-Re alloys, and their stabilities at the application temperatures are critical.     Mineralogical characterization of a highly-weathered soil by the Rietveld Method Caracterização mineralógica de um solo altamente intemperizado pelo Método de Rietveld

    The mineralogical characterization through mineral quantification of Brazilian soils by X-ray diffraction data using the Rietveld Method is not common. A mineralogical quantification of an Acric Ferralsol from the Ponta Grossa region, state of Paraná, Brazil, was carried out using this Method with X-Ray Diffraction data to verify if this method was suitable for mineral quantification of a highly-weathered soil. The A, AB and B3 horizons were fractioned to separate the different particle sizes: clay, silt, fine sand (by Stokes Law) and coarse sand fractions (by sieving), with the procedure free of chemical treatments. X-ray Fluorescence, Inductively Coupled Plasma Atomic Emission Spectrometry, Infrared Spectroscopy and Mössbauer Spectroscopy were used in order to assist the mineral identification and quantification. The Rietveld Method enabled the quantification of the present minerals. In a general way, the quantitative mineralogical characterization by the Rietveld Method revealed that quartz, gibbsite, rutile, hematite, goethite, kaolinite and halloysite were present in the clay and silt fractions of all horizons. The silt fractions of the deeper horizons were different from the more superficial ones due to the presence of large amounts of quartz. The fine and the coarse sand fractions are constituted mainly by quartz. Therefore, a mineralogical quantification of the finer fraction (clay and silt) by the Rietveld Method was successful.<br>A caracterização mineralógica por meio da quantificação dos minerais presentes em solos brasileiros por difração de raios X usando o Método de Rietveld é, ainda, pouco comum. Neste trabalho foi realizada a quantificação mineralógica de um Latossolo Vermelho ácrico da região de Ponta Grossa, Paraná, Brasil, utilizando o Método de Rietveld com dados de Difração de Raios X e também verificado se o método foi adequado na quantificação mineral de um solo altamente intemperizado. Os horizontes A, AB e B3 foram separados fisicamente nas frações: argila, silte, areia fina (por meio da Lei de Stokes) e areia grossa (por peneiramento) sem qualquer tratamento químico. As técnicas de Fluorescência de raios X, Espectroscopia de Emissão Atômica com Fonte de Plasma Induzido, Espectroscopia de Infravermelho e Espectroscopia Mössbauer foram utilizadas para auxiliar na identificação e quantificação dos minerais. O Método de Rietveld possibilitou a quantificação dos minerais presentes nas frações argila e silte de todos os horizontes: quartzo, gibbsita, rutílio, hematita, goethita, caulinita e haloisita. Na fração silte os horizontes mais profundos são diferentes do mais superficial devido à presença de grandes quantidades de quartzo. As frações areia fina e grossa são constituídas principalmente de quartzo. Portanto, o Método de Rietveld foi adequado para quantificação dos minerais, principalmente, das frações mais finas (argila e silte)

    Synthetic seeds can be an alternative for those species in which botanical seeds are not viable. One of the major problems of in vitro plant cultivation is the high level of somaclonal variation. The most common factors affecting somaclonal variation are genotype, explant source, in vitro period and cultivation conditions in which the culture is established. In this work, calli were induced using leaf and stem explants of the commercial potato cultivars Achat, Baraka, Baronesa, Bintje, and Contenda in MS culture media supplemented with 1.65 mM of picloram and 11.5 mM of 2,4-D. Seventy and 90 days after induction, DNA samples of 40 calli were compared concerning the effects of the two explant (leaf and stem) and two growth regulator sources on five potatoes cultivars. A total of 20 arbitrary sequence primers were evaluated. The RAPD pattern generated by these primers suggested a high percentage of polymorphic fragments among the five genotypes, indicating a high level of genetic variation among cultivars. Cultivar Baronesa showed the highest number of polymorphic fragments for all treatments. The cultivar Contenda showed the smallest somaclonal variation, for most of the treatments, except for the treatment which consisted of stem explants, picloram (1.65 mM) application, and a 70-day period of callus formation. 'Contenda' is, therefore, the most suitable cultivar for synthetic seed production.<br>A produção de sementes sintéticas pode ser uma altenativa para culturas, cuja produção de sementes botânicas não é viável. Um dos principais problemas do cultivo de planta in vitro é o alto nível de variação somaclonal resultante. Os fatores mais comuns que afetam a variação somaclonal são genótipo, fonte de explante e duração e condições de cultivo. Neste trabalho, calos foram induzidos utilizando explantes de folha e caule das cultivares comerciais de batata Achat, Baraka, Baronesa, Bintje e Contenda em meio de cultura MS, suplementado com 1.65 µM de picloram ou 11.5 µM de 2,4-D. Após 70 e 90 dias de indução, amostras de DNA de 40 calos foram analisadas por meio de RAPD, em um estudo comparativo entre os efeitos das duas fontes de explante e os dois reguladores de crescimento nas 5 cultivares. Para este propósito, 20 primers de seqüência arbitrária foram testados. No padrão RAPD gerado por estes primers observou-se alta porcentagem de fragmentos polimóficos entre os 5 genótipos, indicando alto nível de variação genética entre as cultivares. A cultivar Baronesa apresentou o mais alto número de fragmentos polimórficos para os tratamentos. A cultivar Contenda apresentou a menor variação somaclonal genética entre os genótipos e tratamentos utilizados, exceto para o tratamento que consistiu de caule como explante, uso de picloram (1.65 mM) e 70 dias para formação de calos. Portanto, 'Contenda' é a cultivar mais adequada para a produção de sementes sintéticas
