Enabling Future Sustainability Transitions: An Urban Metabolism Approach to Los Angeles Pincetl et al. Enabling Future Sustainability Transitions

    Get PDF
Summary: This synthesis article presents an overview of an urban metabolism (UM) approach using mixed methods and multiple sources of data for Los Angeles, California. We examine electric energy use in buildings and greenhouse gas emissions from electricity, and calculate embedded infrastructure life cycle effects, water use and solid waste streams in an attempt to better understand the urban flows and sinks in the Los Angeles region (city and county). This quantification is being conducted to help policy-makers better target energy conservation and efficiency programs, pinpoint best locations for distributed solar generation, and support the development of policies for greater environmental sustainability. It provides a framework to which many more UM flows can be added to create greater understanding of the study area's resource dependencies. Going forward, together with policy analysis, UM can help untangle the complex intertwined resource dependencies that cities must address as they attempt to increase their environmental sustainability

Primary education in Mexico is facing a growing set of challenges that the government has tried to counteract through the use of communication technologies (ICT) in formal education. While these efforts provide support for students and educators, there remains a need for a renewed and contextualized awareness that will re-conceptualize the adverse experiences of students and the importance of resilience in the context of the learning environment. The objective of this document is to give an account of the learning acquired by nine telesecondary students in a rural area of Hidalgo and the student’s benefits of building awareness about the ecology of learning. The paper highlights the process of re-envisioning their experiences that emerged from the various points of views shared in discussion. The study was triangulated by quantitative and qualitative phenomenological and hermeneutical analyses. It was organized into three stages and employed a survey, semi-structured interviews, focus groups and use of the Google-Classroom platform. The hermeneutical analysis of autobiographies and the use of technological resources enhanced the personal analysis of the experiences of the participants. These experiences generated learning that may often be invisible in formal education but can empower critical thinking, collaboration and autonomy of students to become aware of their learning and the scope of their social contribution

Repositioning of Verrucosidin, a purported inhibitor of chaperone protein GRP78, as an inhibitor of mitochondrial electron transport chain complex I.

Verrucosidin (VCD) belongs to a group of fungal metabolites that were identified in screening programs to detect molecules that preferentially kill cancer cells under glucose-deprived conditions. Its mode of action was proposed to involve inhibition of increased GRP78 (glucose regulated protein 78) expression during hypoglycemia. Because GRP78 plays an important role in tumorigenesis, inhibitors such as VCD might harbor cancer therapeutic potential. We therefore sought to characterize VCD's anticancer activity in vitro. Triple-negative breast cancer cell lines MDA-MB-231 and MDA-MB-468 were treated with VCD under different conditions known to trigger increased expression of GRP78, and a variety of cellular processes were analyzed. We show that VCD was highly cytotoxic only under hypoglycemic conditions, but not in the presence of normal glucose levels, and VCD blocked GRP78 expression only when glycolysis was impaired (due to hypoglycemia or the presence of the glycolysis inhibitor 2-deoxyglucose), but not when GRP78 was induced by other means (hypoxia, thapsigargin, tunicamycin). However, VCD's strictly hypoglycemia-specific toxicity was not due to the inhibition of GRP78. Rather, VCD blocked mitochondrial energy production via inhibition of complex I of the electron transport chain. As a result, cellular ATP levels were quickly depleted under hypoglycemic conditions, and common cellular functions, including general protein synthesis, deteriorated and resulted in cell death. Altogether, our study identifies mitochondria as the primary target of VCD. The possibility that other purported GRP78 inhibitors (arctigenin, biguanides, deoxyverrucosidin, efrapeptin, JBIR, piericidin, prunustatin, pyrvinium, rottlerin, valinomycin, versipelostatin) might act in a similar GRP78-independent fashion will be discussed

Rapid verification of terminators using the pGR-blue plasmid and golden gate assembly

The goal of this protocol is to allow for the rapid verification of bioinformatically identified terminators. Further, the plasmid (pGR-Blue) is designed specifically for this protocol and allows for the quantification of terminator efficiency. As a proof of concept, six terminators were bioinformatically identified in the mycobacteriophage Bernal13. Once identified, terminators were then made as oligonucleotides with the appropriate sticky ends and annealed together. Using Golden Gate Assembly (GGA), terminators were then cloned into pGR-Blue. Under visible light, false positive colonies appear blue and positively transformed colonies are white/yellow. After induction of an arabinose inducible promoter (pBad) with arabinose, colony strength can be determined by measuring the ratio of green fluorescent protein (GFP) produced to red fluorescent protein (RFP) produced. With pGR-Blue, the protocol can be completed in as little as three days and is ideal in an educational setting. Additionally, results show that this protocol is useful as a means for understanding in silico predictions of terminator efficiency related to the regulation of transcription

Spectral analysis and new visible and ultraviolet lines of Ar V

A capillary discharge tube was used to record the Ar spectrum in the region of 330-4400 angstrom. We analyzed a set of 109 lines of Ar V, with 10 of them being classified for the first time. Part of these newly identified lines (six in total) corresponds to electric dipole transitions in the visible and ultraviolet regions of the spectrum. We calculated weighted transition rates (gA) for all experimentally observed lines and the corresponding lifetimes using a relativistic Hartree-Fock method with configuration interaction. The equations were modified to consider core polarization (CP) effects. A study on the Si isoelectronic sequence found that considering CP effects is essential for the correct description of the Ar V structure2422COORDENAÇÃO DE APERFEIÇOAMENTO DE PESSOAL DE NÍVEL SUPERIOR - CAPESConsejo Nacional de Investigaciones Cientificas y Tecnicas (CONICET), ArgentinaConsejo Nacional de Investigaciones Cientificas y Tecnicas (CONICET); Coordenaccao de Aperfeicoamento de Pessoal de Nivel Superior (CAPES), Brazil; Comision de Investigaciones Cientificas de la Provincia de Buenos Aires (CIC

Lately, neuroscience has proven key in providing scientific answers to research conducted in other disciplines. Resilience, from the point of view of neuroscience, takes on a new meaning due to the lockdown experienced under the COVID-19 pandemic. This article is the result of an investigation carried out during confinement in Spain, Mexico and Chile. It shows the preliminary results of the analysis of stress risk factors and resilience in neuroscience, identifying possible stress risk factors using socio-demographic data. A descriptive, cross-sectional, correlational and comparative study was conducted in a sample of 784 participants. The Connor-Davidson Resilience Scale (CD-RISC10) was used to identify resilience traits. The findings reveal a non-significant result among the countries, but statistically significant results in the level of education variable, which is associated with cognitive flexibility. The possible stress risk factors did not evince significant results. Thus, promoting resilience at an early age comes as a relevant finding. As argued by epigenetics, to immerse oneself in contexts where meaningful bonds are promoted, is essential to improve vital processes such as resilience and prevent potential stress risk factors.

The thirteen times ionized tungsten is isoelectronic with PmI. Wavelengths and transition probabilities for the 5s-5p and 5p-5d transitions of WXIV, identifying the ground state as 4f13 5s2 2F7/2 were calculated. Both, a relativistic Hartree Fock approach, including core-polarization effects, and a purely relativistic multiconfiguration Dirac-Fock method were used for the calculations. Particularly, 5s-5p transitions were compared with experimental results obtained with VUV electron beam ion trap (EBIT) spectroscopy583CONSELHO NACIONAL DE DESENVOLVIMENTO CIENTÍFICO E TECNOLÓGICO - CNPQCOORDENAÇÃO DE APERFEIÇOAMENTO DE PESSOAL DE NÍVEL SUPERIOR - CAPESnão temnão tem17th International conference on the physics of highly charged ionsThis research was supported by the Consejo Nacional de Investigaciones Científicas y Técnicas (CONICET), Argentina, by the Coordenaçăo de Aperfeiçoamento de Pessoal de Nível Superior (CAPES) and by the Conselho Nacional de Pesquisa (CNPq), Brazil.
