    Retrofitting housing with lightweight green roof technology in Sydney, Australia, and Rio de Janeiro, Brazil

The built environment contributes around half of total greenhouse gas emissions and with 87% of residential buildings that we will have by 2050 already built, it is vital to adopt sustainable retrofitting practices. The question is: what are the viable solutions? One answer may be green roof retrofitting. The environmental benefits include reduced operational carbon emissions, reduced urban heat island effect, increased bio-diversity, housing temperature attenuation and reduced stormwater run-off. The economic benefits are the reduced maintenance costs and lower running costs. The social gain is the creation of spaces where people have access to green areas. However, the barriers to retrofitting include the perceptions of structural adequacy, the risk of water damage, high installation and maintenance costs, as well as access and security issues. Many Australian and Brazilian residential buildings have metal sheet roofs, a lightweight material with poor thermal performance. During the summer, temperatures in Sydney and Rio de Janeiro reach 45 degrees Celsius, and in both cities, rainfall patterns are changing, with more intense downpours. Furthermore, many residential buildings are leased, and currently, tenants are restricted by the modifications that they can perform to reduce running costs and carbon emissions. This research reports on an experiment on two small-scale metal roofs in Sydney and Rio de Janeiro to assess the thermal performance of portable small-scale modules. The findings are that considerable variation in temperature was found in both countries, indicating that green roof retrofitting could lower the cooling energy demand considerably

    Retrofitted green roofs and walls and improvements in thermal comfort

Increased urbanization has led to a worsening in the quality of life for many people living in large cities in respect of the urban heat island effect and increases of indoor temperatures in housing and other buildings. A solution may be to retrofit existing environments to their former conditions, with a combination of green infrastructures applied to existing walls and rooftops. Retrofitted green roofs may attenuate housing temperature. However, with tall buildings, facade areas are much larger compared to rooftop areas, the role of green walls in mitigating extreme temperatures is more pronounced. Thus, the combination of green roofs and green walls is expected to promote a better thermal performance in the building envelope. For this purpose, a modular vegetated system is adopted for covering both walls and rooftops. Rather than temperature itself, the heat index, which comprises the combined effect of temperature and relative humidity is used in the evaluation of thermal comfort in small scale experiments performed in Sydney - Australia, where identical timber framed structures prototypes (vegetated and non-vegetated) are compared. The results have shown a different understanding of thermal comfort improvement regarding heat index rather than temperature itself.     © 2017 The Authors. With increasing densification in urban settlements, environmental issues are a challenge in the sustainable development of all cities globally. Considering that the built environment releases almost half of the total greenhouse gas emissions, an effective solution to mitigating the impacts of increasing temperatures can be the improved performance of existing buildings. Furthermore 87% of the buildings we will have in 2050 are already built. Retrofitting roofs and walls with a living vegetated system such as green roofs and walls could be an upgrade option, increasing sustainable construction. The benefits are improved thermal performance but also improved air quality, stormwater attenuation, increased bio-diversity and lower heating and / or cooling energy consumption. No empirical data exists for Sydney and Rio de Janeiro and the question is; what is the extent of thermal improvement with retrofitted green walls and roof in timber framed and blockwork structures? This study analyses both effects and benefits of the green roofs and walls through an experiment in two countries: one in Sydney, Australia; a timber framed construction, and another one in Rio de Janeiro, Brazil; with blockwork construction. This difference in the material choice was made according to the most common type of construction for housing in each country. In each site, the walls and the roof of one of the prototypes were covered with plants and compared to the performance of an unplanted but otherwise identical prototype. The thermal performance was analysed by observing the temperature variation simultaneously in a non-vegetated and vegetated structure. The initial findings show that the combination of green roof and green walls have a relevant role in temperature attenuation. These results indicate, that this lightweight retrofit green technology could not only represent an important advance on sustainable development, but can that it also lead to more comfortable internal conditions for humans living in dense urban environments

    Characteristics of temporal patterns of cortisol and luteinizing hormone in primiparous, postpartum, anovular, suckled, beef cows exposed acutely to bulls

    BACKGROUND: The physiological mechanism by which bulls stimulate resumption of ovarian cycling activity in postpartum, anovular, suckled cows after calving may involve the concurrent activation of the hypothalamic-hypophyseal-ovarian (HPO) axis and hypothalamic-hypophyseal-adrenal (HPA) axis. Thus, the objectives of this experiment were to determine if characteristics of temporal patterns of cortisol and luteinizing hormone (LH) in postpartum, anovular, beef cows are influenced by acute exposure to bulls. The null hypotheses were that daily, temporal characteristics of cortisol and LH concentration patterns do not differ between cows exposed acutely to bulls or steers. METHODS: Sixteen cows were assigned randomly 67 +/- 4 (+/- SE) after calving to be exposed to bulls (EB, n = 8) or steers (ES, n = 8) 5 h daily for 9 d (D 0 to 8). Blood samples were collected daily from each cow via jugular catheters at 15-min intervals for 6 h from 1000 to 1600 h each day. The 5-h exposure period began 1 h after the start of the intensive bleeding period. Characteristics of cortisol and LH concentration patterns (mean, baseline, pulse frequency, pulse amplitude, and pulse duration) were identified by PULSAR analyses. RESULTS: Mean cortisol concentrations decreased (P < 0.05) in cows in both treatments from D 0 to D 2. Thereafter, mean cortisol concentrations stabilized and did not differ (P > 0.10) between EB and ES cows. The decrease in mean cortisol concentrations in EB and ES cows from D 0 to D 2 was attributed to cows acclimatizing to intensive blood sampling and handling procedures. Consequently, analyses for characteristics of cortisol and LH concentration patterns included D 2 through 8 only. Cortisol mean and baseline concentrations, and pulse amplitude did not differ (P > 0.10) between EB and ES cows. However, cortisol pulse duration tended to be longer (P = 0.09) and pulse frequency was lower (P = 0.05) in EB than ES cows. LH pulse frequency was greater (P = 0.06) in EB than ES cows. All other characteristics of LH concentration patterns did not differ (P > 0.10) between EB and ES cows. Characteristics of cortisol concentration patterns were not related to characteristics of LH concentration patterns for ES cows (P > 0.10). However, as cortisol pulse amplitude increased, LH pulse amplitude decreased (b1 = -0.04; P < 0.05) for EB cows. CONCLUSIONS: In conclusion, exposing primiparous, postpartum, anovular, suckled cows to bulls for 5-h daily over a 9-d period did not alter mean concentrations of cortisol or LH compared to mean concentrations of cortisol and LH in cows exposed to steers. However, exposing cows to bull in this manner altered characteristics of temporal patterns of both LH and cortisol by increasing LH pulse frequency and decreasing cortisol pulse frequency. Interestingly, in cows exposed to bulls, as amplitude and frequency of cortisol pulses decreased, amplitudes of LH pulses increased and frequency of LH pulses tended to increase. Thus, the physiological mechanism of the biostimulatory effect of bulls may initially involve modification of the HPA axis and these changes may facilitate activation of the HPO axis and resumption of ovulatory cycles in postpartum, anovular, suckled cows

    A mutate-and-map protocol for inferring base pairs in structured RNA

    Chemical mapping is a widespread technique for structural analysis of nucleic acids in which a molecule's reactivity to different probes is quantified at single-nucleotide resolution and used to constrain structural modeling. This experimental framework has been extensively revisited in the past decade with new strategies for high-throughput read-outs, chemical modification, and rapid data analysis. Recently, we have coupled the technique to high-throughput mutagenesis. Point mutations of a base-paired nucleotide can lead to exposure of not only that nucleotide but also its interaction partner. Carrying out the mutation and mapping for the entire system gives an experimental approximation of the molecules contact map. Here, we give our in-house protocol for this mutate-and-map strategy, based on 96-well capillary electrophoresis, and we provide practical tips on interpreting the data to infer nucleic acid structure.Comment: 22 pages, 5 figure

    Bioavailability, Antipsoriatic Efficacy and Tolerability of a New Light Cream with Mometasone Furoate 0.1%

    Mometasone furoate, a potent glucocorticoid (class III) with a favorable benefit/risk ratio, has emerged as a standard medication for the treatment of inflammatory skin disorders. The purpose of the investigation presented here was to determine the noninferiority of a topical mometasone formulation, a light cream (O/W 60/40 emulsion) with mometasone furoate 0.1% (water content of 33%) versus marketed comparators. Using the vasoconstrictor assay, a strong blanching effect of the new cream (called Mometasone cream) comparable to that of a mometasone comparator, a fatty cream with mometasone furoate 0.1%, could be demonstrated. Thus, the topical bioavailability of the active ingredient mometasone furoate (0.1%) was regarded to be similar for Mometasone cream and the mometasone comparator. Using the psoriasis plaque test, a strong antipsoriatic effect comparable to that of the mometasone comparator was found for Mometasone cream after 12 days of occlusive treatment. A nearly identical reduction in the mean infiltrate thickness and similar mean AUC values were noted with both formulations confirmed by clinical assessment data. The noninferiority of Mometasone cream to its active comparator with re-spect to the AUC of change to baseline in infiltrate thickness was demonstrated. Both medications were well tolerated. Overall, Mometasone cream and the mometasone comparator showed similar efficacy and tolerability. Mometasone cream, in addition to its high potency and good tolerability, provides the properties of a light cream, which might make this new medication particularly suitable for application on acutely inflamed and sensitive skin.     Parental investment by skin feeding in a caecilian amphibian

    Although the initial growth and development of most multicellular animals depends on the provision of yolk, there are many varied contrivances by which animals provide additional or alternative investment in their offspring(1). Providing offspring with additional nutrition should be favoured by natural selection when the consequent increased fitness of the young offsets any corresponding reduction in fecundity(2). Alternative forms of nutrition may allow parents to delay and potentially redirect their investment. Here we report a remarkable form of parental care and mechanism of parent-offspring nutrient transfer in a caecilian amphibian. Boulengerula taitanus is a direct-developing, oviparous caecilian(3), the skin of which is transformed in brooding females to provide a rich supply of nutrients for the developing offspring. Young animals are equipped with a specialized dentition, which they use to peel and eat the outer layer of their mother's modified skin. This new form of parental care provides a plausible intermediate stage in the evolution of viviparity in caecilians. At independence, offspring of viviparous and of oviparous dermatotrophic caecilians are relatively large despite being provided with relatively little yolk.     Cyfip1 haploinsufficient rats show white matter changes, myelin thinning, abnormal oligodendrocytes and behavioural inflexibility

    The biological basis of the increased risk for psychiatric disorders seen in 15q11.2 copy number deletion is unknown. Previous work has shown disturbances in white matter tracts in human carriers of the deletion. Here, in a novel rat model, we recapitulated low dosage of the candidate risk gene CYFIP1 present within the 15q11.2 interval. Using diffusion tensor imaging, we first showed extensive white matter changes in Cyfip1 mutant rats, which were most pronounced in the corpus callosum and external capsule. Transmission electron microscopy showed that these changes were associated with thinning of the myelin sheath in the corpus callosum. Myelin thinning was independent of changes in axon number or diameter but was associated with effects on mature oligodendrocytes, including aberrant intracellular distribution of myelin basic protein. Finally, we demonstrated effects on cognitive phenotypes sensitive to both disruptions in myelin and callosal circuitry
