    Mutagenic and Spectroscopic Investigation of pH Dependent CooA DNA Binding

    The carbon monoxide (CO) sensing heme protein, CooA, is a transcription factor which exists in several bacteria that utilize CO as an energy source. CooA positively regulates the expression of coo genes in the presence of CO such that the corresponding proteins may metabolize CO. The present studies have yielded the unexpected result that Fe(III) CooA binds DNA tightly at pH \u3c 7, deviating from all previously reported work which indicate that CooA DNA binding is initiated only when the exogenous CO effector reacts with the Fe(II) CooA heme. This observation suggests that the disruption of one or more salt bridges upon effector binding may be a critical feature of the normal CooA activation mechanism. To test this possibility, several protein variants that eliminated a selected salt bridge for the CooA homolog from Rhodospirillum rubrum were prepared via site-directed mutagenesis. Samples of these variant proteins, which were overexpressed in Escherichia coli, were then characterized by spectroscopic methods and functional assays to investigate the impact these mutations had on CooA heme coordination structure and DNA-binding activity. Results of this work are presented in light of the accepted CooA activation mechanism

    Consequences of narrow cyclotron emission from Hercules X-1

    The implications of the recent observations of a narrow cyclotron line in the hard X-ray spectrum of Hercules X-1 are studied. A Monte Carlo code is used to simulate the X-ray transfer of an intrinsically narrow feature at approximately 56 keV through an opaque, cold magnetospheric shell. The results of this study indicate that if a narrow line can be emitted by the source region, then only about 10% of the photons remain in a narrow feature after scattering through the shell. The remaining photons are scattered into a broad feature (FWHM approximately 30 keV) that peaks near 20 keV. Thus, these calculations indicate that the intrinsic source luminosity of the cyclotron line is at least an order of magnitude greater than the observed luminosity

    A photometric function is proposed to describe the diffuse reflection of radiation by particulate materials. Both multiple scattering and the dominant effects of particle shadowing are included and the function is verified by comparisons with the photometries of laboratory surfaces. Brightness measurements of planetary and other diffusely scattering surfaces can be used to calculate the brightness for geometries other than those used in the measurements and for which the Minnaert function does not apply.     Simulation study of a geometric shape factor technique for estimating earth-emitted radiant flux densities from wide-field-of-view radiation measurements

    Geometric shape factors were computed and applied to satellite simulated irradiance measurements to estimate Earth emitted flux densities for global and zonal scales and for areas smaller than the detector field of view (FOV). Wide field of view flat plate detectors were emphasized, but spherical detectors were also studied. The radiation field was modeled after data from the Nimbus 2 and 3 satellites. At a satellite altitude of 600 km, zonal estimates were in error 1.0 to 1.2 percent and global estimates were in error less than 0.2 percent. Estimates with unrestricted field of view (UFOV) detectors were about the same for Lambertian and limb darkening radiation models. The opposite was found for restricted field of view detectors. The UFOV detectors are found to be poor estimators of flux density from the total FOV and are shown to be much better as estimators of flux density from a circle centered at the FOV with an area significantly smaller than that for the total FOV
