    Quantifying phase-amplitude coupling in neuronal network oscillations

    Neuroscience time series data from a range of techniques and species reveal complex, non-linear interactions between different frequencies of neuronal network oscillations within and across brain regions. Here, we briefly review the evidence that these nested, cross-frequency interactions act in concert with linearly covariant (within-frequency) activity to dynamically coordinate functionally related neuronal ensembles during behaviour. Such studies depend upon reliable quantification of cross-frequency coordination, and we compare the properties of three techniques used to measure phase-amplitude coupling (PAC)--Envelope-to-Signal Correlation (ESC), the Modulation Index (MI) and Cross-Frequency Coherence (CFC)--by standardizing their filtering algorithms and systematically assessing their robustness to noise and signal amplitude using artificial signals. Importantly, we also introduce a freely-downloadable method for estimating statistical significance of PAC, a step overlooked in the majority of published studies. We find that varying data length and noise levels leads to the three measures differentially detecting false positives or correctly identifying frequency bands of interaction; these conditions should therefore be taken into careful consideration when selecting PAC analyses. Finally, we demonstrate the utility of the three measures in quantifying PAC in local field potential data simultaneously recorded from rat hippocampus and prefrontal cortex, revealing a novel finding of prefrontal cortical theta phase modulating hippocampal gamma power. Future adaptations that allow detection of time-variant PAC should prove essential in deciphering the roles of cross-frequency coupling in mediating or reflecting nervous system function

    Student interpretations of the terms in first-order ordinary differential equations in modelling contexts

    A study of first-year undergraduate studentsâ€Č interpretational difficulties with first-order ordinary differential equations (ODEs) in modelling contexts was conducted using a diagnostic quiz, exam questions and follow-up interviews. These investigations indicate that when thinking about such ODEs, many students muddle thinking about the function that gives the quantity to be determined and the equation for the quantity's rate of change, and at least some seem unaware of the need for unit consistency in the terms of an ODE. It appears that shifting from amount-type thinking to rates-of-change-type thinking is difficult for many students. Suggestions for pedagogical change based on our results are made

    To swim or not to swim: A population-level model of Xenopus tadpole decision making and locomotor behaviour

    We present a detailed computational model of interacting neuronal populations that mimic the hatchling Xenopus tadpole nervous system. The model includes four sensory pathways, integrators of sensory information, and a central pattern generator (CPG) network. Sensory pathways of different modalities receive inputs from an "environment"; these inputs are then processed and integrated to select the most appropriate locomotor action. The CPG populations execute the selected action, generating output in motor neuron populations. Thus, the model describes a detailed and biologically plausible chain of information processing from external signals to sensors, sensory pathways, integration and decision-making, action selection and execution and finally, generation of appropriate motor activity and behaviour. We show how the model produces appropriate behaviours in response to a selected scenario, which consists of a sequence of "environmental" signals. These behaviours might be relatively complex due to noisy sensory pathways and the possibility of spontaneous actions

    Contextualising Apartheid at the End of Empire: Repression, ‘Development’ and the Bantustans

    This article examines the global dynamics of late colonialism and how these informed South African apartheid. More specifically, it locates the programmes of mass relocation and bantustan ‘self-government’ that characterised apartheid after 1959 in relation to three key dimensions. Firstly, the article explores the global circulation of idioms of ‘development’ and trusteeship in the first half of the twentieth century and its significance in shaping segregationist policy; secondly, it situates bantustan ‘selfgovernment’ in relation to the history of decolonisation and the partitions and federations that emerged as late colonial solutions; and, thirdly, it locates the tightening of rural village planning in the bantustans after 1960 in relation to the elaboration of anti-colonial liberation struggles, repressive southern African settler politics and the Cold War. It argues that, far from developing policies that were at odds with the global ‘wind of change’, South African apartheid during the 1960s and 1970s reflected much that was characteristic about late colonial strategy

    General anaesthetic and airway management practice for obstetric surgery in England: a prospective, multi-centre observational study

    There are no current descriptions of general anaesthesia characteristics for obstetric surgery, despite recent changes to patient baseline characteristics and airway management guidelines. This analysis of data from the direct reporting of awareness in maternity patients' (DREAMY) study of accidental awareness during obstetric anaesthesia aimed to describe practice for obstetric general anaesthesia in England and compare with earlier surveys and best-practice recommendations. Consenting patients who received general anaesthesia for obstetric surgery in 72 hospitals from May 2017 to August 2018 were included. Baseline characteristics, airway management, anaesthetic techniques and major complications were collected. Descriptive analysis, binary logistic regression modelling and comparisons with earlier data were conducted. Data were collected from 3117 procedures, including 2554 (81.9%) caesarean deliveries. Thiopental was the induction drug in 1649 (52.9%) patients, compared with propofol in 1419 (45.5%). Suxamethonium was the neuromuscular blocking drug for tracheal intubation in 2631 (86.1%), compared with rocuronium in 367 (11.8%). Difficult tracheal intubation was reported in 1 in 19 (95%CI 1 in 16-22) and failed intubation in 1 in 312 (95%CI 1 in 169-667). Obese patients were over-represented compared with national baselines and associated with difficult, but not failed intubation. There was more evidence of change in practice for induction drugs (increased use of propofol) than neuromuscular blocking drugs (suxamethonium remains the most popular). There was evidence of improvement in practice, with increased monitoring and reversal of neuromuscular blockade (although this remains suboptimal). Despite a high risk of difficult intubation in this population, videolaryngoscopy was rarely used (1.9%)

    After the RESTART trial: six guidelines for clinical trials of early stuttering intervention

    © 2019 Royal College of Speech and Language Therapists Background: The Rotterdam Evaluation Study of Stuttering Therapy randomized trial (RESTART) was seminal, comparing the Lidcombe Program with RESTART Demands and Capacities Model-based treatment (RESTART-DCM) for pre-school age children who stutter. Aims: To critique the methods of the RESTART trial to develop guidelines for its systematic replication and extension. Beyond that, to contribute to the refinement of existing methodological guidelines for early stuttering intervention. Method: The discussion is organized around methodological issues of primary outcomes, treatment completion, clinician allegiance, treatment fidelity, age exclusions and no-treatment control reasoning. Main Contribution: We recommend six methodological guidelines to guide future clinical trials comparing the Lidcombe Program with RESTART-DCM, which can be applied to clinical trials of other early stuttering intervention methods: (1) incorporate a continuous measure of primary outcome; (2) ensure that all children in clinical trials have completed treatment; (3) eliminate potential bias due to clinician allegiance; (4) establish treatment fidelity within and beyond the clinic; (5) include children younger than 3 years in clinical trials; and (6) establish an estimate of treatment effect size at some stage of treatment development. Conclusion: In addition to guiding future clinical research comparing RESTART-DCM and Lidcombe Program treatment, these recommendations may extend to influence positively other treatment developments for early stuttering

    A canonical circuit for generating phase-amplitude coupling

    ‘Phase amplitude coupling’ (PAC) in oscillatory neural activity describes a phenomenon whereby the amplitude of higher frequency activity is modulated by the phase of lower frequency activity. Such coupled oscillatory activity – also referred to as ‘cross-frequency coupling’ or ‘nested rhythms’ – has been shown to occur in a number of brain regions and at behaviorally relevant time points during cognitive tasks; this suggests functional relevance, but the circuit mechanisms of PAC generation remain unclear. In this paper we present a model of a canonical circuit for generating PAC activity, showing how interconnected excitatory and inhibitory neural populations can be periodically shifted in to and out of oscillatory firing patterns by afferent drive, hence generating higher frequency oscillations phase-locked to a lower frequency, oscillating input signal. Since many brain regions contain mutually connected excitatory-inhibitory populations receiving oscillatory input, the simplicity of the mechanism generating PAC in such networks may explain the ubiquity of PAC across diverse neural systems and behaviors. Analytic treatment of this circuit as a nonlinear dynamical system demonstrates how connection strengths and inputs to the populations can be varied in order to change the extent and nature of PAC activity, importantly which phase of the lower frequency rhythm the higher frequency activity is locked to. Consequently, this model can inform attempts to associate distinct types of PAC with different network topologies and physiologies in real data
