    Variability of Western Australian isolates of Sclerotinia sclerotiorum and the potential of Local Biological Control Agents

    Stem rot disease caused by Sclerotinia sclerotiorum has emerged as a serious problem for canola (Brassica napus L.) production in Western Australia (WA) over the past few years where crop losses can be up to 40% in the worst affected crops. The biological characteristics and pathogenicity of the pathogen in WA is poorly understood. Also the potential for local biological control agents (BCAs) to be used in the management of the disease has not been explored. This paper provides preliminary data in these fields. One hundred and forty isolates of S. sclerotiorum were collected from WA canola growing regions for identification of biological characteristics which include colour of mycelia, growth rate, production of sclerotia, and pathogenicity. Other fungal isolates with potential biological control activity were collected from southern regions of WA. Colour of mycelia of Sclerotinia isolates varied from white, yellowish white, greyish white, brownish white, grey, dark grey to brown. Each isolate had its 24 and 48 hour growth rate recorded after sub-culture on PDA + ampicillin medium. ANOVA showed highly significant differences between growth rates of isolates 24 and 48 hours after being sub-cultured (P≤0.001). There were significant differences in number of sclerotia produced by each isolate. Two potential fungal biological control agents were found in WA, namely isolate KEN1 and isolate MTB1. These local fungal BCAs were found to be effective in inhibiting in vitro both the growth and ability to produce sclerotia of S. sclerotiorum

    Molecular electronics exploiting sharp structure in the electrode density-of-states. Negative differential resistance and Resonant Tunneling in a poled molecular layer on Al/LiF electrodes

    Density-functional calculations are used to clarify the role of an ultrathin LiF layer on Al electrodes used in molecular electronics. The LiF layer creates a sharp density of states (DOS), as in a scanning-tunneling microscope (STM) tip. The sharp DOS, coupled with the DOS of the molecule leads to negative differential resistance (NDR). Electron transfer between oriented molecules occurs via resonant tunneling. The I-V characteristic for a thin-film of tris (8-hydroxyquinoline)- aluminum (AlQ) molecules, oriented using electric-field poling, and sandwiched between two Al/LiF electrodes is in excellent agreement with theory. This molecular device presents a new paradigm for a convenient, robust, inexpensive alternative to STM or mechanical break-junction structures.Comment: 5 pages, 3 figure

    Pathogenicity test of Western Australian isolates of Sclerotinia sclerotiorum in canola

Hundreds of isolates of S. sclerotiorum have been collected from different canola growing regions of WA. As the majority of WA isolates of S. sclerotiorum have not been analyzed for their genetic characterization, analysis of genetic variation of WA isolates will be undertaken using classical and molecular techniques such as pathogenicity test, mycelial compatibility groups (MCGs), ITS sequencing, and cluster analysis. The experiments which started in March 2013, aim to use classical and molecular tools to identify groups of WA isolates of S. sclerotiorum from which isolates will be selected for the main studies on the management of S. sclerotiorum in canola. Accurate information of genetic diversity through research on characterization of the pathogen will lead to better understanding of the pathogen and will also benefit the breeding programs particularly aiming at breeding for disease resistance and moreover, could lead to developing better techniques for managing the disease.     The use of rapid review methods in health technology assessments: 3 case studies.

    BACKGROUND: Rapid reviews are of increasing importance within health technology assessment due to time and resource constraints. There are many rapid review methods available although there is little guidance as to the most suitable methods. We present three case studies employing differing methods to suit the evidence base for each review and outline some issues to consider when selecting an appropriate method. METHODS: Three recently completed systematic review short reports produced for the UK National Institute for Health Research were examined. Different approaches to rapid review methods were used in the three reports which were undertaken to inform the commissioning of services within the NHS and to inform future trial design. We describe the methods used, the reasoning behind the choice of methods and explore the strengths and weaknesses of each method. RESULTS: Rapid review methods were chosen to meet the needs of the review and each review had distinctly different challenges such as heterogeneity in terms of populations, interventions, comparators and outcome measures (PICO) and/or large numbers of relevant trials. All reviews included at least 10 randomised controlled trials (RCTs), each with numerous included outcomes. For the first case study (sexual health interventions), very diverse studies in terms of PICO were included. P-values and summary information only were presented due to substantial heterogeneity between studies and outcomes measured. For the second case study (premature ejaculation treatments), there were over 100 RCTs but also several existing systematic reviews. Data for meta-analyses were extracted directly from existing systematic reviews with new RCT data added where available. For the final case study (cannabis cessation therapies), studies included a wide range of interventions and considerable variation in study populations and outcomes. A brief summary of the key findings for each study was presented and narrative synthesis used to summarise results for each pair of interventions compared. CONCLUSIONS: Rapid review methods need to be chosen to meet both the nature of the evidence base of a review and the challenges presented by the included studies. Appropriate methods should be chosen after an assessment of the evidence base

    Developing a digital intervention for cancer survivors: an evidence-, theory- and person-based approach

    This paper illustrates a rigorous approach to developing digital interventions using an evidence-, theory- and person-based approach. Intervention planning included a rapid scoping review which identified cancer survivors’ needs, including barriers and facilitators to intervention success. Review evidence (N=49 papers) informed the intervention’s Guiding Principles, theory-based behavioural analysis and logic model. The intervention was optimised based on feedback on a prototype intervention through interviews (N=96) with cancer survivors and focus groups with NHS staff and cancer charity workers (N=31). Interviews with cancer survivors highlighted barriers to engagement, such as concerns about physical activity worsening fatigue. Focus groups highlighted concerns about support appointment length and how to support distressed participants. Feedback informed intervention modifications, to maximise acceptability, feasibility and likelihood of behaviour change. Our systematic method for understanding user views enabled us to anticipate and address important barriers to engagement. This methodology may be useful to others developing digital interventions

    Enhancing crop resilience by harnessing the synergistic effects of biostimulants against abiotic stress

    Plants experience constant exposed to diverse abiotic stresses throughout their growth and development stages. Given the burgeoning world population, abiotic stresses pose significant challenges to food and nutritional security. These stresses are complex and influenced by both genetic networks and environmental factors, often resulting in significant crop losses, which can reach as high as fifty percent. To mitigate the effects of abiotic stresses on crops, various strategies rooted in crop improvement and genomics are being explored. In particular, the utilization of biostimulants, including bio-based compounds derived from plants and beneficial microbes, has garnered considerable attention. Biostimulants offer the potential to reduce reliance on artificial chemical agents while enhancing nutritional efficiency and promoting plant growth under abiotic stress condition. Commonly used biostimulants, which are friendly to ecology and human health, encompass inorganic substances (e.g., zinc oxide and silicon) and natural substances (e.g., seaweed extracts, humic substances, chitosan, exudates, and microbes). Notably, prioritizing environmentally friendly biostimulants is crucial to prevent issues such as soil degradation, air and water pollution. In recent years, several studies have explored the biological role of biostimulants in plant production, focusing particularly on their mechanisms of effectiveness in horticulture. In this context, we conducted a comprehensive review of the existing scientific literature to analyze the current status and future research directions concerning the use of various biostimulants, such as plant-based zinc oxide, silicon, selenium and aminobutyric acid, seaweed extracts, humic acids, and chitosan for enhancing abiotic stress tolerance in crop plants. Furthermore, we correlated the molecular modifications induced by these biostimulants with different physiological pathways and assessed their impact on plant performance in response to abiotic stresses, which can provide valuable insights

    Assessing the perceived impact of post Minamata amalgam phase down on oral health inequalities: a mixed-methods investigation

    Background: Data from countries that have implemented a complete phase out of dental amalgam following the Minamata agreement suggest increased costs and time related to the placement of alternatives with consumers absorbing the additional costs. This aim of this study was to investigate the impact of a complete phase out of dental amalgam on oral health inequalities in particular for countries dependent on state run oral health services. Methods: A mixed methods component design quantitative and qualitative study in the United Kingdom. The quantitative study involved acquisition and analysis of datasets from NHS Scotland to compare trends in placement of dental amalgam and a survey of GDPs in Yorkshire, UK. The qualitative study involved analysis of the free text of the survey and a supplementary secondary analysis of semi-structured interviews and focus groups with GDPs (private and NHS), dental school teaching leads and NHS dental commissioners to understand the impact of amalgam phase down on oral health inequalities. Results: Time-trends for amalgam placement showed that there was a significant (p < 0.05) reduction in amalgam use compared with composites and glass ionomers. However dental amalgam still represented a large proportion (42%) of the restorations (circa 1.8 million) placed in the 2016–2017 financial year. Survey respondents suggest that direct impacts of a phase down were related to increased costs and time to place alternative restorations and reduced quality of care. This in turn would lead to increased tooth extractions, reduced access to care and privatisation of dental services with the greatest impact on deprived populations. Conclusion: Amalgam is still a widely placed material in state run oral health services. The complete phase down of dental amalgam poses a threat to such services and threatens to widen oral health inequalities. Our data suggest that a complete phase out is not currently feasible unless appropriate measures are in place to ensure cheaper, long-lasting and easy to use alternatives are available and can be readily adopted by primary care oral health providers

    Association between routine and standardized blood pressure measurements and left ventricular hypertrophy among patients on hemodialysis

    <p>Abstract</p> <p>Background</p> <p>Left ventricular (LV) hypertrophy is common among patients on hemodialysis. While a relationship between blood pressure (BP) and LV hypertrophy has been established, it is unclear which BP measurement method is the strongest correlate of LV hypertrophy. We sought to determine agreement between various blood pressure measurement methods, as well as identify which method was the strongest correlate of LV hypertrophy among patients on hemodialysis.</p> <p>Methods</p> <p>This was a post-hoc analysis of data from a randomized controlled trial. We evaluated the agreement between seven BP measurement methods: standardized measurement at baseline; single pre- and post-dialysis, as well as mean intra-dialytic measurement at baseline; and cumulative pre-, intra- and post-dialysis readings (an average of 12 monthly readings based on a single day per month). Agreement was assessed using Lin's concordance correlation coefficient (CCC) and the Bland Altman method. Association between BP measurement method and LV hypertrophy on baseline cardiac MRI was determined using receiver operating characteristic curves and area under the curve (AUC).</p> <p>Results</p> <p>Agreement between BP measurement methods in the 39 patients on hemodialysis varied considerably, from a CCC of 0.35 to 0.94, with overlapping 95% confidence intervals. Pre-dialysis measurements were the weakest predictors of LV hypertrophy while standardized, post- and inter-dialytic measurements had similar and strong (AUC 0.79 to 0.80) predictive power for LV hypertrophy.</p> <p>Conclusions</p> <p>A single standardized BP has strong predictive power for LV hypertrophy and performs just as well as more resource intensive cumulative measurements, whereas pre-dialysis blood pressure measurements have the weakest predictive power for LV hypertrophy. Current guidelines, which recommend using pre-dialysis measurements, should be revisited to confirm these results.</p
