    Pulmonary giant cells and their significance for the diagnosis of asphyxiation

    This study was performed to prove whether the detection of polynuclear giant cells in lungs is useful for the diagnosis of asphyxiation due to throttling or strangulation. Therefore, lung specimens of 54 individuals with different natural and unnatural causes of death were investigated. In most lungs examined numerous alveolar macrophages with 1-2 nuclei were found. Polynuclear giant cells, which were arbitrarily defined as alveolar macrophages containing 3 or more nuclei, were observed in all groups investigated except in the cases of hypoxia due to covering the head with plastic bags. Apparent differences between the other groups in particular an increased number in cases of throttling or strangulation, could not be observed. Immunohistochemical investigations confirmed the hypothesis that the observed polynuclear giant cells were derived from alveolar macrophages. The immunohistochemical analysis of the proliferation marker antigen Ki 67 revealed no positive reaction in the nuclei of polynuclear giant cells indicating that these cells had not developed shortly before death by endomitosis as an adaptative change following reduction in oxygen supply. The results provide evidence that the detection of pulmonary polynuclear giant cells cannot be used as a practical indicator for death by asphyxiation due to throttling or strangulation

    Eat or sleep : availability of winter prey explains mid-winter and spring activity in an arctic Calanus population

    Copepods of the genus Calanus have adapted to high levels of seasonality in prey availability by entering a period of hibernation during winter known as diapause, but repeated observations of active Calanus spp. have been made in January in high latitude fjords which suggests plasticity in over-wintering strategies. During the last decade, the period of Polar Night has been studied intensively in the Arctic. A continuous presence of an active microbial food web suggests the prevalence of low-level alternative copepod prey (such as microzooplankton) throughout this period of darkness. Here we provide further evidence of mid-winter zooplankton activity using a decadal record of moored acoustics from Kongsfjorden, Svalbard. We apply an individual based life-history model to investigate the fitness consequences of a range of over-wintering strategies (in terms of diapause timing and duration) under a variety of prey availability scenarios. In scenarios of no winter prey availability ((Formula presented.)), the optimal time to exit diapause is in March. However, as P win increases (up to 40μgCL −1), there is little fitness difference in copepods exiting diapause in January compared to March. From this, we suggest that Calanus are able (in energetic terms) to either i) exit diapause early to deal with uncertainty in spring bloom timing, or ii) remain active throughout winter if diapause is not possible (i.e., environment not deep enough, or not enough lipid reserves built up over the previous summer). The range of viable overwintering strategies increases with increasing P win, suggesting that there is more flexibility for Calanus spp. in a scenario of non-zero P win

    The originally of this church is mainly in its planform and the reinforced concrete roof, in the shape of a hyperbolic paraboloid. In the annex, next to the church, there is a house for the priest, confession boxes, class rooms and other dependencies. Below the church there is a large hall, with a floor area of 619 m2 and two meeting halls, each of 80 m2 area. The church itself has seats for 480 people, and an additional chapel with 70 seats, as well as confession boxes, the baptistry and sacristy. This project is a fine example of how large areas may be roofed over without intermediate supports, at a reasonable price, by means of reinforced concrete shells.Su originalidad consiste en la forma de huso de la planta y, sobre todo, en el paraboloide hiperbólico de hormigón armado que constituye la cubierta de la iglesia. En el edificio anexo están: la casa parroquial, locutorios, aulas, etc. En el de la iglesia y en su nivel inferior se distribuyen: una gran nave de 619 m2 y dos salas de reuniones de unos 80 m2 cada una; en la planta superior se encuentran: la nave destinada al culto, con capacidad para 480 personas sentadas, una capilla con 70 asientos, los confesionarios, el baptisterio y la sacristía.     IL-4Rα Signaling in Keratinocytes and Early IL-4 Production Are Dispensable for Generating a Curative T Helper 1 Response in Leishmania major-Infected C57BL/6 Mice.

    Experimental infection with the protozoan parasite Leishmania major has been extensively used to understand the mechanisms involved in T helper cell differentiation. Following infection, C57BL/6 mice develop a small self-healing cutaneous lesion and they are able to control parasite burden, a process linked to the development of T helper (Th) 1 cells. The local presence of IL-12 has been reported to be critical in driving Th1 cell differentiation. In addition, the early secretion of IL-4 was reported to potentially contribute to Th1 cell differentiation. Following infection with L. major, early keratinocyte-derived IL-4 was suggested to contribute to Th1 cell differentiation. To investigate a putative autocrine role of IL-4 signaling on keratinocytes at the site of infection, we generated C57BL/6 mice deficient for IL-4Rα expression selectively in keratinocytes. Upon infection with L. major, these mice could control their inflammatory lesion and parasite load correlating with the development of Th1 effector cells. These data demonstrate that IL-4 signaling on keratinocytes does not contribute to Th1 cell differentiation. To further investigate the source of IL-4 in the skin during the first days after L. major infection, we used C57BL/6 IL-4 reporter mice allowing the visualization of IL-4 mRNA expression and protein production. These mice were infected with L. major. During the first 3 days after infection, skin IL-4 mRNA expression was observed selectively in mast cells. However, no IL-4 protein production was detectable locally. In addition, early IL-4 blockade locally had no impact on subsequent Th1 cell differentiation and control of the disease. Taken together, the present data rule out a major role for skin IL-4 and keratinocyte IL-4Rα signaling in the development of a Th1 protective immune response following experimental infection with L. major

    Living as a commensal, Candida albicans must adapt and respond to environmental cues generated by the mammalian host and by microbes comprising the natural flora. These signals have opposing effects on C. albicans, with host cues promoting the yeast-to-hyphal transition and bacteria-derived quorum-sensing molecules inhibiting hyphal development. Hyphal development is regulated through modulation of the cyclic AMP (cAMP)/protein kinase A (PKA) signaling pathway, and it has been postulated that quorum-sensing molecules can affect filamentation by inhibiting the cAMP pathway. Here, we show that both farnesol and 3-oxo-C12-homoserine lactone, a quorum-sensing molecule secreted by Pseudomonas aeruginosa, block hyphal development by affecting cAMP signaling; they both directly inhibited the activity of the Candida adenylyl cyclase, Cyr1p. In contrast, the 12-carbon alcohol dodecanol appeared to modulate hyphal development and the cAMP signaling pathway without directly affecting the activity of Cyr1p. Instead, we show that dodecanol exerted its effects through a mechanism involving the C. albicans hyphal repressor, Sfl1p. Deletion of SFL1 did not affect the response to farnesol but did interfere with the response to dodecanol. Therefore, quorum sensing in C. albicans is mediated via multiple mechanisms of action. Interestingly, our experiments raise the possibility that the Burkholderia cenocepacia diffusible signal factor, BDSF, also mediates its effects via Sfl1p, suggesting that dodecanol's mode of action, but not farnesol or 3-oxo-C12-homoserine lactone, may be used by other quorum-sensing molecules

    The time-dependent localization of Ki 67 antigen-positive cells in human skin wounds

    A total of 77 human skin wounds with a post-infliction interval between 3 h and 7 months were investigated and the proliferation marker antigen Ki 67 was visualized in paraffin sections using a specific monoclonal antibody (MIB). The re-built epidermal layer covering the former lesional area showed only a few basal cells positively staining for Ki 67 antigen. No enhanced reactivity was found when compared to uninjured skin. In basal cells of the epidermis adjacent to the wound area, however, varying numbers of positive cells occurred, but no information useful for a reliable time estimation of skin wounds could be obtained due to the considerable variability in the number of Ki 67 positive epidermal basal cells found in non-damaged skin. Fibroblastic cells in the wound area revealed an increased number of Ki 67-positive sites which could first be detected in a 1.5-day-old skin lesion. Positive results could be obtained in every specimen investigated after a post-infliction interval of 6 days up to 1.5 months. Only the scar tissue of the oldest wound examined (wound age 7 months) revealed no increase in the number of positively staining fibroblasts. Therefore, positive results indicate a wound age of at least approximately 1.5 days and the lack of an increased number of positive fibroblastic cells in a sufficient number of specimens indicates at a wound age of less than 6 days, but cannot totally exclude longer post-infliction intervals

    The Arctic Ocean is, on average, the shallowest of Earth’s oceans. Its vast continental shelf areas, which account for approximately half of the Arctic Ocean’s total area, are heavily influenced by the surrounding land masses through river run-off and coastal erosion. As a main area of deep water formation, the Arctic is one of the main «engines» of global ocean circulation, due to large freshwater inputs, it is also strongly stratified. The Arctic Ocean’s complex oceanographic configuration is tightly linked to the atmosphere, the land, and the cryosphere. The physical dynamics not only drive important climate and global circulation patterns, but also control biogeochemical cycles and ecosystem dynamics. Current changes in Arctic sea-ice thickness and distribution, air and water temperatures, and water column stability are resulting in measurable shifts in the properties and functioning of the ocean and its ecosystems. The Arctic Ocean is forecast to shift to a seasonally ice-free ocean resulting in changes to physical, chemical, and biological processes. These include the exchange of gases across the atmosphere-ocean interface, the wind-driven ciruclation and mixing regimes, light and nutrient availability for primary production, food web dynamics, and export of material to the deep ocean. In anticipation of these changes, extending our knowledge of the present Arctic oceanography and these complex changes has never been more urgent

    Pan-Arctic diel vertical migration during the polar night

    Diel vertical migration (DVM) has generally been assumed to cease during the polarnight in the high Arctic, although recent studies have shown the occurrence of lunar vertical migrations (LVMs) and shallow DVMs. Here, we quantified when and where full-depth (>20 m), solar-mediated DVM exists on a pan-Arctic scale. We observed the scattering population, most likely to be comprised of zooplankton, using 300 kHz acoustic Doppler current profilers (ADCPs). We quantified the presence/absence of DVM, and found that DVM continues throughout the year to at least 20 m at all locations south of 74° N. North of 77° N, DVM ceases for a period of time duringthe polar night. The dates of this cessation accurately align with the date of the winter solstice (±2 d). Between 74 and 77° N, DVM presence/absence is variable. Acoustic data sampled at 89° N, however, showed no evidence of DVM at any time during the year — a new observation. Using indicators of presence/absence of sea ice from ADCPs and satellite-derived sea ice concentration data, we revealed that local variations in sea ice cover directly determine the continuation or cessationof DVM during the polar night. Earlier-forming and higher-concentration sea ice causes a cessation in DVM, whereas low-concentration or late-forming sea ice results in continuous DVM when comparing migrations at similar latitudes

    Reversible brain edema associated to flow diverter stent procedures: a retrospective single-center study to evaluate frequency, clinical evolution, and possible mechanism

    Background Hemorrhage and ischemia after flow diverter stent (FDS) procedures for intracranial aneurysms are the most common complications and have been extensively described. Temporary brain edema (TBE) is an unknown complication that could be associated with particular FDS procedures. Objective To estimate the frequency, clinical presentation, imaging findings, and possible mechanisms associating TBE with FDS. Methods Unruptured aneurysms treated with FDS implantation performed in our service from June 2015 to March 2018 were reviewed. Medical antecedents, endovascular procedure, clinical assessments before and after treatment, aneurysm characteristics, and image records were collected. Artery diameters of patients in whom TEB developed were also calculated to investigate any correlation between TBE and anatomic descriptors. Results A total of 179 FDS procedures in 176 patients were reviewed. Six patients (3.4%) presented with symptomatic TBE, and all TBE patients had undergone FDS implantation from the middle cerebral artery (MCA) to the internal carotid artery (ICA). A Pearson product-moment correlation coefficient (PPCC) found smaller MCA diameters and MCA/ICA ratios in these 6 patients (respectively PPCC = −0.619, P < 0.04; PPCC = −0.647, P < 0.03). Hemorrhagic and ischemic complications were less frequent than TBE (2.3% and 1.1% vs. 3.4%). Conclusions TBE was more frequent than ischemic or hemorrhagic complications after FDS in this study. TBE seemed to be associated with a particular FDS positioning in small arteries, inducing flow changes and disruption of the blood–brain barrier
