    Genome profiling of ovarian adenocarcinomas using pangenomic BACs microarray comparative genomic hybridization

    <p>Abstract</p> <p>Background</p> <p>Routine cytogenetic investigations for ovarian cancers are limited by culture failure and poor growth of cancer cells compared to normal cells. Fluorescence <it>in situ </it>Hybridization (FISH) application or classical comparative genome hybridization techniques are also have their own limitations in detecting genome imbalance especially for small changes that are not known ahead of time and for which FISH probes could not be thus designed.</p> <p>Methods</p> <p>We applied microarray comparative genomic hybridization (A-CGH) using one mega base BAC arrays to investigate chromosomal disorders in ovarian adenocarcinoma in patients with familial history.</p> <p>Results</p> <p>Our data on 10 cases of ovarian cancer revealed losses of 6q (4 cases mainly mosaic loss), 9p (4 cases), 10q (3 cases), 21q (3 cases), 22q (4 cases) with association to a monosomy X and gains of 8q and 9q (occurring together in 8 cases) and gain of 12p. There were other abnormalities such as loss of 17p that were noted in two profiles of the studied cases. Total or mosaic segmental gain of 2p, 3q, 4q, 7q and 13q were also observed. Seven of 10 patients were investigated by FISH to control array CGH results. The FISH data showed a concordance between the 2 methods.</p> <p>Conclusion</p> <p>The data suggest that A-CGH detects unique and common abnormalities with certain exceptions such as tetraploidy and balanced translocation, which may lead to understanding progression of genetic changes as well as aid in early diagnosis and have an impact on therapy and prognosis.</p

    The Importance of Urban Eco-gardens for Biodiversity and Human Sustainability: A Case Study from Palestine

    The situation in Palestine is of concern where global threats of climate change, overexploitation, habitat destruction, invasive species, and pollution are compounded by occupation and conflict. Thus, almost 1/3rd of vascular plant species are rare and over 50 are listed as endangered or rare based on their abundance and presence in grids studied earlier. Here, we describe the development of a conservation botanic garden that works via research, education, and direct in situ and ex situ conservation of plant species. The garden now boasts 381 species of vascular plants (63 are rare). The team scientifically and selectively introduced some rare and endemic species, developed educational modules, and engaged the community in all aspects of planning and growth of this garden. It has acted as a model for threatened and protected areas in the state of Palestine, as well as becoming a national oasis for both wildlife and humans. The lessons learned from this experience include: 1) principles of minimal intervention in eco-friendly ways producing zones of permaculture and gardening towards conservation (ex situ and in situ conservation), while allowing botanic garden functionality, 2) involvement of staff, volunteers, experts, and community in education and conservation efforts, 3) value of research in plants and animals for integrated ecosystem management. The outcome of this work is a maximally utilitarian garden for areas like education, direct conservation, research, and human satisfaction whilst ensuring long-term sustainability in a nascent state in the midst of a difficult political situation

    Flora and Conservation Issues in Two Protected Areas in Palestine: Wadi Al-Zarqa Al-Ulwi and Wadi Qana

    The Palestinian environment suffered from abuse and neglect for decades. As a nascent state, Palestine aspired to protect and restore its environment, in line with international standards and conventions. Yet, many challenges remain, including a lack of data, the science&ndash;policy&ndash;practice gaps, and the lack of sovereignty. The management of protected areas (PAs) is particularly challenging under these circumstance. Two key protected areas in the northern West Bank (Wadi Qana and Wadi Al-Zarqa Al-Ulwi) were surveyed, with a focus on plant communities and dealing with threats. The two areas had similar Mediterranean flora with differences, including in rare plants. Both areas have important but different wetlands in their buffer zones. The data show that the protection of the two areas is important because of the differences noted, and both areas are valued as IUCN category IV (habitat/species management area) protection. Such protection is possible by working with local communities and bridging the science&ndash;policy&ndash;practice gaps
