    Calliffusion: Chinese Calligraphy Generation and Style Transfer with Diffusion Modeling

    In this paper, we propose Calliffusion, a system for generating high-quality Chinese calligraphy using diffusion models. Our model architecture is based on DDPM (Denoising Diffusion Probabilistic Models), and it is capable of generating common characters in five different scripts and mimicking the styles of famous calligraphers. Experiments demonstrate that our model can generate calligraphy that is difficult to distinguish from real artworks and that our controls for characters, scripts, and styles are effective. Moreover, we demonstrate one-shot transfer learning, using LoRA (Low-Rank Adaptation) to transfer Chinese calligraphy art styles to unseen characters and even out-of-domain symbols such as English letters and digits.Comment: 5pages, International Conference on Computational Creativity, ICC

    BmILF and I-motif Structure Are Involved in Transcriptional Regulation of \u3cem\u3eBmPOUM2\u3c/em\u3e in \u3cem\u3eBombyx mori\u3c/em\u3e

    Guanine-rich and cytosine-rich DNA can form four-stranded DNA secondary structures called G-quadruplex (G4) and i-motif, respectively. These structures widely exist in genomes and play important roles in transcription, replication, translation and protection of telomeres. In this study, G4 and i-motif structures were identified in the promoter of the transcription factor gene BmPOUM2, which regulates the expression of the wing disc cuticle protein gene (BmWCP4) during metamorphosis. Disruption of the i-motif structure by base mutation, anti-sense oligonucleotides (ASOs) or inhibitory ligands resulted in significant decrease in the activity of the BmPOUM2 promoter. A novel i-motif binding protein (BmILF) was identified by pull-down experiment. BmILF specifically bound to the i-motif and activated the transcription of BmPOUM2. The promoter activity of BmPOUM2 was enhanced when BmILF was over-expressed and decreased when BmILF was knocked-down by RNA interference. This study for the first time demonstrated that BmILF and the i-motif structure participated in the regulation of gene transcription in insect metamorphosis and provides new insights into the molecular mechanism of the secondary structures in epigenetic regulation of gene transcription

    Localized High-Concentration Electrolytes Get More Localized Through Micelle-Like Structures

    Liquid electrolytes in batteries are typically treated as macroscopically homogeneous ionic transport media despite having complex chemical composition and atomistic solvation structures, leaving a knowledge gap of microstructural characteristics. Here, we reveal a unique micelle-like structure in a localized high-concentration electrolyte (LHCE), in which the solvent acts as a surfactant between an insoluble salt in diluent. The miscibility of the solvent with the diluent and simultaneous solubility of the salt results in a micelle-like structure with a smeared interface and an increased salt concentration at the centre of the salt-solvent clusters that extends the salt solubility. These intermingling miscibility effects have temperature dependencies, wherein an exemplified LHCE peaks in localized cluster salt concentration near room temperature and is utilized to form a stable solid-electrolyte interphase (SEI) on Li-metal anode. These findings serve as a guide to predicting a stable ternary phase diagram and connecting the electrolyte microstructure with electrolyte formulation and formation protocols to form stable SEI for enhanced battery cyclability

    GPX8 regulates pan-apoptosis in gliomas to promote microglial migration and mediate immunotherapy responses

    IntroductionGliomas have emerged as the predominant brain tumor type in recent decades, yet the exploration of non-apoptotic cell death regulated by the pan-optosome complex, known as pan-apoptosis, remains largely unexplored in this context. This study aims to illuminate the molecular properties of pan-apoptosis-related genes in glioma patients, classifying them and developing a signature using machine learning techniques.MethodsThe prognostic significance, mutation features, immunological characteristics, and pharmaceutical prediction performance of this signature were comprehensively investigated. Furthermore, GPX8, a gene of interest, was extensively examined for its prognostic value, immunological characteristics, medication prediction performance, and immunotherapy prediction potential. ResultsExperimental techniques such as CCK-8, Transwell, and EdU investigations revealed that GPX8 acts as a tumor accelerator in gliomas. At the single-cell RNA sequencing level, GPX8 appeared to facilitate cell contact between tumor cells and macrophages, potentially enhancing microglial migration. ConclusionsThe incorporation of pan-apoptosis-related features shows promising potential for clinical applications in predicting tumor progression and advancing immunotherapeutic strategies. However, further in vitro and in vivo investigations are necessary to validate the tumorigenic and immunogenic processes associated with GPX8 in gliomas

    Modeling of Alpine Grassland Cover Based on Unmanned Aerial Vehicle Technology and Multi-Factor Methods: A Case Study in the East of Tibetan Plateau, China

    Grassland cover and its temporal changes are key parameters in the estimation and monitoring of ecosystems and their functions, especially via remote sensing. However, the most suitable model for estimating grassland cover and the differences between models has rarely been studied in alpine meadow grasslands. In this study, field measurements of grassland cover in Gannan Prefecture, from 2014 to 2016, were acquired using unmanned aerial vehicle (UAV) technology. Single-factor parametric and multi-factor parametric/non-parametric cover inversion models were then constructed based on 14 factors related to grassland cover, and the dynamic variation of the annual maximum cover was analyzed. The results show that (1) nine out of 14 factors (longitude, latitude, elevation, the concentrations of clay and sand in the surface and bottom soils, temperature, precipitation, enhanced vegetation index (EVI) and normalized difference vegetation index (NDVI)) exert a significant effect on grassland cover in the study area. The logarithmic model based on EVI presents the best performance, with an R2 and RMSE of 0.52 and 16.96%, respectively. Single-factor grassland cover inversion models account for only 1–49% of the variation in cover during the growth season. (2) The optimum grassland cover inversion model is the artificial neural network (BP-ANN), with an R2 and RMSE of 0.72 and 13.38%, and SDs of 0.062% and 1.615%, respectively. Both the accuracy and the stability of the BP-ANN model are higher than those of the single-factor parametric models and multi-factor parametric/non-parametric models. (3) The annual maximum cover in Gannan Prefecture presents an increasing trend over 60.60% of the entire study area, while 36.54% is presently stable and 2.86% exhibits a decreasing trend

    Targeting MYH9 represses USP14-mediated NAP1L1 deubiquitination and cell proliferation in glioma

    Abstract Myosin heavy chain 9 (MYH9) plays an important role in a number of diseases. Nevertheless, the function of MYH9 in glioma is unclear. The present research aimed to investigate the role of MYH9 in glioma and determine whether MYH9 is involved in the temozolomide chemoresistance of glioma cells. Our results showed that MYH9 increased the proliferation and temozolomide resistance of glioma cells. The mechanistic experiments showed that the binding of MYH9 to NAP1L1, a potential promoter of tumor proliferation, inhibited the ubiquitination and degradation of NAP1L1 by recruiting USP14. Upregulation of NAP1L1 increased its binding with c-Myc and activated c-Myc, which induced the expression of CCND1/CDK4, promoting glioma cell temozolomide resistance and proliferation. Additionally, we found that MYH9 upregulation was strongly related to patient survival and is therefore a negative factor for patients with glioma. Altogether, our results show that MYH9 plays a role in glioma progression by regulating NAP1L1 deubiquitination. Thus, targeting MYH9 is a potential therapeutic strategy for the clinical treatment of glioma in the future

    Molecular pathology and clinical treatment of independent HPV primary serous carcinoma of the uterine cervix (USCC): A case report

    Abstract On October 23, 2020, a 69‐year‐old Chinese female patient was admitted to Yuncheng Hospital due to a history of postmenopausal bleeding and lower abdominal pain for 5 months. The HPV test and pathology results indicated the presence of independent HPV in primary serous carcinoma of the uterine cervix. The genetic testing identified variants of uncertain significance (PAX8 p.Tyr 410 Ter and TP53 p.Asn 247 Ile), microsatellite instability stable (MSI‐S), tumor mutational burden (TMB) 7.33Muts/Mb, and an elevated tumor neoantigen burden. Before undergoing radical hysterectomy treatment, the patient exhibited a positive response to three cycles of intravenous docetaxel (100 mg/3 h) and carboplatin (450 mg/1 h). Following the surgery, she received an additional three cycles of docetaxel (100 mg/3 h) and carboplatin (500 mg/1 h), accompanied by 25 cycles of radiation therapy (DT 46Gy/2Gy/23f). Concurrently, cisplatin (450 mg/1 h) was administered. As of now, the patient has achieved 20 months of disease‐free survival

    Somatic cells can be reprogrammed into induced pluripotent stem cells (iPSCs) in vitro. Previously, a lentivirus induction strategy of introducing Oct4, Sox2, Nanog and Lin28 (OSNL) into the iPSC process has been shown as a possible way to produce chicken iPSCs from chicken embryonic fibroblasts, but the induction efficiency of this method was found to be significantly limiting. In order to help resolve this efficiency obstacle, this study seeks to clarify the associated regulation mechanisms and optimizes the reprogramming strategy of chicken iPSCs. This study showed that glycolysis and the expression of glycolysis-related genes correlate with a more efficient reprogramming process. At the same time, the transcription factors Oct4, Sox2 and Nanog were found to activate the expression of glycolysis-related genes. In addition, we introduced two small-molecule inhibitors (2i-SP) as a “glycolysis activator” together with the OSNL cocktail, and found that this significantly improved the induction efficiency of the iPSC process. As such, the study identifies direct molecular connections between core pluripotency factors and glycolysis during the chicken iPSC induction process and, with its results, provides a theoretical basis and technical support for chicken somatic reprogramming
