    Isolation and complete genomic characterization of H1N1 subtype swine influenza viruses in southern China through the 2009 pandemic

    <p>Abstract</p> <p>Background</p> <p>The swine influenza (SI) is an infectious disease of swine and human. The novel swine-origin influenza A (H1N1) that emerged from April 2009 in Mexico spread rapidly and caused a human pandemic globally. To determine whether the tremendous virus had existed in or transmitted to pigs in southern China, eight H1N1 influenza strains were identified from pigs of Guangdong province during 2008-2009.</p> <p>Results</p> <p>Based on the homology and phylogenetic analyses of the nucleotide sequences of each gene segments, the isolates were confirmed to belong to the classical SI group, with HA, NP and NS most similar to 2009 human-like H1N1 influenza virus lineages. All of the eight strains were low pathogenic influenza viruses, had the same host range, and not sensitive to class of antiviral drugs.</p> <p>Conclusions</p> <p>This study provides the evidence that there is no 2009 H1N1-like virus emerged in southern China, but the importance of swine influenza virus surveillance in China should be given a high priority.</p

    Trends in cognitive function before and after myocardial infarction: findings from the China Health and Retirement Longitudinal Study

    ObjectivesIncident stroke was associated with cognitive dysfunction after stroke and even before stroke. However, cognitive trends prior to myocardial infarction (MI) and the timeline of cognitive decline in a few years following incident MI remain unclear, especially among the Chinese population. We aimed to evaluate whether MI was associated with cognitive change both before and after MI in China.MethodsThis cohort study included 11,287 participants without baseline heart problems or stroke from the China Health and Retirement Longitudinal Study. The exposure was self-reported MI. The outcomes were scores of cognitive functions in five domains, which reflected abilities of episodic memory, visuospatial abilities, orientation, attention and calculation, and global cognition as a summary measure. A Linear mixed model was constructed to explore cognitive function before and after incident MI among the MI participants and the cognitive trends of participants free of MI.ResultsDuring the 7-year follow-up, 421 individuals [3.7% of 11,287, mean (SD) age, 60.0 (9.0) years; 59.1% female] experienced MI events. The cognitive scores of participants of both the MI group and the control group without MI declined gradually as time went by. The annual decline rate of the MI group before incident MI was similar to that of the control group during the whole follow-up period. Incident MI was not associated with acute cognitive decline in all five cognitive domains. Moreover, MI did not accelerate the cognitive decline rate after MI compared with the pre-MI cognitive trends. The decline rate of cognitive function after MI was similar to the rate before MI.ConclusionsDifferent from stroke, participants who had an MI did not show steeper cognitive decline before MI. MI was not associated with acute cognitive decline and accelerated decline in several years after MI. Future studies are needed to learn the mechanisms behind the different patterns of cognitive decline between MI and stroke

    Epidemiology and immunoprotection of nephropathogenic avian infectious bronchitis virus in southern China

    <p>Abstract</p> <p>Background</p> <p>In last three years, 96 suspected poultry farms from different provinces in China were diagnosed for avian infectious bronchitis (IB) survey. Finally, 221 IBV strains were confirmed by dwarf embryo test and RT-PCR assay. By virus recovery trials, 187 of the isolates caused the birds died or distressed from nephritis, which was accordant with the clinical record.</p> <p>Results</p> <p>Based on epidemiology analysis of recent field isolates of nephropathogenic IB in vaccinated farms in China, YL6 strain were used for vaccination and evaluated by antibody titer and challenge tests. The immunoprotection test indicated that the practical application of vaccine based on the recent field strains could finely facilitate controlling the nephropathogenic IB.</p> <p>Conclusions</p> <p>Our study was aim at setting a guide for safeguard against nephropathogenic IBV-caused disease in China.</p

    The Effect of the Antimicrobial Peptide Plectasin on the Growth Performance, Intestinal Health, and Immune Function of Yellow-Feathered Chickens

    The goal of the study was to test the effects of an antibiotic substitute, plectasin, on the growth performance, immune function, intestinal morphology and structure, intestinal microflora, ileal mucosal layer construction and tight junctions, ileal immune-related cytokines, and blood biochemical indices of yellow-feathered chickens. A total of 1,500 one-day-old yellow-feathered chicks were randomly divided into four dietary treatment groups with five replicates in each group and 75 yellow-feathered chicks in each replication, as follows: basal diet (group A); basal diet supplemented with 10 mg enramycin/kg of diet (group B), basal diet supplemented with 100 mg plectasin/kg of diet (group C), and basal diet supplemented with 200 mg plectasin/kg of diet (group D). It was found that the dietary antimicrobial peptide plectasin could improve the ADG and had better F/G for the overall period of 1–63 days. Dietary plectasin can enhance H9N2 avian influenza virus (AIV) and Newcastle disease virus (NDV) antibody levels of yellow-feathered chickens at 21, and 35 days of age. Dietary plectasin can enhance the intestine structure, inhibit Escherichia coli and proinflammatory cytokines in the ileum, and ameliorate the blood biochemical indices of yellow-feathered chickens at 21 days of age. This study indicates that the antimicrobial peptide plectasin has beneficial effects on the growth performance, intestinal health and immune function of yellow-feathered chickens

    Sources of nitrate in a heavily nitrogen pollution bay in Beibu Gulf, as identified using stable isotopes

    Eutrophication, mainly caused by the oversupply of inorganic nitrogen and phosphate, has increased and become a serious environmental problem in the coastal bays of Beibu Gulf, a newly developing industry and port in South China. However, the sources of nitrate are poorly understood in the gulf. In this study, nitrate dual isotopes (δ15N-NO3- and δ18O-NO3-) and ammonium isotopes (δ15N-NH4+) were measured during the rainy season to identify the nitrate sources and elucidate their biogeochemical processes in Xi Bay, a semi-enclosed bay that is strongly affected by human activities in the Beibu Gulf. The results showed that a high dissolved inorganic nitrogen (DIN, 10.24-99.09 ¾mol L-1) was observed in Xi Bay, particularly in the bay mouth. The concentrations of DIN in the bay were 1.5 times higher than that in Qinzhou Bay and 1.7 times than that in Tieshangang Bay, which mainly influenced by the intensive human activities (i.e., industrial and port activities). In addition, lower values of δ15N-NO3- and δ18O-NO3- and higher values of δ15N-NH4+ were observed in the upper bay, suggesting that microbial nitrification occurs in the upper bay, which was the dominant nitrate source in the upper bay (39%). In addition to nitrification, external sources, including sewage and manure (33%), soil N (15%) and fertilizer (11%), contributed to the higher nutrients in the upper bay. In the lower bay, severe nitrogen pollution led to a weaker impact of biological processes on isotopic fractionation, although a high Chl a level (average of 7.47 ¾g L-1) was found in this region. The heavy nitrate pollution in the lower bay mainly originated from sewage and manure (54%), followed by soil N (26%) and fertilizer (17%). The contribution of the nitrate source from atmospheric deposition was relatively low in the bay (&lt;3%). This study suggests that biogeochemical processes have little impact on nitrate dual isotopes under heavy nitrogen pollution, and isotopes are an ideal proxy for tracing nitrogen sources

    Phylogenetic distribution and predominant genotype of the avian infectious bronchitis virus in China during 2008-2009

    <p>Abstract</p> <p>Background</p> <p>The nephropathogenic avian infectious bronchitis (IB) caused unprecedented economic losses to the commercial chicken industry of China in 2008-2009. To investigate the prevalence of nephropathogenic IB in China, eighty IBV isolates from different provinces during 2008-2009 were identified by dwarf embryo test and RT-PCR.</p> <p>Results</p> <p>The strains were mostly isolated in winter and spring with a wide age range of IB outbreaks, from 4 to 69 days. By the virus recovery trials, 70/80 of the strains resulted in the deaths or distresses of birds from nephritis. To learn more about the molecular evolutionary characteristics of the circulating field strains, the coding region of major spike 1 (S1) protein gene of these strains was RT-PCR amplified and sequenced. Compared to the published representative strains, nucleotides and amino acids sequence analysis indicated that the S1 genes of these strains and the reference strains displayed homologies ranging from 75.1% to 99.8% and from 73.1% to 99.8% respectively. S1 protein of the major pandemic strains contained 540 or 542 amino acids with the cleavage site of HRRRR or RRFRR. Phylogenetic analysis revealed that recent field isolates of IBV in China were mostly belonged to A2-branch (QXIBV-branch) and HN08-branch, only one isolate was belonged to Gray-branch and M41-branch respectively. Most of the 80 strains showed evolutionarily distant from vaccine strains.</p> <p>Conclusions</p> <p>The results of this study suggested that nephropathogenic IBVs were mainly A2-like strains in China during 2008-2009.</p

    cDNA-AFLP analysis reveals differential gene expression in compatible interaction of wheat challenged with Puccinia striiformis f. sp. tritici

    <p>Abstract</p> <p>Background</p> <p><it>Puccinia striiformis </it>f. sp. <it>tritici </it>is a fungal pathogen causing stripe rust, one of the most important wheat diseases worldwide. The fungus is strictly biotrophic and thus, completely dependent on living host cells for its reproduction, which makes it difficult to study genes of the pathogen. In spite of its economic importance, little is known about the molecular basis of compatible interaction between the pathogen and wheat host. In this study, we identified wheat and <it>P. striiformis </it>genes associated with the infection process by conducting a large-scale transcriptomic analysis using cDNA-AFLP.</p> <p>Results</p> <p>Of the total 54,912 transcript derived fragments (TDFs) obtained using cDNA-AFLP with 64 primer pairs, 2,306 (4.2%) displayed altered expression patterns after inoculation, of which 966 showed up-regulated and 1,340 down-regulated. 186 TDFs produced reliable sequences after sequencing of 208 TDFs selected, of which 74 (40%) had known functions through BLAST searching the GenBank database. Majority of the latter group had predicted gene products involved in energy (13%), signal transduction (5.4%), disease/defence (5.9%) and metabolism (5% of the sequenced TDFs). BLAST searching of the wheat stem rust fungus genome database identified 18 TDFs possibly from the stripe rust pathogen, of which 9 were validated of the pathogen origin using PCR-based assays followed by sequencing confirmation. Of the 186 reliable TDFs, 29 homologous to genes known to play a role in disease/defense, signal transduction or uncharacterized genes were further selected for validation of cDNA-AFLP expression patterns using qRT-PCR analyses. Results confirmed the altered expression patterns of 28 (96.5%) genes revealed by the cDNA-AFLP technique.</p> <p>Conclusion</p> <p>The results show that cDNA-AFLP is a reliable technique for studying expression patterns of genes involved in the wheat-stripe rust interactions. Genes involved in compatible interactions between wheat and the stripe rust pathogen were identified and their expression patterns were determined. The present study should be helpful in elucidating the molecular basis of the infection process, and identifying genes that can be targeted for inhibiting the growth and reproduction of the pathogen. Moreover, this study can also be used to elucidate the defence responses of the genes that were of plant origin.</p

    Research Progress on Immunomodulatory Activity of Polysaccharides from Blue Foods

    With the current over-exploitation of terrestrial resources, multi-dimensional in-depth utilization of aquatic resources has attracted widespread attention. Blue foods including marine animals and algae supply nutrients to 3.2 billion people around the world. Polysaccharides are an important class of nutritional components in “blue foods”, and have various biological activities such as antitumor, anti-inflammatory, and anti-allergic functions. The exertion of these physiological functions is closely related to their immunomodulatory activities. Therefore, this article reviews recent progress on the classification, composition and immunomodulatory activities of polysaccharides in blue foods with a focus on the regulatory effects of polysaccharides in marine mollusks and seaweeds on innate/adaptive immune responses, the antitumor effects of pro-inflammatory polysaccharides, and their role in immune adjuvants, and the role of anti-inflammatory polysaccharides in autoimmune diseases and inflammation in an effort to enrich the understanding of the different immune effects of marine polysaccharides and to provide theoretical support for the development of immunomodulatory functional foods based on polysaccharides from blue foods
