    Geological Control on Permeability Heterogeneity in Deeply Buried Dolomite Rocks

    Geological deposition and diagenesis in carbonate rocks are strongly affected by sea level fluctuation, which can result in significant heterogeneity in pore types, elastic properties, and permeability. In this dissertation research, the mechanism of dolomitization of deeply buried carbonate reservoirs in Sichuan Basin, China is investigated in comparison with shallow carbonate microbialite in Alabama, USA. Based on rock physics, geological factors controlling elastic properties and permeability heterogeneity of carbonate rocks are further studied quantitatively, integrating core, well log and seismic data. Core and log measurements were first integrated to estimate pore type variation within the Feixianguan carbonate reservoirs in Sichuan Basin, China. The results show that cementation factor (m) from Archie’s equation and frame flexibility factor (γμ) from a rock physics model are the most effective pore structure indicators for distinguishing diagenetic pore types of the recrystallized rocks, and thus, for classifying permeability-porosity relationships. The variation of γμ is then applied to correlate pore type variations with sea level cyclicities of different orders. The results show that a general trend of γμ curve correlates with eustatic sea level cycle, while the two prominent changes of the curve differentiated the three relative sea level cycles by recognizing the episodic subaerial dissolution. Each cycle is characterized by the initial development of moldic porosity (γμ 4) with decreasing pore size and increasing γμ. The pore type evolution corresponds to the process of seepage refluxing associated with sea level regression. Further integrated analysis of γμ variation, and δ^18 O and δ^13 C isotopes, cathodoluminescence images, confirms that reflux dolomitization played a key role in the studied Feixianguan Formation. Dolomitization started at shallow tidal flat or lagoon in the form of protodolomite by microbial dolomitization, which continued to grow following the seepage reflux of sea water. After the burial, the early dolomite was further recrystallized. Seismic inversion is also applied to estimate permeability, using the different permeability-porosity relations constrained by γμ from the core and log analysis.     Microscale Investigation of Arsenic Distribution and Species in Cement Product from Cement Kiln Coprocessing Wastes

    To improve the understanding of the immobilization mechanism and the leaching risk of Arsenic (As) in the cement product from coprocessing wastes using cement kiln, distribution and species of As in cement product were determined by microscale investigation methods, including electron probe microanalysis (EPMA) and X-ray absorption spectroscopy. In this study, sodium arsenate crystals (Na3AsO412H2O) were mixed with cement production raw materials and calcined to produce cement clinker. Then, clinker was mixed water to prepare cement paste. EPMA results showed that As was generally distributed throughout the cement paste. As content in calcium silicate hydrates gel (C-S-H) was in low level, but higher than that in other cement mineral phases. This means that most of As is expected to form some compounds that disperse on the surfaces of cement mineral phases. Linear combination fitting (LCF) of the X-ray absorption near edge structure spectra revealed that As in the cement paste was predominantly As(V) and mainly existed as Mg3(AsO4)2, Ca3(AsO4)2, and Na2HAsO4

    Infection of hepatitis B virus in extrahepatic endothelial tissues mediated by endothelial progenitor cells

    BACKGROUND: Hepatitis B virus (HBV) replication has been reported to be involved in many extrahepatic viral disorders; however, the mechanism by which HBV is trans-infected into extrahepatic tissues such as HBV associated myocarditis remains largely unknown. RESULTS: In this study, we showed that human cord blood endothelial progenitor cells (EPCs), but not human umbilical vein endothelial cells (HUVECs) could be effectively infected by uptake of HBV in vitro. Exposure of EPCs with HBV resulted in HBV DNA and viral particles were detected in EPCs at day 3 after HBV challenge, which were peaked around day 7 and declined in 3 weeks. Consistently, HBV envelope surface and core antigens were first detected in EPCs at day 3 after virus challenge and were retained to be detectable for 3 weeks. In contrast, HBV covalently closed circular DNA was not detected in EPCs at any time after virus challenge. Intravenous transplantation of HBV-treated EPCs into myocardial infarction and acute renal ischemia mouse model resulted in incorporation of HBV into injured heart, lung, and renal capillary endothelial tissues. CONCLUSION: These results strongly support that EPCs serve as virus carrier mediating HBV trans-infection into the injured endothelial tissues. The findings might provide a novel mechanism for HBV-associated myocarditis and other HBV-related extrahepatic diseases as well

    Utjecaj sastava podloge na povećanje biomase micelija i proizvodnje egzopolimera s pomoću gljive Hericium erinaceus CZ-2

    In this work, the effects of medium composition and fermentation parameters on the simultaneous production of mycelial biomass and exopolymer by medicinal mushroom Hericium erinaceus CZ-2 were investigated in shake flask cultures using one-factor-at-a-time method and orthogonal array design. Results showed that the most suitable carbon, nitrogen, mineral sources, and cofactors for the mycelial biomass and exopolymer production were: corn flour combined with 1 % glucose, yeast extract, KH2PO4 and corn steep liquor. The intuitive analysis of orthogonal array design results indicated that the effects of nutritional requirement on the mycelial growth of Hericium erinaceus CZ-2 were in regular sequence of corn flour combined with 1 % glucose > yeast extract > corn steep liquor > KH2PO4, and those on exopolymer production were in the order of corn flour combined with glucose > KH2PO4 > yeast extract > corn steep liquor. The maximal yield of mycelial biomass (16.07 g/L) was obtained when the composition of the culture medium was (in g/L): corn flour 30, glucose 10, yeast extract 3, KH2PO4 1, CaCO3 0.5, and 15 mL/L of corn steep liquor; while the maximal exopolymer yield (1.314 g/L) was achieved when the composition of medium was (in g/L): corn flour 30, glucose 10, yeast extract 5, KH2PO4 3, CaCO3 0.5, and 15 mL/L of corn steep liquor. In the 15-litre scale-up fermentation, the maximum mycelial biomass yield of 20.50 g/L was achieved using the optimized medium.U ovom je radu ispitan utjecaj sastava podloge i uvjeta fermentacije na istodobnu proizvodnju biomase micelija i egzopolimera s pomoću ljekovite gljive Hericium erinaceus CZ-2. Pokusi su provedeni na kulturama uzgojenim na tresilici, primjenom jednofaktorske metode i ortogonalnog modela. Rezultati su pokazali da su najbolji izvori dušika, minerala i kofaktora za proizvodnju biomase micelija i egzopolimera kukuruzno brašno s 1 % glukoze, ekstrakt kvasca, KH2PO4 i kukuruzni ekstrakt. Analizom ortogonalnog modela utvrđeno je da na povećanje micelija redom utječu: kukuruzno brašno s 1 % glukoze, zatim ekstrakt kvasca, kukuruzni ekstrakt i KH2PO4, a na proizvodnju egzopolimera kukuruzno brašno s glukozom, zatim KH2PO4, ekstrakt kvasca te kukuruzni ekstrakt. Najveći je prinos biomase micelija (16,07 g/L) postignut s ovim sastavom podloge (u g/L): kukuruzno brašno 30, glukoza 10, ekstrakt kvasca 3, KH2PO4 1, CaCO3 0,5 i 15 mL/L kukuruznog ekstrakta, a najveći je prinos egzopolimera (1,314 g/L) dobiven korištenjem podloge (u g/L) od: kukuruznog brašna 30, glukoze 10, ekstrakta kvasca 5, KH2PO4 3, CaCO3 0,5 i 15 mL/L kukuruznog ekstrakta. Povećanjem obujma fermentacije na 15 L dobiven je najveći prinos biomase micelija od 20,50 g/L pri optimalnim uvjetima

    Genome-wide association study on serum alkaline phosphatase levels in a Chinese population

    Background: Serum alkaline phosphatase (ALP) is a complex phenotype influenced by both genetic and environmental factors. Recent Genome-Wide Association Studies (GWAS) have identified several loci affecting ALP levels; however, such studies in Chinese populations are limited. We performed a GWAS analyzing the association between 658,288 autosomal SNPs and serum ALP in 1,461 subjects, and replicated the top SNPs in an additional 8,830 healthy Chinese Han individuals. The interactions between significant locus and environmental factors on serum ALP levels were further investigated. Results: The association between ABO locus and serum ALP levels was replicated (P = 2.50 × 10-21, 1.12 × 10-56 and 2.82 × 10-27 for SNP rs8176720, rs651007 and rs7025162 on ABO locus, respectively). SNP rs651007 accounted for 2.15% of the total variance of serum ALP levels independently of the other 2 SNPs. When comparing our findings with previously published studies, ethnic differences were observed across populations. A significant interaction between ABO rs651007 and overweight and obesity was observed (FDR for interaction was 0.036); for individuals with GG genotype, those with normal weight and those who were overweight or obese have similar serum ALP concentrations; minor allele A of rs651007 remarkably reduced serum ALP levels, but this effect was attenuated in overweight and obese individuals. Conclusions: Our findings indicate that ABO locus is a major determinant for serum ALP levels in Chinese Han population. Overweight and obesity modifies the effect of ABO locus on serum ALP concentrations

    The 5p15.33 Locus Is Associated with Risk of Lung Adenocarcinoma in Never-Smoking Females in Asia

    Genome-wide association studies of lung cancer reported in populations of European background have identified three regions on chromosomes 5p15.33, 6p21.33, and 15q25 that have achieved genome-wide significance with p-values of 10−7 or lower. These studies have been performed primarily in cigarette smokers, raising the possibility that the observed associations could be related to tobacco use, lung carcinogenesis, or both. Since most women in Asia do not smoke, we conducted a genome-wide association study of lung adenocarcinoma in never-smoking females (584 cases, 585 controls) among Han Chinese in Taiwan and found that the most significant association was for rs2736100 on chromosome 5p15.33 (p = 1.30×10−11). This finding was independently replicated in seven studies from East Asia totaling 1,164 lung adenocarcinomas and 1,736 controls (p = 5.38×10−11). A pooled analysis achieved genome-wide significance for rs2736100. This SNP marker localizes to the CLPTM1L-TERT locus on chromosome 5p15.33 (p = 2.60×10−20, allelic risk = 1.54, 95% Confidence Interval (CI) 1.41–1.68). Risks for heterozygote and homozygote carriers of the minor allele were 1.62 (95% CI; 1.40–1.87), and 2.35 (95% CI: 1.95–2.83), respectively. In summary, our results show that genetic variation in the CLPTM1L-TERT locus of chromosome 5p15.33 is directly associated with the risk of lung cancer, most notably adenocarcinoma
