    Serine 129 Phosphorylation of α-Synuclein Cross-Links with Tissue Transglutaminase to Form Lewy Body-Like Inclusion Bodies

    Intraneuronal depositions of α-synuclein have been implicated in the pathogenesis of Parkinsons's disease (PD). Previous reports have identified the crosslinking between α-synuclein and tTG (tissue transglutaminase) in both PD patients and the cellular model. However, no researches have been conducted to further investigate their interaction in physiological conditions. To address this question, we generated the SH-SY5Y cell line which stably expressed the wild-type or mutant (Ser129Ala) α-synuclein. After the treatment with okadaic acid, α-synuclein started forming aggregates upon the activation of tTG. Coimmunoprecipitation assays revealed a decreased interaction of the mutant α-synuclein S129A with tTG compared with the wild-type α-synuclein. Cells expressing the wild-type α-synuclein showed increased eosinophilic cytoplasmic inclusion bodies that resembled Lewy bodies compared with the mutant. Double immunofluorescence staining confirmed the colocalization of the phosphorylated α-synuclein and the tTG in the cells. The S129A mutant demonstrated a lesser degree of colocalization than the wild type

    Hemophilia a patients with inhibitors: Mechanistic insights and novel therapeutic implications

    The development of coagulation factor VIII (FVIII) inhibitory antibodies is a serious complication in hemophilia A (HA) patients after FVIII replacement therapy. Inhibitors render regular prophylaxis ineffective and increase the risk of morbidity and mortality. Immune tolerance induction (ITI) regimens have become the only clinically proven therapy for eradicating these inhibitors. However, this is a lengthy and costly strategy. For HA patients with high titer inhibitors, bypassing or new hemostatic agents must be used in clinical prophylaxis due to the ineffective ITI regimens. Since multiple genetic and environmental factors are involved in the pathogenesis of inhibitor generation, understanding the mechanisms by which inhibitors develop could help identify critical targets that can be exploited to prevent or eradicate inhibitors. In this review, we provide a comprehensive overview of the recent advances related to mechanistic insights into anti-FVIII antibody development and discuss novel therapeutic approaches for HA patients with inhibitors

    In vitro expression and analysis of the 826 human G protein-coupled receptors

    ABSTRACT G protein-coupled receptors (GPCRs) are involved in all human physiological systems where they are responsible for transducing extracellular signals into cells. GPCRs signal in response to a diverse array of stimuli including light, hormones, and lipids, where these signals affect downstream cascades to impact both health and disease states. Yet, despite their importance as therapeutic targets, detailed molecular structures of only 30 GPCRs have been determined to date. A key challenge to their structure determination is adequate protein expression. Here we report the quantification of protein expression in an insect cell expression system for all 826 human GPCRs using two different fusion constructs. Expression characteristics are analyzed in aggregate and among each of the five distinct subfamilies. These data can be used to identify trends related to GPCR expression between different fusion constructs and between different GPCR families, and to prioritize lead candidates for future structure determination feasibility

    Inhibition of 26S Protease Regulatory Subunit 7 (MSS1) Suppresses Neuroinflammation

    Recently, researchers have focused on immunosuppression induced by rifampicin. Our previous investigation found that rifampicin was neuroprotective by inhibiting the production of pro-inflammatory mediators, thereby suppressing microglial activation. In this study, using 2-dimensional gel electrophoresis (2-DE) and mass spectrometry (MS), we discovered that 26S protease regulatory subunit 7 (MSS1) was decreased in rifampicin-treated microglia. Western blot analysis verified the downregulation of MSS1 expression by rifampicin. As it is indicated that the modulation of the ubiquitin-26S proteasome system (UPS) with proteasome inhibitors is efficacious for the treatment of neuro-inflammatory disorders, we next hypothesized that silencing MSS1 gene expression might inhibit microglial inflammation. Using RNA interference (RNAi), we showed significant reduction of IkBα degradation and NF-kB activation. The production of lipopolysaccharides-induced pro-inflammatory mediators such as inducible nitric oxide synthase (iNOS), nitric oxide, cyclooxygenase-2, and prostaglandin E2 were also reduced by MSS1 gene knockdown. Taken together, our findings suggested that rifampicin inhibited microglial inflammation by suppressing MSS1 protein production. Silencing MSS1 gene expression decreased neuroinflammation. We concluded that MSS1 inhibition, in addition to anti-inflammatory rifampicin, might represent a novel mechanism for the treatment of neuroinflammatory disorders

On the one hand, it alleviates the contradiction between the city and ecological environment protection and guides ecological and green development in the suburbs of the city. On the other hand, it creates a better living environment for the city and promotes the sustainable development of urban–rural integration. This paper combines the sustainable design concept of permaculture with the resource conditions and advantages of the comprehensive project base and applies the concept of permaculture to the development of the “air vegetable plot” urban farm. At the same time, we use the integration strategy of agriculture and tourism, through the “Internet + Agriculture” path, to build a multi-functional industry of life, production and ecology; to promote the sustainable development of rural industries with the digitization of agricultural industries; to promote the common prosperity of rural areas; and to open a new model of “Internet + Future Agriculture”

    Photocatalysis: A Possible Vital Contributor to the Evolution of the Prebiotic Atmosphere and the Warming of the Early Earth

    The evolution of the early atmosphere was driven by changes in its chemical composition, which involved the formation of some critical gases. In this study, we demonstrate that nitrous oxide (N2O) can be produced from Miller’s early atmosphere (a mixture of CH4, NH3, H2, and H2O) by way of photocatalysis. Both NH3 and H2O were indispensable for the production of N2O by photocatalysis. Different conditions related to seawater and reaction temperature are also explored. N2O has a strong greenhouse gas effect, which is more able to warm the Earth than other gases and offers a reasonable explanation for the faint young Sun paradox on the early Earth. Moreover, the decomposition of N2O into N2 and O2 can be boosted by soft irradiation, providing a possible and important origin of atmospheric O2 and N2. The occurrence of O2 propelled the evolution of the atmosphere from being fundamentally reducing to oxidizing. This work describes a possible vital contribution of photocatalysis to the evolution of the early atmosphere

    An immunosuppressive subtype of senescent tumor cells predicted worse immunotherapy response in lung adenocarcinoma

    Summary: Senescent tumor cells (STCs) can induce immunosuppression, promoting tumor progression and therapy resistance. However, the specific characteristics of immunosuppressive STC have not been thoroughly investigated. This study aimed to characterize and elucidate the immunosuppressive phenotype of STC in lung adenocarcinoma by employing single-cell and bulk transcriptomics, as well as serum proteomics profiling. We identified senescence-related genes specific to tumors and identified Cluster10 of STC as the immunomodulatory subtype. Cluster10 exhibited a distinct secretome dominated by cytokines such as CXCL1, CXCL2, and CXCL8 and showed activation of transcription factors associated with cytokine secretion, including NFKB1, RELA, and STAT3. Notably, Cluster10 demonstrated the highest degree of intercellular communication among all cell types, with interactions as LGALS9-TIM3 and MIF-CD74. Furthermore, Cluster10 showed significant associations with poor prognosis and diminished response to immunotherapy. Analysis of serum proteomics data from our in-house cohort identified CXCL8 as a potential marker for predicting immunotherapeutic outcomes

    A Transformed Triangular Vegetation Index for Estimating Winter Wheat Leaf Area Index

    Leaf area index (LAI) is a key parameter in plant growth monitoring. For several decades, vegetation indices-based empirical method has been widely-accepted in LAI retrieval. A growing number of spectral indices have been proposed to tailor LAI estimations, however, saturation effect has long been an obstacle. In this paper, we classify the selected 14 vegetation indices into five groups according to their characteristics. In this study, we proposed a new index for LAI retrieval-transformed triangular vegetation index (TTVI), which replaces NIR and red bands of triangular vegetation index (TVI) into NIR and red-edge bands. All fifteen indices were calculated and analyzed with both hyperspectral and multispectral data. Best-fit models and k-fold cross-validation were conducted. The results showed that TTVI performed the best predictive power of LAI for both hyperspectral and multispectral data, and mitigated the saturation effect. The R2 and RMSE values were 0.60, 1.12; 0.59, 1.15, respectively. Besides, TTVI showed high estimation accuracy for sparse (LAI < 4) and dense canopies (LAI > 4). Our study provided the value of the Red-edge bands of the Sentinel-2 satellite sensors in crop LAI retrieval, and demonstrated that the new index TTVI is applicable to inverse LAI for both low-to-moderate and moderate-to-high vegetation cover
