    Computational analysis of pathological images enables a better diagnosis of TFE3 Xp11.2 translocation renal cell carcinoma

    TFE3 Xp11.2 translocation renal cell carcinoma (TFE3-RCC) generally progresses more aggressively compared with other RCC subtypes, but it is challenging to diagnose TFE3-RCC by traditional visual inspection of pathological images. In this study, we collect hematoxylin and eosin- stained histopathology whole-slide images of 74 TFE3-RCC cases (the largest cohort to date) and 74 clear cell RCC cases (ccRCC, the most common RCC subtype) with matched gender and tumor grade. An automatic computational pipeline is implemented to extract image features. Comparative study identifies 52 image features with significant differences between TFE3-RCC and ccRCC. Machine learning models are built to distinguish TFE3-RCC from ccRCC. Tests of the classification models on an external validation set reveal high accuracy with areas under ROC curve ranging from 0.842 to 0.894. Our results suggest that automatically derived image features can capture subtle morphological differences between TFE3-RCC and ccRCC and contribute to a potential guideline for TFE3-RCC diagnosis

    Meta-analysis Followed by Replication Identifies Loci in or near CDKN1B, TET3, CD80, DRAM1, and ARID5B as Associated with Systemic Lupus Erythematosus in Asians

    Systemic lupus erythematosus (SLE) is a prototype autoimmune disease with a strong genetic involvement and ethnic differences. Susceptibility genes identified so far only explain a small portion of the genetic heritability of SLE, suggesting that many more loci are yet to be uncovered for this disease. In this study, we performed a meta-analysis of genome-wide association studies on SLE in Chinese Han populations and followed up the findings by replication in four additional Asian cohorts with a total of 5,365 cases and 10,054 corresponding controls. We identified genetic variants in or near CDKN1B, TET3, CD80, DRAM1, and ARID5B as associated with the disease. These findings point to potential roles of cell-cycle regulation, autophagy, and DNA demethylation in SLE pathogenesis. For the region involving TET3 and that involving CDKN1B, multiple independent SNPs were identified, highlighting a phenomenon that might partially explain the missing heritability of complex diseases

    Performance Assessment of Hydrological Models Considering Acceptable Forecast Error Threshold

    It is essential to consider the acceptable threshold in the assessment of a hydrological model because of the scarcity of research in the hydrology community and errors do not necessarily cause risk. Two forecast errors, including rainfall forecast error and peak flood forecast error, have been studied based on the reliability theory. The first order second moment (FOSM) and bound methods are used to identify the reliability. Through the case study of the Dahuofang (DHF) Reservoir, it is shown that the correlation between these two errors has great influence on the reliability index of hydrological model. In particular, the reliability index of the DHF hydrological model decreases with the increasing correlation. Based on the reliability theory, the proposed performance evaluation framework incorporating the acceptable forecast error threshold and correlation among the multiple errors can be used to evaluate the performance of a hydrological model and to quantify the uncertainties of a hydrological model output

    Experimental Research on Fluid Coupling Flexible Actuator

    In the field of micromechanics, piezoelectric actuator has attracted great attention for its high-frequency response, high displacement resolution, and high output force. However, its prospect of practical application has been largely limited by the displacement of micrometer. A fluid coupling flexible actuator was proposed, which utilizes resonance to enlarge the output displacement. The actuator uses a piezoelectric oscillator as an excitation source, fluid as the transmission medium and a flexible diaphragm for the displacement output. On the condition that the fluid is inviscid and incompressible, mathematical formulation of the membrane vibration theory has been analyzed. Then, the prototype is made. The displacement is amplified 21 times to 1.106 mm when driving frequency is 127 Hz. The flexible diaphragm appears the largest displacement output when driving frequency is close to one of the system’s natural frequency. Then, the points with zero amplitude form a circle on the surface of flexible diaphragm and the movement direction of the flexible diaphragm is opposite on different sides of the circle. In fact, rather than vibrates at the first resonance frequency, the membrane in the essay is vibrating at a certain higher-order resonance frequency. The experimental results are mainly consistent with the theoretical analysis

    A Novel Flood Forecasting Method Based on Initial State Variable Correction

    The influence of initial state variables on flood forecasting accuracy by using conceptual hydrological models is analyzed in this paper and a novel flood forecasting method based on correction of initial state variables is proposed. The new method is abbreviated as ISVC (Initial State Variable Correction). The ISVC takes the residual between the measured and forecasted flows during the initial period of the flood event as the objective function, and it uses a particle swarm optimization algorithm to correct the initial state variables, which are then used to drive the flood forecasting model. The historical flood events of 11 watersheds in south China are forecasted and verified, and important issues concerning the ISVC application are then discussed. The study results show that the ISVC is effective and applicable in flood forecasting tasks. It can significantly improve the flood forecasting accuracy in most cases

    Construction of the Three Gorges Dam attenuates the relationship between air temperature, river water temperature and baseflow

    Baseflow is a critical hydrological variable linked to a variety of environmental factors. The construction of a dam may modify baseflow generation and adjust its environmental dependence via the regulation of discharge. However, its effects on the relationships among air temperature, water temperature, and baseflow remain unclear. Here, we use copula functions to explore the relationships among these variables in the Yangtze River and show how they have changed in response to the Three Gorges Dam (TGD). Because of the buffering effects of the baseflow, baseflow is negatively correlated to water temperature, and the relationship between baseflow and water temperature lessens as air temperature rises. Construction of the TGD attenuates the relationship between baseflow and water temperature when air temperature is low, and this attenuation is especially prominent in winter and spring. Our findings will enhance environmental protection in the Yangtze River and improve the ecological operations of the TGD.</p
