Isolasi Enzim Horseradish Peroksidase (Hrp) Dari Kultur Sel Daun Armoracia Lapatifolia Dengan Cara Fraksinasi Menggunakan Amonium Sulfat
Isolation of HRP ( Horseradish Peroksidase ) from Armoracia lapatifolia plant cell culture was conducted by fractionation using ammonium sulfate. Optimation of thefractionation process was carried out by concentrating the crude enzyme, investigating the fractionation stages and precipitation with ammonium sulfate. Two-stage fractionation namely with 0 - 40% and 40 - 85 % saturation of ammonium sulfate gave better results in terms of enzyme activity. In 0 - 40% saturation, the purity was 23fold with 11.4 %yield. The 20 highest yield, 60% and purify of 43 folds were showed by fraction which were obtained at fractionation 40 - 85 % saturation
Effects of Medium Composition on Oxytetracycline Production by Streptomyces Rimosus Atcc 33022
The economical production of antibiotics to some extent depends on the availabilily of cheap substrates. The work reported in the present paper deals with the fermentative production of oxytetracycline by Streptomyces rimosus ATCC 33022 using commerciol high fructose syrup (HFS), vitamin B complex and citric acid of technical grode. The effects of concentration of high fructose syrup (0.5 - 2.5 %, v/v), commercial vitamin B complex (0.03 - 0.07 %, w/v) and the citric acid (0.34 - 1.28 %. w/v) were examined in this study. It was found that fermentation medium (medium-MHFS) containing high fructose syrup 1.0 % produced maximum activily of oxytetracycline after 4 days incubation period. Fermentation tnedium (mediwn-MBpleJ containing 0.05 % commerciol vitamin B complex showed maximum acrivily after 3 days incubation. While the addition of citric acid (0.64 %) to the fermentation medium (medium-MCA) was found optimumfor production oxytetracycline
Pengaruh Sumber Karbon Terhadap Produksi Enzim Inulinfruktotransferase Dari Nonomuraea SP
The aims of this research is to produce inulinfructotransferase enzyme with potentially benefit of bacteria Actinomycetes which was isolated from Indonesia soils to produce Difructoanhydride-Ill (DFA III), a compound has physiology activities to enhance calsium absorption of rats. cows and human colons so that the materials can be applied as antiosteoporosis. The processing of inulinfructotransferase enzyme from Nonomuraea sp by usingshakeflask level and study the influence of carbohydrate source were conducted. This enzyme has optimum temperature at 65rtC,higher than optimum temperature which has found before and the activities was stable at 70dc for 20 minutes. So that the enzyme has potentially applicable for industry scale. The results indicated that optimation condition for processing inulinfructotransferase enzyme of Nonomuraea sp : temperature sore. incubation time 48 hours.carbon source of inulin 20 g/L, nitrogen source 5g/L, noyeast extract added, the enzyme activities was 7,3 Unit/ml; However,when medium contains yeast extract 0,2 gIL,the inulinfructotransferase enzyme activity was 14,6 Unit/mL
Kandungan Inulin Dari Umbi Dahliasp Yang Ditanam Pada Jenis Tanah Vertisol, Inceptisol Dan Andisol
Tujuan dari penelitian ini adalah menentukan kandungan inulin dari umbi Dahlia sp yang ditanam didaerah sejuk seperti Cianjur, Lembang dan Sukabumi pada jenis tanah yang berbeda dalam rangka rencana produksi di Jawa Barat. Inulin adalah oligosacharida yang terjadi secara alami dengan komposisi gabungan fruktooligosacharida dari oligomer dengan derajat polimerisasi yang berbeda. Umbi dahlia dari bunga berwarna ungu, merah, kuning dan putih yang ditanam di tiga lokasi digunakan sebagai sumber inulin. Metode ekstraksi yang digunakan yaitu cara ekstraksi berdasarkan kelarutan inulin dalam air pada suhu 800 C dan pengendapan dilakukan dengan etanol 30%. Hasil penelitian menunjukkan kadar inulin dari umbi bunga warna merah yang ditanam pada tanah Inceptisols (Sukabumi) memiliki kadar inulin tertinggi yaitu sebesar 17,99%, gula total 19.13%, karbohidrat 8.02%, kadar abu 0,15% sementara kadar inulin terendah pada jenis tanah Vertisols (Cianjur) yaitu 14.90%, gula total 26.61%, karbohidrat 8.66% kadar abu 0.17% sementara jenis tanahAndisols (Lembang) menghasilkan inulin dengan  kadar inulin 11.84%, gula total 12.48%, karbohidrat 9.38% dan kadar abu 0.09%. Kata kunci :Umbidahlia , inulin, vertisols inceptisols, andisols, karbohidrat The objectives of this research were to measure inulin content of Dahlia sp tubers widely planted in cool area such as Cianjur, Lembang and Sukabumi at different soil type in order to produce inulin in West Java. Inulin is a naturally occurring fructooligosaccharide composed of a mixture of oligomers of varying degrees of polymerization. Dahlia tuber of flower such as violet, red, yellow and white colour was planted on different location used as the source for inulin. The extraction methods was used base on inulin dissolve in water at 800 C and precipitation was carried out by using ethanol 30%.The results indicated that inulin powder of red flower was planted on inceptisols ( Sukabumi) has the highest level of inulin that was 17.99%, total glucose 19.13%, carbohydrate 8.02%, ash 0.15% .The lowest inulin content was detected in vertisols type ( Cianjur) as much as 14.90%, total glucose 26.61%, carbohydrate 8.66%, ash 0.17% meanwhile  andisols type has inulin concentration 11.84%, total glucose 12.84%, carbohydrate 9.38% and ash 0.09%
