    Knowth passage-grave in Ireland: An instrument of precision astronomy?

    Knowth is one of three large monuments at the Neolithic complex in the bend of the River Boyne in County Meath, Ireland. The others are Newgrange and Dowth. All three have obvious solar alignments but whereas the alignment to the winter solstice sunrise at Newgrange has been extensively researched and interpreted, little has been attempted regarding the way that astronomy functions at Knowth and Dowth. This paper treats the evidence for solar and lunar alignments at Knowth. Knowth has two internal passages with entrances at the east and west. The paper draws on new surveys as well as interpretations of the evidence at Knowth that includes rock art engraved on kerbstones around the circumference. Particular engravings on kerbstone K52 are interpreted as depicting astronomical cycles. It is argued that, while Knowth’s passages function in relation to the equinoxes, they are not internally orientated to match exactly the equinoctial directions. Rather, it seems that they may have been constructed and used to facilitate the harmonisation of the solar and lunar cycles - much in the same way as does the equinoctial Judeo-Christian festival of Easter. The paper concludes by suggesting that like Newgrange, Knowth may be an astronomical instrument that enabled its builders and users to construct accurate calendars and counting systems, which in turn facilitated calculated planning and was a fundamental structuring principle for their ritual lives and cosmological beliefs

    Efficacy of a compulsory homework programme for increasing physical activity and improving nutrition in children: A cluster randomised controlled trial

    Background: Most physical activity interventions in children focus on the school setting; however, children typically engage in more sedentary activities and spend more time eating when at home. The primary aim of this cluster randomised controlled trial was to investigate the effects of a compulsory, health-related homework programme on physical activity, dietary patterns, and body size in primary school-aged children. Methods: A total of 675 children aged 7-10 years from 16 New Zealand primary schools participated in the Healthy Homework study. Schools were randomised into intervention and control groups (1:1 allocation). Intervention schools implemented an 8-week applied homework and in-class teaching module designed to increase physical activity and improve dietary patterns. Physical activity was the primary outcome measure, and was assessed using two sealed pedometers that monitored school- and home-based activity separately. Secondary outcome measures included screen-based sedentary time and selected dietary patterns assessed via parental proxy questionnaire. In addition, height, weight, and waist circumference were measured to obtain body mass index (BMI) and waist-to-height ratio (WHtR). All measurements were taken at baseline (T0), immediately post-intervention (T1), and 6-months post-intervention (T2). Changes in outcome measures over time were estimated using generalised linear mixed models (GLMMs) that adjusted for fixed (group, age, sex, group x time) and random (subjects nested within schools) effects. Intervention effects were also quantified using GLMMs adjusted for baseline values. Results: Significant intervention effects were observed for weekday physical activity at home (T1 [P < 0.001] and T2 [P = 0.019]), weekend physical activity (T1 [P < 0.001] and T2 [P < 0.001]), BMI (T2 only [P = 0.020]) and fruit consumption (T1 only [P = 0.036]). Additional analyses revealed that the greatest improvements in physical activity occurred in children from the most socioeconomically deprived schools. No consistent effects on sedentary time, WHtR, or other dietary patterns were observed. Conclusions: A compulsory health-related homework programme resulted in substantial and consistent increases in children's physical activity - particularly outside of school and on weekends - with limited effects on body size and fruit consumption. Overall, our findings support the integration of compulsory home-focused strategies for improving health behaviours into primary education curricula.     Optimizing exosomal RNA isolation for RNA-Seq analyses of archival sera specimens

    Exosomes are endosome-derived membrane vesicles that contain proteins, lipids, and nucleic acids. The exosomal transcriptome mediates intercellular communication, and represents an understudied reservoir of novel biomarkers for human diseases. Next-generation sequencing enables complex quantitative characterization of exosomal RNAs from diverse sources. However, detailed protocols describing exosome purification for preparation of exosomal RNA-sequence (RNA-Seq) libraries are lacking. Here we compared methods for isolation of exosomes and extraction of exosomal RNA from human cell-free serum, as well as strategies for attaining equal representation of samples within pooled RNA-Seq libraries. We compared commercial precipitation with ultracentrifugation for exosome purification and confirmed the presence of exosomes via both transmission electron microscopy and immunoblotting. Exosomal RNA extraction was compared using four different RNA purification methods. We determined the minimal starting volume of serum required for exosome preparation and showed that high quality exosomal RNA can be isolated from sera stored for over a decade. Finally, RNA-Seq libraries were successfully prepared with exosomal RNAs extracted from human cell-free serum, cataloguing both coding and non-coding exosomal transcripts. This method provides researchers with strategic options to prepare RNA-Seq libraries and compare RNA-Seq data quantitatively from minimal volumes of fresh and archival human cell-free serum for disease biomarker discovery

    Young people and environmental affordances in urban sustainable development

    Background: Cities are at the fore of sustainability challenges of the twenty-first century, and many, particularly in Asia and Africa, are predominantly youthful spaces. Understanding young people's experiences in urban environments is therefore important as we strive to achieve both the Sustainable Development Goals and the Paris Agreement. Two amenities identified in the urban Sustainable Development Goal 11, transport and public and green space, are specifically recognised as applying to youth. Yet, there is little analysis that explicitly considers how youth experience these amenities across the Global North and South, and no current measures for understanding progress in youth experiences of green space and transport. Results: This paper provides a comparative analysis of young people's experiences with local transport and green space in seven diverse urban communities (Christchurch, New Zealand; Dhaka, Bangladesh; Lambeth/London, UK; Makhanda, South Africa; New Delhi, India; São Paulo, Brazil; and Yokohama, Japan). Our study contributes to a growing body of literature that seeks to listen to child and youth perspectives to understand their environmental experiences. We examine the 'affordances' young residents aged 12 to 24 years currently associate with green space and transport amenities. Affordances are defined here as the inter-relationships between what a local environment offers young people and their perceptions and actions. Drawing on focus groups and interviews conducted with 332 young people, we identify five affordances young people associate in relation to transport and public space across these diverse urban settings: (1) social inclusion and belonging; (2) autonomy; (3) physical comfort and security; (4) relaxation and reflection; and (5) health and fitness. Conclusions The paper contributes to growing interdisciplinary research interest in measuring affordances as a way to advance the Sustainable Development Goals in an urban context. In providing a comparative account of young people's experiences across diverse contexts, our discussion highlights how affordances in relation to transport or public and green space can help understand the multiple interconnections between the well-being of young people and sustainability. In particular, we argue that it is not merely the provision of transport or public and green space that matters, but the nuanced meaning of places and experiences as understood by local communities that needs to be recognised if we are to better support urban youth wellbeing and advance sustainable development goals

    Youth Attitudes and Participation in Climate Protest

    This article examines youth participation the school climate strikes of 2018 and 2019 (also known as #Fridays4Future), through an exploratory study conducted in seven diverse cities. Despite the international nature of the climate strikes, we know little about the factors that influenced youth participation in these protests beyond the global North. This matters because youth of the global South are disproportionately impacted by climate change and there is growing concern that the climate movement is dominated by narratives that marginalize the voices and priorities of Indigenous communities and people of color. In this context, the exploratory research reported here aimed to compare the attitudes of climate protesters (n = 314) and their non-protester peers (n = 1,217), in diverse city samples drawn from a wider study of children and youth aged 12–24 years, living in Christchurch (New Zealand); Dhaka (Bangladesh); Lambeth, London (United Kingdom); Makhanda (South Africa); New Delhi (India); São Paulo (Brazil); and Yokohama (Japan). Using cross-sectional data (N = 1,531) and binary logistic regression models, researchers examined three common explanations for youth participation in protest: availability (biographical and structural), political engagement (reported individual and collective efficacy of strikers and non-strikers), and self-reported biospheric values amongst participants. Results indicate that even in diverse city samples, structural availability (civic skills and organizational membership) predicted strike participation across city samples, but not political engagement (self-efficacy and collective efficacy). Youth who reported that ‘living in harmony with nature and animals’ was important for their wellbeing, were also more likely to strike than their peers. Descriptive statistics indicated that the majority (85 percent) of all protestors in this study agreed climate change was a serious issue and a startling 65 percent said that they think about climate change “all the time”. Reported rates of youth climate protest participation varied across city samples as did the extent to which participants reported having friends take part or expecting climate change to have a personal impact. While the study is exploratory, it points to the need for more extensive research to understand the diversity of youth participation in ‘global climate strikes’

    Metabolic profiling of aortic stenosis and hypertrophic cardiomyopathy identifies mechanistic contrasts in substrate utilization

    Aortic stenosis (AS) and hypertrophic cardiomyopathy (HCM) are distinct disorders leading to left ventricular hypertrophy (LVH), but whether cardiac metabolism substantially differs between these in humans remains to be elucidated. We undertook an invasive (aortic root, coronary sinus) metabolic profiling in patients with severe AS and HCM in comparison with non‐LVH controls to investigate cardiac fuel selection and metabolic remodeling. These patients were assessed under different physiological states (at rest, during stress induced by pacing). The identified changes in the metabolome were further validated by metabolomic and orthogonal transcriptomic analysis, in separately recruited patient cohorts. We identified a highly discriminant metabolomic signature in severe AS in all samples, regardless of sampling site, characterized by striking accumulation of long‐chain acylcarnitines, intermediates of fatty acid transport across the inner mitochondrial membrane, and validated this in a separate cohort. Mechanistically, we identify a downregulation in the PPAR‐α transcriptional network, including expression of genes regulating fatty acid oxidation (FAO). In silico modeling of β‐oxidation demonstrated that flux could be inhibited by both the accumulation of fatty acids as a substrate for mitochondria and the accumulation of medium‐chain carnitines which induce competitive inhibition of the acyl‐CoA dehydrogenases. We present a comprehensive analysis of changes in the metabolic pathways (transcriptome to metabolome) in severe AS, and its comparison to HCM. Our results demonstrate a progressive impairment of β‐oxidation from HCM to AS, particularly for FAO of long‐chain fatty acids, and that the PPAR‐α signaling network may be a specific metabolic therapeutic target in AS
