    Physiological, Anatomical and Metabolic Implications of Salt Tolerance in the Halophyte Salvadora persica under Hydroponic Culture Condition

    Salt tolerance mechanism of an extreme halophyte Salvadora persica was assessed by analysing growth, nutrient uptake, anatomical modifications and alterations in levels of some organic metabolites in seedlings imposed to various levels of salinity (0, 250, 500 and 750 mM NaCl) under hydroponic culture condition. After 21 days of salt treatment, plant height, leaf area and shoot biomass decreased with increase in salinity whereas the leaf succulence increased significantly with increasing salinity in S. persica. The RWC% of leaf increased progressively in salt-treated seedlings as compared to control. Na+ contents of leaf, stem and root increased in dose-dependent manner whereas there was no significant changes in K+ content. There was significant alterations in leaf, stem and root anatomy by salinity. The thickness of epidermis and spongy parenchyma of leaf increased in salt treated seedlings as compared to control, whereas palisade parenchyma decreased dramatically in extreme salinity (750 mM NaCl). There was a significant reduction in stomatal density and stomatal pore area of leaf with increasing salinity. Anatomical observations of stem showed that the epidermal cells diameter and thickness of cortex decreased by salinity whereas thickness of hypodermal layer, hypodermal cell diameter, pith area and pith cell diameter increased by high salinity. The root anatomy showed an increase in epidermal thickness by salinity whereas diameters of epidermal cells and xylem vessels decreased. Total soluble sugar content remained unchanged at all levels of salinity whereas reducing sugar content increased by 2-fold at high salinity (750 mM NaCl). The starch content of leaf decreased progressively in NaCl treated seedlings as compared to control. Total free amino acid content did not change at low salinity (250 mM), whereas it increased significantly at higher salinity (500 and 750 mM NaCl). The proline content increased in the NaCl treated seedlings as compared to control. There was no significant changes in polyphenol levels in leaf at all levels of salinity. The results from the present study reveal that seedlings imposed with various levels of salinity experience physiological, biochemical and anatomical modifications in order to circumvent under extreme saline environment.     Clinical features and predictors of in hospital mortality in patients with intra cerebral haemorrhage

    Background: Intracerebral haemorrhage (ICH) accounts for 15-20% of all strokes and is associated with significant morbidity and mortality. The present study was done to assess the risk factors for ICH and determine the factors responsible for poor outcome in ICH.Methods: Consecutive patients of ICH were assessed for modifiable and non-modifiable risk factors, followed by detailed examination with emphasis on ICH score and CT scan findings.Results: There were 200 patients with ICH; 108 males and 92 females. The prevalence of ICH was maximum in the age group of 61-70 years (34%). Hypertension was present in 68% of the patients and was the single most important modifiable risk factor. The most common presenting symptom was altered sensorium present in 58% patients. The mean hematoma volume in our study was 44+/-45 cm3. 45 patients with a GCS <5 and both the patients with ICH score 5 expired. On multivariate analysis, there was a significant correlation of mortality with GCS and hematoma volume.Conclusions: Hypertension is the commonest risk factor for ICH. Patients with a low GCS score, large hematoma volume and a higher ICH score have a poor prognosis and higher probability of mortality.

    Endoscopic Ultrasound-Guided Random Omental Fine Needle Aspiration: A Novel Technique for the Diagnosis of Peritoneal Carcinomatosis

    Background/Aims Diagnostic abdominal paracentesis has been described in literature to have variable sensitivity of 50%–75% for the detection of peritoneal carcinomatosis (PC). We believe that random needle aspirates from the omentum, even in the absence of obvious deposits by endoscopic ultrasound (EUS), could prove malignancy in patients with PC. Methods Consecutive patients who underwent EUS for diagnosis and staging of cancer and found to have ascites were included after obtaining informed consent. EUS-guided fine needle aspiration (EUS-FNA) from random sites in the omentum was performed through the transgastric route using a linear echoendoscope. Results Fifty-four patients underwent EUS during October 2015 to April 2017 for detection, staging, or FNA of a suspected malignant lesion. Ascites was seen in 17 patients and 15 patients who fulfilled the criteria were included. The procedure was successful in all patients. Cytology was suggestive of malignancy in 12 (80%) but not suggestive of malignancy in 3 (20%) patients. Three patients who tested negative had hyperbilirubinemia with biliary obstruction. Their ascitic fluid analysis result was also negative. Conclusions Random FNA of the omentum in patients with malignancy-related ascites is highly effective in the diagnosis of PC and could be employed during EUS evaluation of malignancies

    Molecular profiling of ETS and non‐ETS aberrations in prostate cancer patients from northern India

    BACKGROUNDMolecular stratification of prostate cancer (PCa) based on genetic aberrations including ETS or RAF gene‐rearrangements, PTEN deletion, and SPINK1 over‐expression show clear prognostic and diagnostic utility. Gene rearrangements involving ETS transcription factors are frequent pathogenetic somatic events observed in PCa. Incidence of ETS rearrangements in Caucasian PCa patients has been reported, however, occurrence in Indian population is largely unknown. The aim of this study was to determine the prevalence of the ETS and RAF kinase gene rearrangements, SPINK1 over‐expression, and PTEN deletion in this cohort.METHODSIn this multi‐center study, formalin‐fixed paraffin embedded (FFPE) PCa specimens (n = 121) were procured from four major medical institutions in India. The tissues were sectioned and molecular profiling was done using immunohistochemistry (IHC), RNA in situ hybridization (RNA‐ISH) and/or fluorescence in situ hybridization (FISH).RESULTSERG over‐expression was detected in 48.9% (46/94) PCa specimens by IHC, which was confirmed in a subset of cases by FISH. Among other ETS family members, while ETV1 transcript was detected in one case by RNA‐ISH, no alteration in ETV4 was observed. SPINK1 over‐expression was observed in 12.5% (12/96) and PTEN deletion in 21.52% (17/79) of the total PCa cases. Interestingly, PTEN deletion was found in 30% of the ERG‐positive cases (P = 0.017) but in only one case with SPINK1 over‐expression (P = 0.67). BRAF and RAF1 gene rearrangements were detected in ∼1% and ∼4.5% of the PCa cases, respectively.CONCLUSIONSThis is the first report on comprehensive molecular profiling of the major spectrum of the causal aberrations in Indian men with PCa. Our findings suggest that ETS gene rearrangement and SPINK1 over‐expression patterns in North Indian population largely resembled those observed in Caucasian population but differed from Japanese and Chinese PCa patients. The molecular profiling data presented in this study could help in clinical decision‐making for the pursuit of surgery, diagnosis, and in selection of therapeutic intervention.     CicArVarDB: SNP and InDel database for advancing genetics research and breeding applications in chickpea

    Molecular markers are valuable tools for breeders to help accelerate crop improvement. High throughput sequencing technologies facilitate the discovery of large-scale variations such as single nucleotide polymorphisms (SNPs) and simple sequence repeats (SSRs). Sequencing of chickpea genome along with re-sequencing of several chickpea lines has enabled the discovery of 4.4 million variations including SNPs and InDels. Here we report a repository of 1.9 million variations (SNPs and InDels) anchored on eight pseudomolecules in a custom database, referred as CicArVarDB that can be accessed at http://cicarvardb.icrisat.org/. It includes an easy interface for users to select variations around specific regions associated with quantitative trait loci, with embedded webBLAST search and JBrowse visualisation. We hope that this database will be immensely useful for the chickpea research community for both advancing genetics research as well as breeding applications for crop improvement
