    Evaluation of a low-cost optical particle counter (Alphasense OPC-N2) for ambient air monitoring

    A fast-growing area of research is the development of low-cost sensors for measuring air pollutants. The affordability and size of low-cost particle sensors makes them an attractive option for use in experiments requiring a number of instruments such as high-density spatial mapping. However, for these low-cost sensors to be useful for these types of studies their accuracy and precision need to be quantified. We evaluated the Alphasense OPC-N2, a promising low-cost miniature optical particle counter, for monitoring ambient airborne particles at typical urban background sites in the UK. The precision of the OPC-N2 was assessed by co-locating 14 instruments at a site to investigate the variation in measured concentrations. Comparison to two different reference optical particle counters as well as a TEOM-FDMS enabled the accuracy of the OPC-N2 to be evaluated. Comparison of the OPC-N2 to the reference optical instruments shows some limitations for measuring mass concentrations of PM1, PM2.5 and PM10. The OPC-N2 demonstrated a significant positive artefact in measured particle mass during times of high ambient RH (>85%) and a calibration factor was developed based upon °-Köhler theory, using average bulk particle aerosol hygroscopicity. Application of this RH correction factor resulted in the OPC-N2 measurements being within 33% of the TEOM-FDMS, comparable to the agreement between a reference optical particle counter and the TEOM-FDMS (20%). Inter-unit precision for the 14 OPC-N2 sensors of 22±13% for PM10 mass concentrations was observed. Overall, the OPC-N2 was found to accurately measure ambient airborne particle mass concentration provided they are (i) correctly calibrated and (ii) corrected for ambient RH. The level of precision demonstrated between multiple OPC-N2s suggests that they would be suitable devices for applications where the spatial variability in particle concentration was to be determined

    Effect of aerosol composition on the performance of low-cost optical particle counter correction factors

    There is considerable interest in using low-cost optical particle counters (OPCs) to supplement existing routine air quality networks that monitor particle mass concentrations. In order to do this, low-cost OPC data need to be comparable with particle mass reference instrumentation; however, there is currently no widely agreed upon methodology to accomplish this. Aerosol hygroscopicity is known to be a key parameter to consider when correcting particle mass concentrations derived from low-cost OPCs, particularly at high ambient relative humidity (RH). Correction factors have been developed that apply κ-Köhler theory to correct for the influence of water uptake by hygroscopic aerosols. We have used datasets of co-located reference particle measurements and low-cost OPC (OPC-N2, Alphasense) measurements, collected in four cities on three continents, to explore the performance of this correction factor. We provide evidence that the elevated particle mass concentrations, reported by the low-cost OPC relative to reference instrumentation, are due to bulk aerosol hygroscopicity under different RH conditions, which is determined by aerosol composition and, in particular, the levels of hygroscopic aerosols (sulfate and nitrate). We exploit measurements made in volcanic plumes in Nicaragua, which are predominantly composed of sulfate aerosol, as a natural experiment to demonstrate this behaviour in the ambient atmosphere; the observed humidogram from these measurements closely resembles the calculated pure sulfuric acid humidogram. The results indicate that the particle mass concentrations derived from low-cost OPCs during periods of high RH (>60 %) need to be corrected for aerosol hygroscopic growth. We employed a correction factor based on κ-Köhler theory and observed that the corrected OPC-N2 PM2.5 mass concentrations were within 33 % of reference measurements at all sites. The results indicated that a κ value derived in situ (using suitable reference instrumentation) would lead to the most accurate correction relative to co-located reference instruments. Applying a κ values from the literature in the correction factor also resulted in improved OPC-N2 performance, with the measurements being within 50 % of the reference values. Therefore, for areas where suitable reference instrumentation for developing a local correction factor is lacking, using a literature κ value can result in a reasonable correction. For locations with low levels of hygroscopic aerosols and low RH values, a simple calibration against gravimetric measurements (using suitable reference instrumentation) would likely be sufficient. Whilst this study generated correction factors specific for the Alphasense OPC-N2 sensor, the calibration methodology developed is likely amenable to other low-cost PM sensors

    Correction of the cystic fibrosis defect in vitro by retrovirus-mediated gene transfer

    We have used retrovirus-mediated gene transfer to demonstrate complementation of the cystic fibrosis (CF) defect in vitro. Amphotropic retroviruses were used to transduce a functional cystic fibrosis transmembrane conductance regulator (CFTR) cDNA into CFPAC-1, a pancreatic adenocarcinoma cell line derived from a patient with CF that stably expresses the chloride transport abnormalities characteristic of CF. CFPAC-1 cells were exposed to control virus (PLJ) and CFTR-expressing virus (PLJ-CFTR); viral-transduced clones were isolated and subjected to molecular and physiologic analysis. RNA analysis detected a viral-derived CFTR transcript in all of the PLJ-CFTR clones that contained unrearranged proviral sequences. Agents that increase intracellular cAMP stimulated 125I efflux in PLJ-CFTR clones but not PLJ clones. Whole-cell patch-clamp performed on three responding clones showed that the anion efflux responses were due to cAMP stimulation of Cl conductance. This paper hopes to make an overview of historical, diplomatic, economic and political aspects and present events, as well as to offer a perspective of what the future might hold for the relationship between Brazil, the Caribbean Community and its member countries

    Nutrient intakes and nutritional biomarkers in pregnant adolescents: a systematic review of studies in developed countries

    Background: Babies born to adolescent mothers have been shown to have poorer outcomes compared to those born to adults. Nutritional status may have an important role to play in improving the health of pregnant adolescents; however there is a lack of evidence regarding the adequacy of adolescent diets during pregnancy. This systematic review aims to examine what is known about the nutritional status of adolescent pregnant women. Methods: A systematic search of the literature identified 21 studies which met the inclusion criteria for the review. Primary research papers using any methods were included where they were published in English between January 1995 and May 2015 and included measurements of nutrient intakes or biological markers of nutritional status in pregnant women aged 11-19 years. Individual study data was first summarised narratively before study means were pooled to give an estimate of nutritional status in the population. Results: The results show that individual studies reported intakes of energy, fibre and a number of key micronutrients which were below recommended levels. Biological markers of iron and selenium status also showed cause for concern. Pooled analysis of individual means as a percentage of UK Dietary Reference Intakes showed intakes of vitamin D (34.8 % CI 0-83.1) to be significantly below recommendations (p=0.05). Serum selenium levels were also found to be low (61.8 μg/L, CI 39-84). Conclusions: This review has identified a number of areas where the nutritional status of pregnant adolescents is sub-optimal, which may have implications for the health of adolescent mothers and their babies. It was not however possible to examine the impact of supplement use or socio-demographic characteristics which limits the interpretation these results. Further work is needed to establish the characteristics of those most at risk within this population, how this differs from adult pregnant women and the role of supplementation in achieving adequate nutrition

    Transcriptional Response of Zebrafish Embryos Exposed to Neurotoxic Compounds Reveals a Muscle Activity Dependent hspb11 Expression

    Acetylcholinesterase (AChE) inhibitors are widely used as pesticides and drugs. Their primary effect is the overstimulation of cholinergic receptors which results in an improper muscular function. During vertebrate embryonic development nerve activity and intracellular downstream events are critical for the regulation of muscle fiber formation. Whether AChE inhibitors and related neurotoxic compounds also provoke specific changes in gene transcription patterns during vertebrate development that allow them to establish a mechanistic link useful for identification of developmental toxicity pathways has, however, yet not been investigated. Therefore we examined the transcriptomic response of a known AChE inhibitor, the organophosphate azinphos-methyl (APM), in zebrafish embryos and compared the response with two non-AChE inhibiting unspecific control compounds, 1,4-dimethoxybenzene (DMB) and 2,4-dinitrophenol (DNP). A highly specific cluster of APM induced gene transcripts was identified and a subset of strongly regulated genes was analyzed in more detail. The small heat shock protein hspb11 was found to be the most sensitive induced gene in response to AChE inhibitors. Comparison of expression in wildtype, ache and sopfixe mutant embryos revealed that hspb11 expression was dependent on the nicotinic acetylcholine receptor (nAChR) activity. Furthermore, modulators of intracellular calcium levels within the whole embryo led to a transcriptional up-regulation of hspb11 which suggests that elevated intracellular calcium levels may regulate the expression of this gene. During early zebrafish development, hspb11 was specifically expressed in muscle pioneer cells and Hspb11 morpholino-knockdown resulted in effects on slow muscle myosin organization. Our findings imply that a comparative toxicogenomic approach and functional analysis can lead to the identification of molecular mechanisms and specific marker genes for potential neurotoxic compounds

    A Randomized Trial Examining the Effects of Parent Engagement on Early Language and Literacy: The Getting Ready Intervention

    Language and literacy skills established during early childhood are critical for later school success. Parental engagement with children has been linked to a number of adaptive characteristics in preschoolers including language and literacy development, and family-school collaboration is an important contributor to school readiness. This study reports the results of a randomized trial of a parent engagement intervention designed to facilitate school readiness among disadvantaged preschool children, with a particular focus on language and literacy development. Participants included 217 children, 211 parents, and 29 Head Start teachers in 21 schools. Statistically significant differences in favor of the treatment group were observed between treatment and control participants in the rate of change over 2 academic years on teacher reports of children’s language use (d = 1.11), reading (d = 1.25), and writing skills (d = .93). Significant intervention effects on children’s direct measures of expressive language were identified for a subgroup of cases where there were concerns about a child’s development upon entry into preschool. Additionally, other child and family moderators revealed specific variables that influenced the treatment’s effects
