    A New Marker on Chicken Hematopoietic Cells is Defined by a Monoclonal Antibody Raised Against a V ß Chain of the Human TCR

    In this paper, we show that a mouse monoclonal antibody, 111-427, specific for the V ß 5.3 chain of the human T-cell receptor (TCR) for antigen, also reacts with chicken hematopoietic cells. Our data indicate that the majority of 111-427 positive cells among peripheral blood leucocytes (PBL) are thrombocytes. This antibody also recognizes two in vitro cell lines, III-C5, an IL-2-dependent T-cell-line and HD11, a macrophage cell line. In addition, erythrocytes and a minor subpopulation of thymus and spleen cells are also stained by the monoclonal antibody (mAb). No specific immunoprecipitation could be detected from 125I radiolabeled cell lysates. By Western blotting techniques, the 111- 427 mAb identifies a single band of apparent molecular weight 91 kD, unaffected by reduction, from III-C5 and HD11 cell lysates. This band is absent in negative cell control lysates. On thrombocytes, the apparent molecular weight of the band is shifted to 87 kD. These results indicate that the mAb does not recognize the chicken T-cell receptor for antigen, but a cell surface marker shared primarily between thrombocytes and erythrocytes.     There is a growing evidence that obesity is not only a weight problem, but it is linked to adverse neurocognitive outcomes. Besides obesity, frontal lobe based cognitive deficits in depressed patients are confirmed, and interactions between depression and obesity are known. In our study we investigated the relationship between cognitive functioning, mood and female obesity. Our findings revealed reduced mental flexibility and sustained attention capacity in obesity together with the presence of depressive mood. The mediating role of depression is confirmed. Positive emotion was associated with cognitive functions independently from BMI.     Gdt1p, a new player involved in calcium homeostasis in Saccharomyces cerevisiae? Development of organelles-localized calcium and pH-sensitive probes.

    The Ca2+ ion is a ubiquitous intracellular messenger involved in various signaling processes. The basal concentration of cytosolic calcium is maintained at a very low level. Upon stimulation, a transient increase of cytosolic Ca2+ originated from internal and/or external stores is observed. Therefore, a complex toolkit of pumps, exchangers and channels localized in the different compartments of the cell are essential to tightly control this mechanism. Although a significant number of those are already well known, there are still some lacking actors in the Saccharomyces cerevisiae Ca2+-signaling toolkit. Recently, we have identified a novel family of putative Ca2+/H+ antiporters (UPF0016), whose Gdt1p is the yeast member. Gdt1p is a six-transmembrane spans protein located to the Golgi apparatus. It possesses a characteristic conserved hydrophilic motif in two transmembrane spans. Phenotypical assays and cytosolic Ca2+ concentration measurement suggested that Gdt1p could play an important role in Ca2+ homeostasis. However, the precise function of Gdt1p remains unclear. In order to elucidate the physiological function of Gdt1p related to calcium and pH homeostasis within the secretory pathway, we try to get direct evidences of Ca2+/H+ transport. For this purpose, we are addressing genetically-encoded Ca2+ sensors and H+ probes to the lumen of different organelles (endoplasmic reticulum, Golgi apparatus, vacuole). Aequorin, GAP (GFP-Aequorin Probe) and pHluorin are bioluminescent/fluorescent probes useful for Ca2+ or pH measurement, respectively. So far, we correctly localized these sensors to the endoplasmic reticulum and the Golgi apparatus lumens. However, the low expression level does not allow us to distinguish specific signal from noise. The next challenge is then to increase the expression level of these probes while preserving the right localization

    Background: Partial anomalous pulmonary venous return (PAPVR) is a rare condition that may lead to pulmonary arterial hypertension (PAH). We sought to determine the prevalence of PAPVR, the follow-up rate of incidentally discovered PAPVR, the repercussions of volume and pressure overload on the right ventricle (RV), and the prevalence and predictors of PAH. Methods and results: Fifty PAPVR patients aged ≥18 years were included. All underwent transthoracic echocardiography and multislice imaging. PAPVR prevalence was 0.2% (23/13,606 chest computed tomography reports). Of the 26 incidentally diagnosed patients, 14 (54%) did not have a cardiac follow-up. Seven (14%) patients had PAH, of whom one third (28%) had Eisenmenger syndrome. The left-to-right shunt ratio (Qp/Qs) was the only predictor of PAH. PAPVR-induced volume overload led to increased right chamber volumes (RV basal diameter 45±8 mm, RV mid-diameter 38±9 mm, RV/left ventricle ratio 1.2 ± 0.2, indexed RV end-diastolic area 14±4 cm2/m2, indexed RV end-systolic area 8.6 ± 3.2 cm2/m2) and increased RV functional parameters (tricuspid annular plane systolic excursion 27±5 mm, fractional area change [FAC] 42 ± 10%, global longitudinal strain [GLS] −22 ± 5). In contrast, PAH-induced pressure overload had no impact on right heart volumes, but on RV functional parameters, which were decreased compared to non-PAH patients (FAC 35 ± 13% vs. 43 ± 10%, p = 0.049; GLS -18 ± 5 vs. −23 ± 4%, p = 0.01). Conclusions: PAPVR prevalence was 0.2%. When incidentally diagnosed, its management was often neglected despite potentially serious consequences (14% PAH). Only the Qp/Qs ratio was predictive of PAH. PAPVR-induced volume overload was shown to increase right heart volumes, while PAH-induced pressure overload caused alterations of RV functional parameters
