    Temporary/Permanent Dual Cross-Link Gels Formed of a Bioactive Lactose-Modified Chitosan

    Mounting evidences have recognized that dual cross-link and double-network gels can promisingly recapitulate the complex living tissue architecture and overcome mechanical limitations of conventional scaffolds used hitherto in regenerative medicine. Here, dual cross-link gels formed of a bioactive lactose-modified chitosan reticulated via both temporary (boric acid-based) and permanent (genipin-based) cross-linkers are reported. While boric acid rapidly binds to lactitol flanking diols increasing the overall viscosity, a slow temperature-driven genipin binding process takes place allowing for network strengthening. Combination of frequency and stress sweep experiments in the linear stress\u2013strain region shows that ultimate gel strength, toughness, and viscoelasticity depend on polymer-to-genipin molar ratio. Notably, herewith it is demonstrated that linear stretching correlates with strain energy dissipation through boric acid binding/unbinding dynamics. Strain-hardening effect in the nonlinear regime, along with good biocompatibility in vitro, points at an interesting role of present system as biological extracellular matrix substitute

    Partitioning of bronchopulmonary carcinoids in two different prognostic categories by Ki-67 score

    Introduction: Histological distinction between typical and atypical bronchopulmonary car- cinoids is based on mitotic activity and necrosis. Regardless of these two parameters, outcome after surgery is often unpredictable. In this study the prognostic value of different clinico-pathological factors was retrospectively analyzed in a large series of patients with bronchopulmonary carcinoid. Materials and Methods: The long-term post-surgical out- come of 106 radically treated patients affected by bronchopulmonary carcinoid from two Italian centers was correlated with tumor characteristics assessed by combining conven- tional histology with a panel of immunohistochemical markers of neuroendocrine differen- tiation (chromogranin-A, NSE) and proliferation activity (Ki-67 score). Results: Carcinoids were assessed as typical (TC = 75; 70.8%) and atypical (AC = 31; 29.2%). Mean follow-up was 8.3 years (range: 0-20; median: 8.0). All cases expressed neuroendocrine markers. At univariate analysis, tumor recurrence [14/75 TC (18.7%), 15/31 AC (48.4%)] correlated with carcinoid histotype (P = 0.003), tumor size (P = 0.012), mitotic index (P = 0.044), Ki-67 score (P < 0.0001), and synchronous node metastasis (P = 0.037). Of these, Cox multivari- ate analysis confirmed only Ki-67 score as independent predictor of disease recurrence (P = 0.009). The best cut-off for Ki-67 score (calculated by ROC curves) discriminating recurrent vs non-recurrent disease was 4% (sensitivity 79.3%; specificity 83.8%; area under the curve 0.85). By stratifying patients according to this cut-off, a significantly dif- ferent disease-free survival was found (log-rank test P < 0.0001). Conclusion: Ki-67 score accurately separates bronchopulmonary carcinoids in two well-distinct histo-prognostic categories. Ki-67 score predicts the patients outcome better than mitotic count, histotype, and tumor stage and it is therefore helpful in establishing the appropriate follow-up. © 2011 Grimaldi, Muser, Beltrami, Machin, Morelli, Pizzolitto, Talmassons, Marciello, Colao, Monaco, Monaco and Faggiano

    Efectividad del aceite esencial de orégano mexicano de República Dominicana (Lippia graveolens) contra plagas del maíz (Sitophilus zeamais y Fusarium verticillioides)

    The insecticidal and antifungal properties of Mexican oregano (Lippia graveolens) essential oil from the Dominican Republic were investigated under laboratory conditions against two main pests of stored grains: Sitophilus zeamais and Fusarium verticillioides. Although oregano essential oil at 600 ?l/l air did not result in a significant mortality of S. zeamais after 24 hours of exposure by fumigation, this essential oil was a powerful acetylcholinesterase (AChE) inhibitor in vitro. One of the main components of oregano essential oil, p-cymene, presented fumigant toxicity and AChE inhibition activity against the maize weevil. The L. graveolens essential oil antifungal activity against F. verticillioides was evaluated at 50, 100 and 200 ?l/l, and it was found that growth parameters were affected by the presence of oregano essential oil in the media, whereas FB1 production was not inhibited. The results demonstrate that oregano essential oil and p-cymene can be used as alternatives to synthetic pesticides against F. verticillioides and S. zeamais, respectively.Se estudiaron en condiciones de laboratorio las propiedades insecticidas y fungicidas del aceite esencial de orégano mexicano (Lippia graveolens) de República Dominicana contra dos plagas principales de granos almacenados: Sitophilus zeamais y Fusarium verticillioides. Aunque el aceite esencial de orégano a 600 ?l/l de aire no resultó en una mortalidad significativa de S. zeamais después de 24 horas de exposición a la fumigación, fue un potente inhibidor de la acetilcolinesterasa (AChE) in vitro. El p-cimeno, uno de los principales componentes del aceite esencial, presentó actividad fumigante y de inhibición de la AChE contra el gorgojo del maíz. Además se evaluó la actividad antifúngica frente a F. verticillioides a 50, 100 y 200 ?l/l y se encontró que los parámetros de crecimiento del hongo fueron afectados por la presencia del aceite esencial de orégano en el medio, mientras que la producción de fumonisina B1 (FB1) no se inhibió. Los resultados demuestran que el aceite esencial de orégano y el p-cimeno se pueden usar como alternativas a los pesticidas sintéticos contra F. verticillioides y S. zeamais, respectivamente

    Assessment of central venous catheter-associated infections using semi-quantitative or quantitative culture methods

    <p class="MsoNormal" style="margin: 0cm 0cm 0pt; line-height: normal; text-align: justify; mso-layout-grid-align: none;"> Semiquantitative (Maki) and quantitative (Brun- Buisson) culture techniques were employed in the diagnosis of catheter-related bloodstream infections (CRBSI) in patients who have a short-term central venous catheter (inserted for 30 days). The diagnosis of CRBSI was based on the results of semiquantitative and quantitative culture of material from the removed catheters. Catheter tips (118) from 100 patients were evaluated by both methods. Semiquantitative analysis revealed 34 catheters (28.8%) colonized by ≥15 colonyforming units (cfu), while quantitative cultures (34 catheters, 28.8%) showed the growth of ≥10<sup>3</sup> cfu/mL. Bacteremia was confirmed in four patients by isolating microorganisms of identical species from both catheters and blood samples. Using the semiquantitative culture technique on short-term central venous catheter tips, we have shown that with a cut-off level of ≥15 cfu, the technique had 100.0% sensitivity, specificity of 68.4%, 25.0% positive predictive value (PPV) and 100.0% negative predictive value (NPV), efficiency of 71.4% and a prevalence of 9.5%. The quantitative method, with a cut-off limit of ≥10<sup>3</sup> cfu/mL, gave identical values: the sensitivity was 100.0%, specificity 68.4%, positive predictive value (PPV) 25.0%, negative predictive value (NPV) 100.0%, efficiency 71.4% and prevalence 9.5%. We concluded that the semiquantitative and quantitative culture methods, evaluated in parallel, for the first time in Brazil, have similar sensitivity and specificity.     ASSESSMENT OF ACTION OF DISINFECTANTS AGAINST LISTERIA MONOCYTOGENES BIOFILMS

    <p class="MsoNormal" style="margin: 0cm 0cm 0pt; line-height: normal; text-align: justify; mso-layout-grid-align: none;"> The purpose of this study was to assess the action of various disinfectants used in food industry against biofilm cells of Listeria monocytogenes formed on stainless steel surfaces during 24, 72 and 120 hours. Numbers of viable biofilm cells decreased after treatment with all the tested disinfectants (iodine, biguanide, quaternary ammonium compounds, peracetic acid and sodium hypochlorite). Sodium hypochlorite was the most effective disinfectant against the biofilm cells, while biguanide and iodine were the least. Scanning electron microscopy observations demonstrated attached cells on stainless steel surfaces after treatment with all the disinfectants. These observations showed that microorganisms were not completely removed from stainless steel surfaces after treatment with the disinfectants, however, the attachment did not means the viability of remaining cells. The biofilm age in hours (24, 72 and 120) had no apparent influence on resistance of microbiological cells to the disinfectants under study.     Slime production is an important virulence factor of coagulase-negative Staphylococcus spp., allowing them to attach to smooth surfaces of biomaterials, and it has been associated with infections of implanted medical devices. In the present study the production of slime capsules in 27 strains of coagulase-negative Staphylococcus was investigated by culture in Congo Red agar (77.7% positivity), spectrophotometric or microplate method (81.4% positivity) and scanning electron microscopy (88.9% positivity). The resistance of coagulase-negative strains of Staphylococcus to various antimicrobial agents was also determined by agar disk diffusion. The proportion of strains resistant to penicillin G, oxacillin, erythromycin, clindamycin and gentamicin among the slime-producing staphylococci was 88.9%, 70.4%, 81.5%, 66.7% and 59.2%, respectively; all of the coagulase-negative staphylococci were susceptible to vancomycin. The strains isolated from central venous catheters were identified by a conventional method and the API Staph system. The 27 coagulase-negative Staphylococcus strains were identified as: S. saprophyticus (3.7%), S. xylosus (7.4%), S. haemolyticus (14.8%), S. epidermidis (37.0%), S. warneri (14.8%), S. lugdunensis (7.4%), S. hominis (7.4%), S. schleiferi (3.7%) and S. chromogenes (3.7%). It can be concluded that in the most of the coagulase-negative Staphylococcus species there was an association between slime production, the nosocomial origin of the strains and reduced sensitivity to the antibiotics, suggesting a pathogenic potential in the hospital environment.A produção de slime é um importante fator de virulência dos estafilococos coagulase-negativa, permitindo-lhes aderir sobre as superfícies lisas de biomateriais, e por isso, é associada aos processos de infecção de implantes. No presente estudo a produção de slime em 27 cepas de estafilococos coagulase-negativa foi investigada por cultura em ágar vermelho Congo (77,7% de positividade), método espectrofotométrico ou da microplaca (81,4% de positividade) e microscopia eletrônica de varredura (88,9% de positividade). Foi também avaliada a resistência de estafilococos coagulasenegativa a vários antimicrobianos usando a técnica do disco difusão. A porcentagem de resistência à penicilina G, oxacilina, eritromicina, clindamicina e gentamicina em estafilococos produtores de slime foi respectivamente de 88,9%; 70,4%; 81,5%; 66,7% e 59,2%; todos os estafilococos coagulase-negativa foram vancomicina sensíveis. As cepas isoladas de cateter venoso central foram identificadas por método convencional e sistema API Staph. Os 27 estafilococos coagulase-negativa foram identificados como: S. saprophyticus (3,7%), S. xylosus (7,4%), S. haemolyticus (14,8%), S. epidermidis (37,0%), S. warneri (14,8%), S. lugdunensis (7,4%), S. hominis (7,4%), S. schleiferi (3,7%) e S. chromogenes (3,7%). Pode-se concluir que entre a maioria das espécies Staphylococcus coagulase-negativa houve associação entre a produção de slime, origem nosocomial das cepas e reduzida sensibilidade aos antimicrobianos, sugerindo potencial patogênico no ambiente hospitalar.     Effect of grafting solvent in the optimisation of Sba-15 acidity for levulinIc acid production

    Valorisation of lignocellulosic biomass for chemical production is one of the main challenges of the 21st century. Levulinic acid (LA) has been chosen as the target product due to its potential as an intermediate to produce a wide variety of other chemicals. The attention has been focused on the formulation of an active solid acid catalyst for the hydrolysis of glucose to LA. Therefore, a deep modification of SBA-15 with sulfonic groups was performed using post-synthesis grafting method. In particular, this work focuses on the role of different grafting solvents. Since the traditional solvents such as toluene or hexane, are flammable and toxic, a safer and more environmentally friendly solvent, a mixture of water and NaCl has been investigated. It was found that the nature of the solvent highly affects the morphological and chemical features of the materials. Thus, the best catalytic results were obtained with the catalyst prepared in water and NaCl; indeed 30 % and 16 % of LA yield were obtained from fructose and glucose hydrolysis at 180 °C respectively. Water and NaCl mixture guarantees the best distribution of sulfonic groups over the surface, leading to the most balanced acid catalyst
