Định tính và định lượng Huperzine a trong cây Thạch tùng răng cưa (Huperzia serrata) ở Đà Lạt, tỉnh Lâm Đồng

Huperzine A, an alkaloid, was originally isolated from Huperzia serrata. This compound potentially enhances the memory in animal, hence, it has been approved as a drug for the clinical treatment of Alzheimer’s disease, a major disease affecting the elderly population throughout the world. Because Huperzine A is an acetylcholinesterase inhibitor, the presentation of Huperzine A in brain inhibited acetylcholinesterase activity, thus, leading to the increase in concentration of acetylcoline. In Vietnam, H. serrata distributed in Sapa (Lao Cai) and Da Lat (Lam Dong), this species provide valuable pharmaceutical materials to the treatment for Alzheimer’s diseases. In this research, we evaluated the availability of Huperzine A in Huperzia serrata, which was collected from Da Lat (Lam Dong) in two seasons: Spring and Autunm. Thin layer chromatography (TLC) method was used to preliminary qualitative analysis. High performance liquid chromatography (HPLC) method were used for determining Huperzine A content in samples. In the result, Huperzine A is almost existed in leaves of Da Lat Huperzia serrata and equivalent levels of Chinese Huperzia serrata. The Content of Huperzine A was different between two collection samples in Spring and Autumn, by analyze HPLC data, the samples was harvested in Autumn contents 92.5 µg.g-1dry sample and the spring is 75.4 µg.g-1 dry sample. Therefore, the content of Huperzine A in Huperzia serrata’s leaves sample is  harvested in the fall compared with samples collected in the spring is higher 17.1 µg.g-1dry samples.Huperzine A là một alkaloid có nguồn gốc tự nhiên, là hoạt chất chính có trong cây Thạch tùng răng cưa (Huperzia serrata). Chất này được ứng dụng trong việc điều trị lâm sàng bệnh mất trí nhớ ở người cao tuổi Alzheimer. Sự có mặt của Huperzine A làm tăng hàm lượng acetylcholine trong não bằng cách ức chế enzyme acetylcholinesterase. Ở Việt Nam, cây Thạch tùng răng cưa mới được phát hiện ở Sapa (Lào Cai) và Đà Lạt (Lâm Đồng), đây là nguồn dược liệu quý cho y học trong việc điều trị bệnh Alzheimer. Trong nghiên cứu này, chúng tôi đánh giá sự có mặt của Huperzine A ở trong mẫu cây Thạch tùng răng cưa được thu hái tại Đà Lạt vào mùa Xuân và mùa Thu bằng phương pháp sắc kí bản mỏng (TLC) và sắc ký lỏng hiệu năng cao (HPLC). Kết quả cho thấy, có Huperzine A trong mẫu lá cây Thạch tùng răng cưa và hàm lượng tương đương với kết quả phân tích mẫu Thạch tùng răng cưa của Trung Quốc. Hàm lượng của Huperzine A có sự khác nhau giữa hai mùa Xuân và Thu, mẫu Thạch tùng răng cưa thu hái vào mùa Thu có hàm lượng là 0,0925 mg/g mẫu khô và mùa Xuân là 0,0754 mg/g mẫu khô. Như vậy, Huperzine A trong mẫu lá Thạch tùng răng cưa thu hái vào mùa Thu cao hơn so với mẫu thu hái vào mùa Xuân là 0,0171 mg/g mẫu khô
